2.Calcium phosphate nanoparticles as new generation vaccine adjuvants

Adjuvants have been used as critical components of vaccine.They were used to stabilize the antigens and potentiate the immune responses.Dose sparing is also of interests in recent years to be applied in potential pandemic situations or to lower the costs of vaccines.With the emerging nanotechnology in many aspects including the use in biomedical applications,the application of nanomaterials as vaccine adjuvants also attracted a lot of attentions in recent years.With favorable biological properties,such as well-defined and well-formed nanoparticles,biocompatibility,biodegradability and ability to induce humoral immunity and cellular immunity simultaneously,calcium phosphate nanoparticles (CaP) have the potentials to be developed as an immune potentiator to be used as vaccine adjuvants.Here,we reviewed the basic properties of CaP and their applications as vaccine adjuvants with antigens of different modalities such as inactivated virus,protein and naked DNAs.The mechanism of the adjuvanting effects is briefly described.Further the development of CaP as vaccine adjuvants with some designed surface modification could lead to next generation vaccine adjuvants with improved immune potentiating properties and safety profiles.
3.Effects of propofol combined with resveratrol pretreatment on hepatic ischemia-reperfusion injury in rats

Objective To evaluate the effects of propofol combined with resveratrol on hepatic ischemiareperfusion (I/R) injury in rats.Methods One hundred and eight male Sprague-Dawley rats,aged 6-7 weeks,weighing 220-270 g,were randomly divided into 6 groups (n =18 each):sham operation group (group S) ; I/R group; solvent group (group TW-80) ; propofol group (group P) ; resveratrol group (group R) ; propofol combined with resveratrol group (group P + R).The animals were anesthetized with intraperitoneal 10 % chloral hydrate 3.5-4.0 ml/kg.Liver ischemia was produced by clamping the first hepatic portal for 30 min,followed by 12 h reperfusion.In group P,propofol was infused intravenously at 10 mg· kg-1 ·h-1 starting from 10 min before ischemia until the end of operation.In group R,resveratrol 10 mg/kg was injected intravenously at 10 min before ischemia.In group P + R,resveratrol 10 mg/kg was injected intravenously and then propofol was infused intravenously at 10 mg· kg-1· h-1 starting from 10 min before ischemia until the end of operation.The equal volume of normal saline was given instead in groups S and I/R,and the equal volume of TW-80 was given instead in group TW-80.Six rats in each group were chosen at 3,6 and 12 h of reperfusion and blood samples were taken from the superior and inferior vena cava for measurement of serum alanine aminotransferase (ALT) and aspartate amino transferase (AST) activities.The rats were then sacrificed and livers were removed for microscopic examination and for determination of superoxide dismutase (SOD) and myeloperoxidase (MPO) activities,inducible nitric oxide synthase (iNOS) expression,and malondialdehyde (MDA) and nitric oxide (NO) contents in liver tissues.Results Compared with group S,the serum ALT and AST activities,and MDA and NO contents,MPO activity and iNOS expression in liver tissues were significantly increased,and the activities of SOD were decreased in the other four groups (P <0.05).Compared with group I/R,the serum ALT and AST activities,and MDA and NO contents,MPO activity and iNOS expression in liver tissues were significantly decreased,and the activities of SOD were increased in groups P,R and P + R (P < 0.05).Compared with groups P and R,SOD activity was significantly increased,and the other parameters were decreased in group P + R (P < 0.05).The pathological changes were significantly attenuated in groups P,R and P + R compared with group I/R.Conclusion Pretreatment with propofol and resveratrol can attenuate hepatic I/R injury in rats by increasing antioxidation and inhibiting lipid peroxidation and reducing production of endogenous NO,and the combination of the two agents provides better efficacy than etheir alone.
4.Research progress of the relationship between hydrogen peroxide and glioma cell apoptosis

Glioma is the most common intracranial malignant tumor,with high incidence and recurrence rate,high fatality rate and the characteristics of low cure rate.Current treatment is given priority to with surgical treatment,auxiliary comprehensive therapy such as radiotherapy and chemotherapy.Hydrogen peroxide (H2O2) is a intermediate product of the cell oxygen metabolism which is a universal phenomenon in aerobic organisms.H2O2 is indispensable at various stages of tumor cell proliferation,infiltration and metastasis.Studies of the production of H2O2 and its function,the mechanism of apoptosis and the relationship between H2O2 and glioma cells can provide corresponding guidances for looking for a target gene for the treatment of gliomas.
5.Osteogenic ability of cryopreserved bone marrow stromal cells complex in vivo

BACKGROUND: Our previous studies have demonstrated that cryopreserved bone marrow stromal cells (BMSCs) still maintain high survival rate, cell proliferation and osteogenic differentiation potentials after thawing. However, this result needs confirmed in vivo environment. OBJECTIVE: To explore the effects of cryopreserved BMSCs and collagenic membrane BME-10X complex on type Ⅰ collagen synthesis in vivo. METHODS: Beagle dog BMSCs were cultured in vitro and cryopreserved for 12 months, which were thawed and prepared complexes with collagenic membrane. The complexes were cultured with mineralization induction medium or normal medium for 5 days, followed by implanting into nude mice. The specimens were harvested and analyzed by gross observation, histopathological and immunohistochemistry at 4 weeks after implantation. The collagenic membrane cultured with mineralization induction medium served as controls. RESULTS AND CONCLUSION: In the control group, the boundary of collagenic membrane was distinctly, without cell growth around boundary or intra collagenic membrane, additionally, there was little type Ⅰ collagen. In the non-induction group, cells grew into collagenic membrane, trabes-like collagen formed, and type Ⅰ collagen distribution increased at 4 weeks. In the induction group, scaffold degraded, more cells grew, and plenty of collagen formed osteoid-like tissues. The distribution of typeⅠcollagen was obviously increased than that of other groups. The findings demonstrated that cryopreserved BMSCs possess strong osteogenic differentiation potentials after proliferation and induction combined with collagenic membranes in vitro.
6.Clinical observation of pregabalin in the treatment of painful diabetic peripheral neuropathy

Objective To observe the efficacy of pregabalin in the treatment of painful diabeticperipheral neuropathy.Methods In a 8-week study,50 patients were randomly assigned into two groups:the therapy group and the control group.The patients in the therapy group were treated with pregabalin 150 mg/d,and the patients in the control group were treated with carbamazepine 200 mg/d.The dose of pregabalin was increased to 300 mg/d till pain was alleviated in the therapy group,and the dose of carbamazepine was increased to 600 mg/d till pain was alleviated in the control group.The therapeutic effect and VAS score in the two groups were observed and evaluated.Results After 1 and 8 weeks' treatment,the therapeutic effect of therapy group were significantly superior to the control group ( At 1 week:U =2.028,P =0.046 ; At 8 week:U =3.540,P <0.001 ).After 1 and 8 weeks' treatment,the VAS scores of therapy group were significantly lower than the control group ( At 1 week:5.13 ± 1.76 vs.6.74 ± 1.52,P < 0.05 ; At 8 week:1.13 ± 0.45 vs 3.27 ± 1.04,P < 0.05).There were 5 patients (20%) and 10 patients (40%) had adverse reaction in the therapy and the control groups respectively.Conclusion Pregabalin is effective and safe for the treatment of painful diabetic peripheral neuropathy.
7.Pathological changes of the livers from 39 patients with hepatic failure

Objective To explore the pathological changes of the livers from hepatic failure (HF)patients and its association with clinical disease stages.Methods Thirty-nine patients with liver failure caused by HBV infections were investigated,and none accompanied with hepatocellular carci- noma.The sections of tissue were taken from the liver after liver transplantation and stained with he- matoxylin eosin(H&E)or RT(reticular fiber)staining.The pathological features were analyzed and compared between the clinical and pathological diagnosis.Results 1.The range and the grade of the pathological changes were all well-proportioned in the whole liver but quite asymmetrical in the same spicemen.2.4 cases with clinical diagnosis of cirrhosis(active stage)were in accordance with the pathological diagnosis.Only 17 in 35cases can be pathologically diagnosed as chronic severe hepatitis (SH),while the other 18 cases were pathologically diagnosed as cirrhosis(active stage).Conclu- sion There were a great inconsistency between the clinical and pathological diagnosis.

9.Improved pelvic floor reconstruction with polypropylene mesh for repair of pelvic organ prolapse

BACKGROUND:Synthetic patch as an important substitute to pelvic tissue can replace damaged pelvic fascia tissue, and has been widely used in the pelvic floor reconstruction. OBJECTIVE:To observe the efficacy and complications of pelvic floor reconstruction with mesh, and to explore its safety and effectiveness. METHODS: Forty-five pelvic organ prolapse female patients were divided into two groups according to patient’s wilingness: experimental group, pelvic floor reconstruction with mesh (n=25); control group, transvaginal hysterectomy combined with vaginal wal repair (n=20). Perioperative conditions were recorded, and uterine prolapse staging, complications, pelvic floor function, pelvic discomfort, as wel as an objective cure rate were evaluated in the two groups during the folow-up. RESULTS AND CONCLUSION:Compared with the control group, the experimental group was characterized as shorter operative time, less amount of bleeding, milder infections, and faster recovery (AlP < 0.05). At 6 months of folow-up, the score on uterine prolapse staging was higher in the experimental group than the control group; at 12 months, the scores on pelvic floor function and pelvic discomfort as wel as the incidence of complications were significantly lower in the experimental group (P < 0.05), but the objective cure rate was higher compared with the control group (P < 0.05). These findings suggest that pelvic floor reconstruction with mesh for pelvic organ prolapsed can reduce the operative time and blood loss and promote postoperative recovery. Meanwhile, it can significantly improve pelvic floor function, pelvic discomfort, postoperative complications, the rate of exposure, and the objective cure rate, which is safe and effective during the short-term folow-up.
10.Expression of the basic fibroblast growth factor gene after being transfected into rat bone marrow mesen-chymal stem cells

Objective Bone marrow mesenchymal stem cells ( BMSCs) , a kind of stem cells with multiple differentiation po-tentials, exist in the bone marrow and other organizations.This study aimed to investigate the repairing effect of the exogenous basic fi-broblast growth factor ( bFGF) against chronic obstructive pulmonary disease ( COPD) and its action mechanism, and to determine the expression of the bFGF gene in transfected rat BMSCs. Methods BMSCs were isolated, cultured and identified.The recombinant plasmid bFGF-pcDNA3.1 was constructed and sequenced.Liposome-mediated bFGF-pcDNA3.1 plasmid was transfected into the BM-SCs of the rat (bFGF-pcDNA3.1 transfection group), liposome-mediated pcDNA3.1 transfected into the BMSCs (pcDNA3.1 transfec-tion group) , and untransfected BMSCs used as the control.G418 screening was performed for 14 days.The gene and protein expres-sions of bFGF were determined by qRT-PCR and Western blot. Results The full-length sequence of the bFGF gene was consistent with that of the GenBank.The expression of the bFGF gene was significantly higher in the bFGF-pcDNA3.1 transfection group (7.028 ±0.568) than in the pcDNA3.1 transfection group (1.000 ±0.082) and the non-transfection control (1) (P<0.01), but with no statistically significant difference between the latter two groups (P>0.05).The expression of the bFGF protein was also re-markably higher in the bFGF-pcDNA3.1 transfection group (1.017 ±0.054) than in the pcDNA3.1 transfection group (0.217 ± 0.009) and the non-transfection control (0.165 ±0.013) (P<0.05), with no statistically significant difference between the latter two groups (P>0.05). Conclusion Mediated by the liposome reagent, the recombinant eukaryotic expression vector bFGF-pcD-NA3.1 can be transfected into rat BMSCs and expresses the bFGF gene and protein.
