1.Effect of Naohuandan Capsule in Treating Mild and Moderate Cases of Vascular Dementia

To observe the therapeutic effect of Naohuandan Capsule (mainly composed of Radix Ginseng, Radix Rehmanniae Preparata,etc.) in treating mild and moderate cases of vascular dementia (VD).Forty five cases of VD, which are subjected to the diagnostic standard of American Association of Psychiatry (DSM Ⅳ), were randomly allocated to two groups for a 3-month treatment course. Group A (n=22) was treated with Naohuandan Capsule and Group B (n=23) with Naofukang Tablet. Therapeutic effect was evaluated by scoring of mini-mental state examination (MMSE) and activity of daily living (ADL) and serum levels of estradiol (E 2) and testosterone (T) were also observed.Scores of MMSE and ADL were increased, E 2 and T levels were elevated and the ratio of E 2/T was decreased in Group A(P0 05). [Conclusion]Naohuandan Capsule has a definite effect in preventing and treating VD and its mechanism may be related to the increase of E 2 and T levels, decrease of E 2/T ratio and the regulation of neuroendocrine function.

AIM: To observe the changes of NO, ET-1, SOD and MDA levels in plasma of rats exposed to infrasound. METHODS: Using infrasound (frequency: 8 Hz; sound pressure level:130 dB), the rats were exposed for 1 d, 7 d, 14 d, 21 d and 28 d, 2 h daily, then the levels of NO, ET-1, SOD and MDA were measured after exposure. ObjectiveTo study the effect of 8 Hz infrasound on the expression of 5-hydroxytryptamine (5-HT) in rats' hippocampus and temporal cortex.Methods140 male SD rats were randomly divided into the control group, and experimental groups that exposed to infrasound of 8Hz,90dB,100dB and 130dB for 1,7,14,21,28,35,42 days. Experimental groups were exposed to infrasound for 2 hours each day. The control group was only placed in the infrasonic storehouse but without infrasound. Rats' brains were taken as soon as the exposure finished and strained by immunohistochemistry. The content of 5-HT in hippocampus and temporal cortex was detected under an optical microscope.ResultsInfrasound groups had less expression of 5-HT in hippocampus and temporal cortex than the control group (P<0.01), and the least were at the 28th day for 90 dB and 100 dB groups and the 21st day for 130 dB group. Then the expression of 5-HT had an increase in each group.ConclusionThe deceased expression of 5-HT in rats' hippocampus and temporal cortex could result from infrasound of 8 Hz. Rules of change are related to the parameter of infrasound and the 130 dB 8 Hz infrasound can induce greater changes compared with that of 100 dB and 90 dB.
4.Transurethral plasma kinetic enucleation of the prostate for benign prostatic hyperplasia more than 80 mL

Objective To evaluate the clinical effect and safety of transurethral enucleation of prostate with the bipolar plasma kinetic technique(PKEP)in the treatment of benign prostate hyperplasia (BPH)more than 80 mL.Methods The data from the 116 patients who underwent the PKEP were analyzed retrospectively.The clinical parameters include operation time,blood loss, postoperative catheter retention time,postoperative complication rates,the differences of the clinical parameters pre-and postopera-tively were compared,include maximum urine flow rate(Qmax),residual urine(RU),international prostate symptoms score(IPSS), and quality of life(QOL).Results Mean operation time was (87.46±25.01)min,Mean blood loss was (129.15±44.35)mL.Mean resected tissue weight was (77.67±19.56)g.No patient had the transurethral resection syndrome(TURS),All cases were followed up for 3 to 6 months,the clinical parameters pre-and postoperatively was respectively:Qmax(6.04±2.37)mL/s vs.(17.85±2.55) mL/s;RU(116.25±53.18)mL vs.(8.85±7.66)mL;IPSS(25.06±4.23)vs.(5.90± 1.91);QOL(4.85 ±0.65)vs.(1.71± 0.54).Conclusion The transurethral enucleation of prostate with the bipolar PKEP resects the proliferated prostate cleanly,had lesser bleeding and complication rates,had advantages of high safety,and satisfactory efficacy for the treatment of BPH more than 80 mL.
5.Effects ofBaitouweng Decoction on Intestinal Mucosal Healing of Ulcerative Colitis and Study on Partial Mechanism

Objective To detect the levels of tumor necrosis factor-α (TNF-α) and interleukin 10 (IL-10) in the plasma of patients with ulcerative colitis (UC); To investigate the clinical efficacy ofBaitouweng Decoction for UC and its influence on intestinal mucosal healing and partial mechanism.Methods Totally 66 patients with UC were randomly assigned to the treatment group and the control group. Patients in the treatment group were treated with Baitouweng Decoction (oral and enema treatment) and those in the control group took Mesalazine for 4 weeks. Before and after treatment, the therapeutic effects were assessed by Mayo scoring system and the grading of activities was evaluated by Baron Endoscope. Serum levels of TNF-α and IL-10 were detected by ELISA. Besides, another 20 healthy volunteers were recruited as the healthy control group for comparison.Results The endoscopic remission rate was 51.4% (19/35) in treatment group, higher than that of the control group (25.8%, 8/31,P<0.05). The clinical remission rate and efficacy rate in the treatment group were 74.2% and 88.6% respectively, higher than those of the control group (45.1% and 64.5%,P<0.05). The histological remission rate and efficacy rate in the treatment group were 25.7% and 54.3% respectively, higher than those of the control group (16.1% and 35.5%), without statistical significance (P>0.05). Serum levels of TNF-α were obviously positive correlated with Mayo score, Baron score and histology score (r=0.836, 0.735, 0.527,P<0.01). Serum levels of IL-10 were obviously negative correlated with Mayo score, Baron score and histology score (r=-0.704, -0.695, -0.509,P<0.01). Compared with pre-treatment, the serum levels of TNF-α significantly decreased (P<0.01), while the serum levels of IL-10 significantly increased (P<0.01) after treatment withBaitouweng Decoction.ConclusionBaitouweng Decoction (oral and enema treatment) is effective and safe in patients with UC. Its effects might be involved in regulating the imbalance of pro-inflammatory factors and anti-inflammatory factors, and promoting mucosal healing.
6.The role of survivin in the process of colorectal carcinogenesis

Objective To identify the role of survivin gene in colorectal carcinogenesis.Methods Using immunohistochemical method,expression of survivin gene were detected in colorectal mucosal tissues.Results Expression rates of survivin in four tissues were 62.5%,17.2%,0 and 0.there were significant differences between them(P0.05).Conclusion Survivin may play an important role in onset and progression of colorectal cancer.

8.Influence of various compatibilities on puerarin content in Gegen Qinlian Decoction

Object To set up the quantitative method for determining the puerarin in Gegen Qinlian Decoction (GQD) by RP-HPLC and to study the change of puerarin contents in various combinations of GQD. Methods L 8(2 7) orthogonal design and statistic analysis (SPSS 10.0) were used, the puerarin contents in the samples were determined by HPLC. Results Influence of Radix Scutellaria, Rhizoma Coptidis and Radix Glycyrrhizae Preparata on the puerarin content in GQD was insignificant. And interactions between two of three were insignificant too. In this experiment, puperarin in the sedimentation was found in the compatible decoction of Rhizoma Coptidis and Radix Puerariae. Conclusion The puerarin content is not markedly influenced by Radix Scutellaria, Rhizoma Coptidis and Radix Glycyrrhizae Preparata. Maybe, the change of pH is relevant to the sedimentation in the compatible decoction contained Rhizoma Coptidis. and Radix Puerariae
9.Effection of lentiviral-delivered PLC?1 siRNA on human colorectal carcinomas cell lines

Objective: To preare recombinant lentivirus stably suppressing PLC?1 in human colorectal carcinomas Lo-Vo cells, so as to establish LoVo cell lines deficient in PLC1 and to investigate the role of this gene. Methods: Recombinant lentivirus produceing PLC-?1 siRNA were prepared. After LoVo cells were transduced with lentivirust, stably transduced cells were selected by Blasticidin. The protein and mRNA expression of PLC?1 was examined by Western-blot and RT-PCR analysis. The effect of the lentivirus on the cell proliferation and cell adhesion was analyzed by XTT method and ctll adhesion assay, respectively. Results: PLC?1 siRNA knocked down PLC?1 expression in LoVo cells obviously. The silenced efficiency of siRNA transducted by recombinant lentivirus was very high. Adhesion of human colorectal carcinomas LoVo cell Lines was significantly decreased, while proliferation was not affected. Conclusion: Our research confirm that PLC?1 plays an important role in cell adhesion of colorectal carcinomas, and provides experimental evidences for targeting PLC?1 in gene therapy against cancer.
10.Influence of compatibility on content of baicalin in Gegen Qinlian Decoction

Methods By L 8(2 7) orthogonal design and statistics analysis (SPSS10 0), eight decoctions with Radix Scutellariae either alone or incombination with one or more of the other three components were prepared or analyzed. Contents of baicalin in them were determined by RP HPLC. Results It was significant (P0 05) and interactions between two of three were insignificant either. In this experiment, the sedimentation was found in the decoctions contained Radix Scutellariae and Rhizoma Coptidis. Conclusion The content of baicalin is reduced by Radix Puerariae and Rhizoma Coptidis, The resolution of baicalin is not increased by Radix Glycyrrhizae Preparata. Maybe the change of pH leads to the sedimentation in decoctions contained Radix Scutellariae and Rhizoma Coptidis.
