Objective To investigate the characteristics, degradation and biocompatibility of a woven chitosan fiber (CF). Methods Scanning electron microscopic observation, deteronination of density, degree of swelling and tensile strength were performed to characterize the CF. In vitro degradation was observed by immersing CF in buffered aqueous solution of pH 6 containing lysozyme at 37 ℃, while in vivo degradation was studied after intramuscular implantation of CF in rats. Furthermore, rat mesenchymal stem cells were directly cultured on CF to estimate its toxicity. Results The diameter of CF was about 2000?63?m, with a sleek surface. The density and rate of swelling were 1.093?0.27g/cm3 and 2.30?0.21, respectively. The tensile strength was 80.18?1.56MPa. Specific viscosity, measured after the addition of lysozyme to the solution, decreased dramatically within 2 hours compared with that of the control mixture without enzyme. There was obvious inflammatory reaction after intramuscular implantation in the first week after surgery, bat inflammation subsided gradually. Biodegradation was not obvious within 4 weeks, but majority of it was absorbed in twelve weeks. Cells grew well on it with normal morphology, and no inhibition of cell proliferation could be observed. Conclusion Woven CF has a good tensile strength and fairly good biodegradation and biocompatibility, and it can be used in bone and cartilage tissue engineering.

Objective To investigate the correlation of XRCC1 Arg194Trp Arg399Gln Single nucleotide polymor-phism (SNP) with radiotherapy response of squamous cell carcinoma of cervix. Methods Patients with exogenous type cer-vical squamous cell carcinoma confirmed by histopathology were selected for our study. These include:patients in stageⅠ(4 cases), patients in stageⅡ(36 cases), patients in stageⅢ(30 cases), patients in stageⅣ (3 cases). There are 30 patients with tumor diameter less than 4 cm and 43 patients with tumor diameter over 4 cm in our test. There are 36 cases with dose point A less than 80 Gy and 37 cases with dose point A over 80 Gy . Radiotherapy outcomes showed 47 cases of complete re-mission and 26 cases of part remission. Polymorphisms Arg194Trp, Arg399Gln of XRCC1 gene in 73 cervical cancer pa-tients were analyzed by mismatch amplification polymerase chain reaction (MAMA-PCR). Results Arg/Arg, Arg/Trp, TrP/Trp of Arg194Trp genotype distribution were 31 (42.5%), 37 (50.7%), 5 (6.8%) respectively. Arg/Arg, Arg/Gln, Gln/Gln of Arg399Gln distribution were 6 (35.6%), 39 (53.4%), 8 (11.0%) respectively. The response to radiotherapy was not statistical-ly significant in three genotypes, Arg/Arg, Arg/Trp, TrP/Trp of XRCC1 at codon 194(P>0.05). Neither was XRCC1 at codon 399. Multivariate analysis showed that late clinical stage was a risk factor of part remission. Conclusion SNP of XRCC1 gene at codon 194 and codon 399 could not predict clinical response of patients with squamous cell carcinoma of cervix to ra-diotherapy. The patients with advanced cervical cancer had poor response to radiotherapy.

Objective:To study immunological stimulation of synthetic oligodeoxynucleotides(ODN) containing unmethylated CpG dinucleotides(CpG-ODN) on human immune cells.Methods:CpG-ODN was co-cultured with peripheral blood mononuclear cells(PBMC) .The IFN-? and IFN-? in the supernatant were measured by ELISA;the reverse transcription PCR was used to analyze the expression levels of TLR9 mR NA in PBMC;MTT method was used to detect NK-mediated lytic activity of K562 cells.Results:CpG-ODN induced high amounts of IFN-? as well as IFN-? production;the lysis of K562 mediated by CpG-ODN 2216 stimulating NK cells was increased;the CpG-ODN could up-regulate the expression of TLR9 mRNA in PBMC. Conclusion: CpG-ODN 2216 can activate human immune reaction by increasing the production of IFN-? and IFN-?,the expression of TLR9 and NK cell lytic activity.

Objective To study the application of photodynamic therapy (PDT) for dental caries prevention using whole body luminescence fiber, and to investigate the effects of PDT on the content of Ca and P in rat molar enamel.Method The rat dental caries model was established by inoculating with S.mutans.Eighty male rats were randomly divided into five groups, including three experimental groups:17 mW (8 mW/cm2) PDT (group A),34 mW (15 mW/cm2) PDT (group B),68 mW (30 mW/cm2) PDT (group C),a positive control group:20 g/L NaF solution (group D),and a negative control group:0.9% physiological saline (group E).The experimental groups were treated by 40 μg/mL hematoporphyrin monomethyl ether (HMME) and 650 nm diode laser irradiation. The experiments were conducted for 4 weeks. The contents of Ca and P in the molars of each group were measured by inductively coupled plasma emission spectrometry. Results The contents of Ca and P in group B,C and D after PDT were significantly higher than those in group A and E (all P<0.05).The contents of Ca and P in group A showed no significant difference before and after PDT,while those in groups B and C showed significant increase after PDT (all P<O.05).The increment of Ca in group A after PDT was lower than that in group D (P<0.05),while those in group B and C were significantly higher than those in group D (all P<0.05).There was no significant difference in the increment of Ca and P between group B and C after PDT.Conclusions In the range of the experimental parameters, the PDT promoted effect of tooth remineralization is better than 20 g/L NaF.The levels of Ca and P in the tooth enamel can be promoted by PDT treatment, and the contents of Ca and P are related to the pewer of PDT.The effect of low power PDT on the remineralization of enamel is not obvious. The contents of Ca and P in the tooth enamel are increased with laser power of PDT.When the laser power increased to a certain value, the change in contents of the two elements is not obvious.PDT can maintain the tooth remineralization microenvironment.

ObjectiveTo study the changes of cellular immunity caused by intravenous infusion of allogenic rhesus mesenchymal stem cells (MSCs).MethodsMSCs were isolated and cultured. Then the immunomodulatory effects after MSCs infusion were evaluated by means of peripheral blood counts, mixed lymphocyte reaction (MLR) and analysis of lymphocytic subgroup. ResultsMSCs of rehsus were successfully cultivated. No acute toxicities or GVHD were observed in recipients. No obvious changes of peripheral blood counts were present. Recipients A2, A3, A4 were administered with MSC by 4.0 ×105/kg, 1.0 ×106/kg, 2.0×106/kg respectively and relative reaction (RR) of MLR decreased 14 days post MSCs infusion: from 46±2.6 %to 40.4±1.73 % (F =10.19, P =0.023), from (40.9±2.3) % to (33±2.1) % (F =2.593, P =0.013), from 48.3±2.0 % to 39±1.0 % (F =28.431, P =0.003) respectively. The decrease degree (ARR) was positively related to the amount of MSCs(F =27.413, P =0.038). RR was restored within 30 days post MSCs infusion. After MSCs infusion, CD3+ CD3+CD4+ and CD3+CD8+ T-lymphocytes decreased in recipient A4, who was administered with the largest number of MSCs, and restored within 30 days. ConclusionMSCs infusion without any other treatment could temporarily inhibit immunity of T lymphocytes in MLR and the immunity inhibition was positively related to the amount of MSCs.The specific immunological characteristics of MSCs were demonstrated with extensive prospect in clinical research.
