1.Expression of cannabinoid receptor type 2 in skin lesions of psoriasis vulgaris

Objective To investigate the expression of cannabinoid receptor type 2 (CNR2) in skin lesions of psoriasis vulgaris,and to explore its significance.Methods Tissue samples were obtained from lesions and perilesional skin of 20 patients with psoriasis vulgaris,and normal skin of 10 human controls without psoriasis or other autoimmune dermatoses.Real-time fluorescence-based quantitative PCR and immunohistochemistry were performed to determine the mRNA and protein expression levels of CNR2 in these samples.Statistical analysis was carried out by using two-sample t test and chi-square test.Results CNR2 mRNA and protein were expressed in the normal control skin,lesional skin and perilesional skin tissues.As immunohistochemistry showed,CNR2 was distributed diffusedly in the prickle cell layer in lesional skin,but expressed in the basal cell layer of the epidermis in normal control skin and perilesional skin.The mRNA expression level (2-△△α) of CNR2 was significantly higher in lesional skin than in normal control skin and perilesional skin tissues (3.97 vs.1.00 and 1.49,both P < 0.05),but similar between normal control skin and perilesional skin tissues (P > 0.05).The expression rate of CNR2 protein was significantly higher in lesional skin (90% (18/20) than in perilesional skin (15% (3/20),x2 =22.56,P < 0.01) and normal control skin tissues (10% (2/20),c2 =18.37,P< 0.01),while there was no significant difference between normal control skin and perilesional skin tissues (x2 =0.14,P =0.70).Conclusions The mRNA and protein expressions of CNR2 are both increased in skin lesions of psoriasis vulgaris,suggesting that CNR2 may be related to the occurrence and development of psoriasis vulgaris.
2.Clinical analysis of screening for gestational diabetes mellitus in 945 pregnant women with 50 g glucose challenge test

Objective To explore the clinical characteristics of gestational diabetes mellitus (GDM) and its relevant factors.Methods Using two diagnostic criteria set by World Health Organization and Carpenter/Coustan respectively, clinical data of 945 pregnant women who underwent glucose challenge test (GCT) in the Department of Obstetrics and Gynecology of China-Japan Friendship Hospital in Beijing during the past year were retrospectively analyzed.Results Of 945 pregnant women, 338 (35.8%) had 1 h serum glucose level equal to or greater than 7.8 mmol/L(140 mg/dl).Of these 338 women with positive GCT, 228 underwent 75 g oral glucose tolerance test (OGTT) and 102 (44.7%) of 228 were diagnosed as GDM. Women with GDM diagnosed according to either criteria had older age, higher body mass index(BMI) before pregnancy, more weight gain during the first 15 weeks of gestation, higher fasting blood glucose in early pregnancy and higher proportion of those with history of diabetes in their first-degree relatives than those with negative GCT. OGTT for pregnant women at about 30 weeks of gestation showed that insulin sensitivity decreased significantly in those with GDM compared with that in those with normal glucose tolerance, and the peak of insulin secretion delayed significantly in women with GDM.Logistic regression analysis showed that older age, higher BMI before pregnancy, more weight gain in early pregnancy and higher fasting blood glucose in early pregnancy were all independently associated with increased incidence of GDM.Conclusions Older age, higher BMI before pregnancy,more weight gain in early pregnancy,higher fasting blood glucose in early pregnancy,higher proportion of those with history of diabetes in their first-degree relatives and more severe insulin resistance were found in pregnant women with GDM than in those without GDM. It is suggested that those pregnant women with risk factors of GDM should be screened by fasting blood glucose in early pregnancy in order to be diagnosed and treated earlier.

4.Analysis of onset characteristics and clinical features in Chinese population with impaired glucose regulation

65 years and 55~65 years respectively,whereas the frequency of IFG in each age group was close,which was more than IGT in the age group of 25~34 years only.The differences of many clinical indexs among 3 groups were significant.The change tendecny of indexs in IFG/IGT group was more obvious.Conclusion The onset characteristics of age and sex are significantly different between IFG and IGT groups,and the latter is more strongly associated with cardiovascular risk factors than the former.Undoubtedly,IFG/IGT group has the highest cardiovascular risk factors.
5.Preliminary study on new parameter of myocardial elasticity

Objective To discuss the value of the new parameter of myocardial elasticity derived from circumferential strain.Methods Right ventrieular apical pacing was established on the beagle dogs.Before and after pacing,the peak of early diastolic flow velocity (E peak),peak of atrial flow velocity (A peak),and the ratio of A/E were measured.The peak pressure-rise rate (+ dp/dtmax),peak pressure-fall rate (-dp/dt max),left ventricular systolic pressure(LVSP),LV diastolic pressure(LVDP)were measured by cardiac catheterization.The circumferential strain at the levels of base (CSRA),papillary (CSPM) and cardiac apex (CSAP) were measured,and the parameter of myocardial clasticity at the levels of base (EBA),papillary (EPM) and cardiac apex (EAP) were measured and calculated,respectively.Results ①After pacing,the + dp/dt and LVDP were increased,but the-dp/dtmax was decreased (P <0.05).② After pacing,the CSPM and CSAP were all decreased,but EBA,EPM,EA were all increased(P <0.05).③ The areas under the receiver operating characteristic curves (ROC) of CSAP,CSPM,EAP,EPM and EBA were all 1,and the area under the curve of CSBA was 0.687.EAB,EPM and EAP were significant negative correlation with CSPM and CSAP,but are positive correlated with A/E and LVDP (P < 0.05).Conclusions The parameter of myocardial elasticity derived from circumferential strain includes useful information about myocardial deformation and function.The new parameter can be applied to assess the diastolic function of left ventricle.
6.The species and distribution of fleas in plague natural foci of Baotou

Objective To investigate the situation of flea species and distribution in the natural foci of plague in Baotou and provide a scientific basis for prevention and control of the disease.Method Flea specimens collected over the years and literature recordings were analyzed systematically.Results A total of 5 families,19 genera and 39 fleas were recorded,and 9 of them could be naturally infected with plague bacteria (Nosopsyllus laeviceps,Xenopsylla conformis,Neopsylla pleskei,Citellophilus tesquorum,Neopsylla bidentatiformis,Leptopsylla pavlovskii,Ophthalmopsylla praefecta,Rhadinopsylla insolita and Frontopsylla luculenta).There were 34 fleas in the north area,and 14 of them were regional fleas; there were 25 species in south area,and 5 were regional fleas.Conclusion Flea distribution in Baotou City has obvious regional characteristics,and these fleas can be naturally infected by the plague.
7.Role of Rho-kinase and PKC in cyclosporine A regulating vascular reactivity and it relationship to MPTP after traumatic hemorrhagic shock

Objective To investigate the relationship of the beneficial effect of CsA on vascular reactivity to Rho-kinase,protein kinase C ( PKC) and mitochondrial permeability transition pore ( MPTP) in traumatic hemorrhagic shock rats. Methods With traumatic hemor-rhagic shock rats and hypoxia-treated vascular smooth muscle cells ( VSMCs) ,the role of Rho-kinase and PKC in CsA-regulating vascular re-activity following shock and their relationship to MPTP was observed. Meanwhile,the effects of CsA on inflammatory mediators including TNF-α, IL-1β and IL-6 were also studied. Results CsA significantly improved the vascular reactivity of superior mesenteric artery following hemor-rhagic shock. Rho-kinase inhibitor Y27632 significantly antagonized CsA-induced increase of vascular reactivity,while PKC inhibitor stauros-porine had no significant influences on the effects of CsA. Further studies showed that CsA and Rho-kinase agonist U46619 inhibited the o-pening of MPTP in hypoxia-treated VSMCs. In addition,shock induced a significant increase of TNF-αand IL-1β,but CsA did not show a sig-nificant inhibitory effect on their level. Conclusion CsA-induced restoration of vascular reactivity following traumatic hemorrhagic shock via inhibiting MPTP opening,and Rho-kinase participate in this process.
8.Detection of DD3 mRNA level in peripheral blood of patients with prostate cancer and its clinical value

Objective To study the level and clinical value of differential display code 3 (DD3) in peripheral blood of patients with prostate cancer.Methods 27 patients with prostate cancer from April 2014 to April 2015 in Affiliated Hospital of Hangzhou Normal University were researched.26 patients with non prostate diseases were selected as control group.DD3 mRNA levels were detected in peripheral blood of all patients.DD3 absorbance value of three groups of patients,the relationship between the relative content of DD3 mRNA and the clinical characteristics of prostate cancer,DD3 mRNA in urine and peripheral blood of patients with prostate cancer and benign prostatic hyperplasia were observed.Results In the patients with benign hyperplasia of prostate and non prostate diseases,a total of 4 patients did not appear to be DD3 specific bands.In prostate cancer patients,all patients were found to have DD3 specific bands.The relative content of patients with non prostate diseases was (0.18 ± 0.05) copies/mL.The relative content of benign prostatic hyperplasia patients was (0.30 ± 0.09) copies/mL.The relative content of prostate cancer patients was (0.78 ± 0.23) copies/mL.The positive expression rates of DD3 mRNA in peripheral blood and urine of patients with prostate cancer were 85.18% (23/27),51.85% (14/27),respectively,which were significantly higher than those in patients with benign prostatic hyperplasia[21.42% (6/28),7.14% (2/27)] (x2 =22.416,13.319,all P < 0.05).Conclusion The specific expression of DD3 mRNA in peripheral blood of patients with prostate cancer can be used as an effective basis for judging the patients' condition,and it has certain value for the treatment and prognosis of patients with prostate cancer.

Studies in recent years have shown that the mesenchymal stem cells promote nerve repair and improve neurological function after ischemic stroke.As a major source of mesenchymal stem cells,bone mesenchymal stem cells have potential and broad prospects in the treatment of ischemic stroke.This article reviews the sources,characteristics of mesenchymal stem cells,and the pathogenesis of mesenchymal stem cell transplantation in the treatment of ischemic stroke and prospects.
10.Effects of rotenone on NOS activity and NO concentration in caudatum and plasma in rats

Objective To observe the rotenone induced changes of nitric oxide (NO) concentration and nitric oxide synthase (NOS) activity in rat caudate nuclei and blood plasma. Methods Wistar rats were daily administrated with rotenone at the doses of 0.335, 0.670 and 1.005 mg/kg(sc). Animals were sacrificed on days 30, 60 and 90. The NO concentration and the NOS activity in the caudate nuclei and blood plasma were determined by biochemical methods. Results At each time point of days 30, 60 and 90, rotenone induced significant increases in NO concentration and NOS activity in both the caudate nuclei and blood plasma in dose and time dependent manners. Conclusion Long term exposure to rotenone leads to the increase of nitric oxide and nitric oxide synthase in brain tissues and blood plasma in rat.
