1.Drug Use in 1250 Patients with Community Hypertension

OBJECTIVE:To explore the comprehensive model for the prevention and treatment of community hypertension.METHODS:By questionnaire survey,the community hypertension target population was screened and the drug use in hypertensive patients was analyzed statistically.RESULTS:Of the 65 000 community residents investigated,1 250 were found to be hypertensive patients,with a total treatment rate of 80.0%;only 32.8% were treated with at least 2 kinds of drugs,and the overall blood pressure control efficacy was poor.CONCLUSION:The program of prevention and treatment for community hypertensive patients is conducive to patients' long term adherence to medication,scientific and rational drug use and effective and stable control of blood pressure.
2.Oroxylin A ameliorates isoproterenol-induced heart failure model in rats through promoting myocardial autophagy

The protective effect and mechanism of Oroxylin A, a naturally occurring compound in Scutellaria baicalensis Georgi, was investigated in this study. Isoproterenol administration to rats triggered classic cardiac failure, as demonstrated by objective parameters of cardiac dysfunction. Intragastric administration of oroxylin A at the dose of 25, 50 and 100 mg/(kg·d)significantly improved deranged cardiac parameters in the isoproterenol-induced heart failure model in a dose-dependent manner. At the same time, oroxylin A markedly ameliorated cardiac histological changes and down-regulated serum levels of various neuroendocrine factors including norepinephrine, aldosterone, brain natriuretic peptide, endothelin 1, angiotensin II and so on. Mechanistically, augmenting autophagy of myocardial cells via the inhibition of AKT1-RPS6KB1 signaling contributed to the improvement of isoproterenol-induced rat heart failure by oroxylin A. Taken together, these results suggest that oroxylin A ameliorates heart failure through promoting autophagy in myocardial cells.
3.Exploration and clinical validation of the repair mode of the sclerotic zone of steroid-induced osteonecrosis of the femoral head based on Tandem Mass Tags technology

BACKGROUND:The sclerotic zone in the femoral head is an important imaging feature in the progression of steroid-induced femoral head necrosis,which is associated with disease prognosis.Peroxisome proliferator-activated receptor γ coactivator 1α(PGC-1α)has been shown to possess biological activities such as osteogenesis,angiogenesis and anti-mitochondrial apoptosis,which may be closely related to bone repair of steroid-induced femoral head necrosis. OBJECTIVE:To screen for the differential proteins in the sclerotic zone of steroid-induced osteonecrosis of the femoral head versus the normal zone,to screen for hub proteins in the sclerotic zone,and to verify the differential expression of hub proteins in the femoral head specimens following steroid-induced femoral head necrosis,and to to explore the repair pattern of the sclerotic zone following steroid-induced femoral head necrosis. METHODS:Femoral head samples were collected from patients with steroid-induced osteonecrosis of the femoral head receiving total hip arthroplasty.The differentially expressed genes in the sclerotic zone and the normal zone were screened by Tandem Mass Tags and analyzed by GO and KEGG signaling pathways to construct a protein-protein interaction network and screen hub genes.In addition,the expression of hub genes in the sclerotic zone was verified by immunohistochemistry and western blot. RESULTS AND CONCLUSION:Quantitative protein profiling by Tandem Mass Tags revealed that 609 proteins were significantly differentially expressed(Log2FC>1.20,Log2FC<0.84 and P<0.05)in the sclerotic zone of the femoral head compared with the normal zone,of which 290 proteins were upregulated and 319 proteins were downregulated.The GO and KEGG pathway enrichment analyses revealed that among the top 10 enriched pathways,Wnt signaling pathway and life-cycle regulatory pathway were closely related to bone repair;in the life-cycle regulatory pathway,PGC-1α was one of the important proteins.In addition,western blot results verified the low expression of PGC-1α and NRF1 in the sclerotic zone and high expression of Cleaved Caspase-3 in the sclerotic zone compared with the normal zone of steroid-induced femoral head necrosis specimens.Light microscopic immunohistochemical results showed the distribution of PGC-1α,NRF1 and Cleaved Caspase-3 positive expression in the sclerotic and normal zones in the femoral head tissue specimens,indicating the presence of their expression in bone trabeculae,osteoblasts and bone marrow.In contrast,the brown area of the sclerotic zone of femoral head necrosis stained darker and showed more obvious expression of Cleaved Caspase-3.To conclude,in the sclerotic zone of steroid-induced femoral head necrosis,biological behaviors including activation of osteogenesis-related pathways such as Wnt and oxidative apoptosis characterized by low expression of PGC-1 are observed.Low expression of PGC-1α in the sclerotic zone of steroid-induced femoral head necrosis may be associated with the activation of oxidative apoptosis.
4.SNP array analysis of three cases with partial 21q trisomy.

OBJECTIVETo analyze three cases with partial 21q trisomy, and correlate their genotypes with phenotypes.
METHODSG-banding chromosomal analysis and single nucleotide polymorphism (SNP array) were performed for the three cases and their parents.
RESULTSSNP array has detected partial 21q trisomy in three cases and one mother, with variable size and location of the duplications. Case 1 harbored a 12.35 Mb duplication at 21q22.11q22.3, which spanned the Down syndrome critical region. Case 2 harbored a 35.32 Mb duplication at 9p24.3p13.3 and a 14.42 Mb duplication at 21q11.2q21.3, with the former spanning the partial 9p trisomy syndrome critical region excluding the Down syndrome critical region, and was inherited from his mother. Case 3 harbored a 4.17 Mb tetraploidy at 21q11.2q21.1 in the form of mosaicism, which spared the Down syndrome critical region. His mother carried a 4.17 Mb triploidy at 21q11.2q21.1, which was also a mosaicism.
CONCLUSIONPartial 21q trisomy may occur in various forms and its clinical phenotypes are heterogeneous. Combined use of genetic techniques, particularly SNP array, is crucial for diagnosing partial 21q trisomy and delineating its genotype-phenotype correlation.

5.Rabbit Dry Eye Model Induced by a Controlled Drying System

【Objective】To provide a rabbit model for dry eye using a dry environment induced by Controlled Drying System.【Methods】Twenty-four male New Zealand rabbits were used in the experiment. They were randomly divided into control group and dry group,each one with 12 rabbits. The dry group was randomly housed in Controlled Drying System(CDS)for 14 days. The relative humidity,airflow and temperature were kept at(22±4)%,3~4 m/s and(23~25)℃, respectively. The control group were fed in a normal environment where relative humidity,airflow and temperature were kept at 60%~70%,0.2 m/s and(23~25)℃. The Schirmer test,corneal fluorescein staining,conjunctival lissamine green staining were performed during the experimental process on days 0,3,7,and 14. On the last day,the rabbits were euthanized and the eye tissues were made into paraffin-cut sections. After staining,we evaluated the corneal epithelial thickness and goblet cell number in the conjunctiva using light microscopy. MUC5AC in the conjunctival epithelium was detected by immunofluorescence. The apoptosis level changes on the ocular surface were evaluated using Caspase- 3 by immunohistochemistry. 【Results】 Decreased tear production ,increased corneal fluorescein staining and increased conjunctival lissamine green staining were found on days 3,7,and 14 in the dry group compared with the control group(P < 0.001). Corneal epithelial thickness of control group and dry group were (58.0±7.2)μ m and(47.8±7.6)μ m ,which showed corneal epithelial thickness of dry group was decreased(P<0.05). Goblet cells in the conjunctiva of control group and dry group were 15 ± 4 and 10 ± 2,which showed goblet cells of dry group was decreased(P<0.01). The expression of MUC5AC(consistent with goblet cells deficiency) was also reduced. Caspase- 3 was highly expressed on the corneal epithelium in the dry group. IOD/field of control group and dry group were(17±2)% and(20±2)%(P<0.01).【Conclusions】 Dry environment can make rabbits have pathological changes of dry eye on ocular surface epithelium. This dry eye model of rabbit caused by Controlled Drying System would be an effective tool to study the pathogenesis of dry eye.
