Chinese Journal of cardiovascular Rehabilitation Medicine 2016;25(1):1-7
Objective: To explore influence of tanshinone on rabbit malignant arrhythmias and calmodulin signal transduction pathway after myocardial infarction. Methods: A total of 30 rabbits were randomly and equally divided into sham operation group (sham group), model group and tanshinone group. Acute myocardial infarction model was established through ligating left anterior descending branch of coronary. After modeling, the rabbits were randomly and equally divided into model group and tanshinone group. Incidence rate of malignant arrhythmias, action potential duration (APD) of three myocardial layers (namely endocardium layer, myocardium layer and epicardium layer), transmural dispersion of repolarization, Ca2+ concentration, compared among three groups. Results: Incidence rate of malignant arrhythmias in tanshinone group was significantly lower than that of model group (20.0% vs. 70.0%, P<0.01); compared with sham group, there were significant rise in transmural dispersion of repolarization, 90% APD, Ca2+ concentration, expression levels of calmodulin and calmodulin kinase Ⅱ of three layers in model group and tanshinone group (P<0.01 all) ; compared with model group, there were significant reductions in transmural dispersion of repolarization[(46.2±10.9) ms vs.(35.5±8.8) ms], 90% APD [epicardium layer, (231.5±17.4) ms vs.(211.0±16.3) ms], Ca2+ concentration [epicardium layer, (132.0±12.3) mmol/L vs.(102.3±10.3) mmol/L], expression levels of calmodulin [epicardium layer, (0.724±0.014) vs. (0.563±0.014)] and calmodulin kinase Ⅱ[epicardium layer, (0.759±0.019) vs. (0.589±0.017)] in tanshinone group, (P<0.05~0.01). Conclusion: Tanshinone possesses anti-malignant arrhythmias effect after myocardial infarction. Its mechanism may be regulating calmodulin and calmodulin kinase Ⅱ signal transduction pathway.

OBJECTIVE To explore the characteristics of cochlear hair cell injuries in guinea pigs exposed to blast underpressure(BUP). METHODS The guinea pigs were killed 14 days after exposure to experimental BUP. Their basilar membranes were stained by silver nitrate and the hair cell injuries were quantitatively assessed using the light microscope. RESULTS The outer hair cells(OHCs) of the guinea pigs clearly appeared to be injured following exposure to the experimental BUP at a peak underpressure varying between -22.4kPa and -63.3kPa. The most obvious injury was in the second turn and OHC loss was smallest in the first row and more severe in the second and third rows. OHC loss was seen in all of three experimental groups of animals following exposure to BUP. Furthermore, the higher the peak of underpressure, the more obvious the injury of OHCs. Quantitative morphological analysis of cochlear hair cells showed that the total OHC loss rates in all the experimental groups of guinea pigs exposed to BUP were significantly higher than that of control group animals(P

Objective To explore the effect of radiofrequency heating on type Ⅱ collagen expression in a rabbit model of osteoarthritis. Methods Knee osteoarthritis was induced in the right hind legs of 54 male rabbits using modified Hulth modeling. The rabbits were randomly divided into a model group which was not given any special treatment,a Lugua polypeptide group and a radiofrequency hyperthermia group. The Lugua polypeptide group was injected with Lugua polypeptide;the radiofrequency hyperthermia group was treated with radiofrequency irradiation.Six,12 and 18 days after the treatment, the morphological condition of the rats' right femoral medial condyle cartilages were evaluated using modified Mankins scoring and the type Ⅱ collagen content of the cartilage was detected using a quantitative PCR technique. Results At the same time points after treatment, the average Mankins scores were decreased in all the 3 groups, with that of the model group was significantly higher than those of both of the other groups, and the radiofrequency hyperthermia group's average score was significantly better than that of the Lugua polypeptide group. The average type Ⅱ collagen content was significantly increased in all the 3 groups to various extent (the radiofrequency hyperthermia group > Lugua polypeptide group > model group). For the radiofrequency hyperthermia group, the average Mankins score decreased significantly and the average type Ⅱ collagen content increased significantly as the treatment continued. Conclusion Radiofrequency hyperthermia is superior to Lugua polypeptide for treating knee osteoarthritis,at least in rabbits. Its therapeutic effectiveness may be related to a significant increase of type Ⅱ collagen in the cartilage.
