1.THE EXPRESSION OF THE VANILLOID RECEPTOR SUBTYPE 1 IN AFFERENT FIBERS OF THE RAT ESOPHAGUS

Objective To study the expression of the capsaicin receptor,vanilloid receptor subtype 1(VR1) immunoreactivity,their morphology and distribution in afferent fibers of rat esophagus. Methods Laser scanning confocal microscope combined with immunohistochemical double labeling methods were used. Results VRl-like immunoreactivity was observed on nerve fibers and terminalis within mucosa,submucosa,muscle layer and surrounding blood vessel throughout esophagus.Their profiles were fine fibers with some spiny,possessing varicose-like swellings along their lengths.About(92.3?3.7)% VR1 positive fibers co-localized with CGRP immunoreactivity,which representing large majority of afferent fibers in the esophagus was extrinsic in origin with cell bodies located in dorsal root ganglia.In dorsal root ganglion,VR1 was expressed in small-and middle-sized cell bodies.About(41.5?4.5)% VRl-immunoreactive neurons co-stained with CGRP and(67.9?3.2)% CGRP positive neurons co-localized with VR1.In nodose ganglion,the expression of VR1 was similar with dorsal root ganglion,but CGRP immunoreactive neurons very few.Only(4.7?1.4)% VR1 positive neurons co-stained with CGRP.Conclusion These results suggest that VR1 is expressed in afferent fibers in the wall of the rat esophagus which is in origin in dorsal root ganglion.
2.Distributions of purinergic P2X_3 receptor immunoreactive afferent fibers in the rat pharyngeal mucosa

Objective To study the distribution of purinergic P2X_3 receptor immunoreactive afferent fibers in the rat pharyngeal mucosa and provide morphological evidence for exploring the role of ATP in the signal transduction of sensory stimuli. Methods Twelve adult Wistar rats were used and immunofluorescenthistochemical double-labeling technique combined laser confocal scanning microscope was applied in the present study. Results 1.P2X_3 immunoreactive fibers were observed on sections from all parts of pharyngeal mucosa. Two types of positive fibers were found. One was free nerve fibers covered with many varicosities. Another one ramified in mucosa and showed complex arborization endings. Nerve plexus in mucosa were formed by P2X_3 immunoreactive fiber ramifications. 2.Most calcitonin gene-related peptide (CGRP) positive fibers intermingled with P2X_3 immunoreactive fiber arborizations. A few P2X_3 immunoreactive fibers were covered with many varicosities co-localized with CGRP. 3.In petrosal ganglion, most neurons were stained with P2X_3 or CGRP immunoreactivity and small number of P2X_3 immunoreactive neurons co-stained with CGRP. Conclusion These results indicated that the different types of afferent fibers in rat pharyngeal mucosa expressed purinergic P2X_3 receptor immunoreactivity and ATP might be related to nociceptive or physiological signals transduction in rat pharyngeal mucosa.

With the scientific and technological progress and rapid development of the social economy, the extensive application of various electronic equipments has gradually intensified the possibility of ELF-EMF(extreme low frequency electromagnetic fields) exposure, the relationship between ELF-EMF exposure and health effects attracted more and more eyes and many corresponding researches have been finished. In recent years, a lot of epidemiological researches about the relationship between ELF-EMF and leukemia, brain tumor, breast cancer and other tumors have been done at home and abroad, in the present paper the research results were reviewed and the possible mechanism of the effect of ELF-EMF exposure on tumors was discussed.

Objective To investigate the effect of low intensity microwave electromagnetic fields (EMF) on energy metabolism of cerebral cortical neurons of postnatal rats. Methods The cultured neurons were exposed to low intensity 900 MHz continuous microwave EMF (SAR=0.38-3.22 mW/g) 2 hours a day for 4 to 6 consecutive days. The activity of cytochrome oxidase(CCO) was taken as the index for energy metabolism level. The projection from the nucleus of solitary tract (Sol) to the nucleus accumbens (Acb) was examined by using HRP retrograde and anterograde tracing combined with immunocytochemical double-labeling technique. The following results were obtained: (1) when WGA-HRP was injected into the caudal part of the Sol, the labeled fibers, terminals and cell bodies were found in the ventromedial area of the caudal Acb. After injecting HRP into the ventromedial area of the caudal Acb, labeled cell bodies and terminals were found in the ipsilateral and contralateral caudal part of the Sol,mainly in the commissure nucleus and the medial subnucleus of nucleus of solitary tract. (2) After injecting HRP into the Acb and combined with immunocytochemical method, many HRP-TH, HRP-NT, and HRP-CCK double-labeled cell bodies were found in the caudal part of the Sol. The number of the HRP-TH double-labeled cell bodies was most numerous, HRP-NT cells was next and HRP-CCK cells was even less.
7.Analysis of impact factors for 273 medical dispute cases mediated with the people's mediation mechanism in Guangdong province

Objective To analyze impact factors for resolving cases of medical disputes with people's mediation mechanism,for improvement of this mechanism.Methods This paper picked 273 cases mediated by Guangdong Medical Dispute People Meditation Committee from June 2010 to December 2011,for analysis from doctor-factors,patient-factors,mediator-factors and composite factors.Results Under such mechanism,doctor-factor has not significant difference on the mediation success ratio and mediation timeliness of the cases; the success rates of cases of death,disability,and no obvious impairment suffered by patients are 68.57%,55.81% and 76.80% respectively.Such results are found by chi-square test as P<0.05,significant statistically.With cases reported via hospitals,the mediation timeliness is much better than other cases.Conclusion The mechanism is advantageous in effectively avoiding influences of case factors,yet emphasis should be placed on mediation of disability cases,and doctors should be encouraged to report the cases,contributing to higher success rate of mediation.
8.Meta-analysis of Clinical Effects of Topical Administration of Phenytoin on Wound Healing

Objective:To evaluate the clinical effects of topical administration of phenytoin ( PHT) on wound healing. Methods:The clinical trials on PHT topically used for wound healing were collected from Cocharne Library and PubMed ( from database establish-ment to May, 2016. Meta-analysis was performed using RevMan 5. 0 software and Stata 12. 0 software. Results:A total of 15 stud-ies involving 1 048 patients were included. Topical PHT treatment was significantly associated with complete healing rate (OR=3. 28, 95%CI:1. 23-8. 75, P=0. 02), production rate of health granulation tissue (OR=2. 18,95%CI:1. 33-3. 59, P=0. 002) and aver-age percentage reduction of wound surface size (SMD=1. 77, 95%CI:0. 53-3. 02, P<0. 000 01). However, heterogeneity existed in complete healing rate and average percentage reduction of wound surface size among the studies. Meta-regression analysis showed that wound types (P=0. 02) and treatment periods(P=0. 08) were associated with the heterogeneity of complete healing rate outcomes, and mean age was associated with the heterogeneity of average percentage reduction of wound surface size(P=0. 07). Conclusion:Meta-analysis suggests that topical PHT treatment has significant positive clinical effect on wound healing. There is heterogeneity among the studies, so topical PHT treatment still should be applied in clinical practice prudently.
9.The association of DNMT3a, DNMT3b expression with the state of promoter methylation of ERαgene and ERαexpression in sporadic breast cancer

Objective To investigate the correlationship between DNMT3a, DNMT3b protein expressions and the state of promoter methylation of ERα gene and ERα protein expression in the development of sporadic breast cancer. Methods A total of 180 patients with sporadic breast cancer and 30 patients with breast fibroadenoma were included in this study. The expressions of DNMT3a and DNMT3b protein were detected by immunohistochemical method. The state of promoter methylation of ERα gene was detected by methylation specific PCR in 97 patients with sporadic breast cancer. Results There were no significant differences in positive expression rates of DNMT3a and DNMT3b protein between breast fibroadenoma and breast cancer. There were higher expression levels of DNMT3a and DNMT3b in breast cancer patient of Ⅲ~Ⅳstages than those of Ⅰ~Ⅱstages. The expression of DNMT3a was significantly higher in patients with lymph node metastasis than that of patients without lymph node metastasis (P<0.05). Of 97 cases of breast cancer patients, ERα gene promoter methylation occurred in 39 cases (40.2%). The positive expression of DNMT3a protein was positively correlated with the ERα gene methylation (rS=0.250). The DNMT3a protein expression showed a significant influence to the overall survival (OS) in patients of breast cancer (P=0.035), no significant influence to the disease-free survival (DFS) (P=0.064). DNMT3b protein expression showed no significant influence to OS and DFS of patients with breast cancer (P=0.914 and 0.961). Conclusion The positive expressions of DNMT3a and DNMT3b are correlated with the invasion, metastasis and poor prognosis of sporadic breast cancer. DNMT3a was positively correlated with the state of ERα gene promoter methylation. The inhibition of DNMT3a and DNMT3b may have advantages in the prevention and treatment of sporadic breast cancer.
10.Expressions of EGF and bFGF in human prostate

Purpose:To study the expression and functional way of EGF and bFGF in human prostatic tissue.Method:To detect the expression of EGF and bFGF in 6 cases of human normal prostate (NP) and 27 cases of benign prostatic hyperplasia (BPH) specimens using mRNA dot blot, in situ hybridization and immunohistochemistry.Results:There was no expression of EGF mRNA in any of the prostate exmined,weakly immunohistochemical staining of EGF was observed in 2 cases of 6 NP and 5 of 27 BPH sepcimens,there was no statistic difference between NP and BPH (P>0.05).The total amount of bFGF mRNA in BPH tissue is much than that in NP tissue,aboundant bFGF mRNA was revealed in epithelial cells of 6 cases of NP tissues,but no bFGF;low amount of bFGF was expressed,according with the level of its mRNA both in basal and stromal cells of NP tissues;No signal for bFGF mRNA was found in epithelial cells of BPH tissues but bFGF protein was found on surface of local proliferating epithelial cells.Markedly increased expression of bFGF mRNA and its protein were present in basal and stromal cells of BPH tissues,especially in the region of local proliferating stromal cells.Conclusion:There are no EGF secretory cells in human normal or hyperplastic prostate; overexpression of bFGF in basal and stromal cells of human prostate caused irregular hyperplasia both of stromal elements and glandular epithelium via autocrine and paracrine pathways.
