2.The validation study of Chinese version of Sunnybrook of the facial nerve grading system

Objective We have revised the Sunnybrook facial nerve grading system which has been widely recog?nized in the world to a Chinese version and verified its credibility and validity. Methods First we translated and edited the Sunnybrook facial nerve grading system using Chinese.Nine doctors, who were selected from different hospitals, then scored twice on the same video of 90 patients with facial nerve palsy using the Chinese version of the Sunnybrook facial nerve grading system and grading standard separately at one month interval. The consistency and the repeatability of the Chinese version of the Sunnybrook facial nerve grading system were evaluated. Results The Cronbach 's alpha value was 0.92 and the average coefficient of repeatability (CR) was 12.72, indicating that the Chinese version of the Sunny?brook facial nerve grading system had a high degree of internal consistency and repeatability. The ICC values on the as?sessment and evaluation for the internal measurement were 0.97 and 0.98, respectively. The scored results were compara?ble between doctors who were not familiar with this disease and those who were very familiar with the disease. The aver?age ICC was 0.97(95%CI:0.95~0.99). Conclusions The Sunnybrook facial nerve grading system has good reliability and consistency and hereby recommend Chinese doctors to use it.
3.Study on the management and its effectiveness evaluation of 70538 patients with serious mental illness at home in Gansu province

Objective To analysis the family management of patients with serious mental illness in Gansu prov-ince, and evaluate its effectiveness to lay the foundation for the better community-based rehabilitation management and treatment for the people with serious mental illness. Method According to Treatment-management specification of severe mental illness (2012) and specification of national basic public health services (2011), the information such as home ther-apy, medicine taken, and dangerous behavior happened were collected from patients who were confirmed as serious men-tal illness in January 2015 to April 2016 in Gansu province. Result Up to April 30th, 2016, totally 70538 serious mental illnesses were identified, and about 81.10%(57195) were under management. The rate of treatment of serious mental ill-ness was 69.9%(48009/68663) in Gansu province, and Bipolar affective disorder (76.0%) was the highest (P<0.001). The rate of taking medication in patients under family-management was 42.16%(24113/57195), and the rate of taking regular medication was 24.74%(14150/57195). The rate of taking medication in Paranoiac psychosis patients (51.98%) was the highest (P<0.001), and the rate of taking regular medication in Mental disorders caused by epilepsy (31.02%) was the highest (P<0.001). Conclusion The rate of family-management, treatment, taking medication and take regular medica-tion of serious mental illness in Gansu province are all below the national average level. A special training should be giv-en to medical professionals to strengthen their capacity to treat serious mental illness.
4.Effect of Post-conditioning in Brain Injury Induced by Myocardial IR on Inflammatory Factor and GFAP

Objective:To evaluate the effect of post-conditioning in brain injury induced by myocardial I/R on inflammatory factor and GFAP.Methods:Male Sprague-Dawley rats were randomly allocated into 3 groups (n=8):group Sham,group IR,group IPost.Myocardial IR was induced by occlusion of the anterior descending branch of the left coronary artery for 30 min.group IPost received 3 cycles of 10 s reperfusion followed by 10 s ischemia at the end of myocardial ischemia.The rats were sacrificed at 120 rain of reperfusion and the brains were removed for microscopic examination,inflammatory factors and GFAP.Results:Compared with group Sham,IL-6,IL-8 were significantly increased,IL-10 was down-regulated in group IR(P<0.01).Post-conditioning can decrease IL-6,IL-8 and up-regulated IL-10(P<0.01).When compared with group Sham,the expression of GFAP was higher in group IR(P<0.05),however,the GFAP in group IPost is the most among these three groups(P<0.01).Conclusion:Post-conditioning could protect brain by decreasing inflammatory factors,increasing GFAP,which both from brain injury induced by myocardial ischemia reperfusion.
5.LMO2 in prostate stromal cells promotes prostate cancer cells proliferation and invasion through ;paracrine of IL-11 and FGF-9

Background and purpose:The previous research has found that the prostate stromal cells derived from different prostate zones have distinct effect on prostate epithelial cells. We also revealed that LMO2 protein was highly expressed in PZ stromal cells (PZSCs) and prostate cancer associated fibroblasts (CAFs) compared with TZ stromal cells. This study investigated the effect of LMO2 protein in prostate stromal cells on proliferation and invasion of prostate cancer PC-3 cells and its mechanisms. Methods:Lentivirus overexpression vectors were used to establish LMO2-overexpressed prostate WPMY-1 stromal cell line. shRNA plasmids were used to suppress LMO2 in CAFs. LMO2 mRNA and protein level of both WPMY-1 and CAFs were evaluated by real-time fluorescent quantitative polymerase chain reaction (RTFQ-PCR) and Western blot. Then, PC-3 cells were co-cultured with different prostate stromal cells and the in vitro proliferation and invasion of PC-3 were measured by CCK-8 and matrigel invasion assays respectively. Results:When co-cultured with LMO2-overexpressed prostate stromal cells, both proliferation and in-vasion of PC-3 were improved. However, when co-cultured with CAFs which have inhibited expression of LMO2, the proliferation and invasion of PC-3 were reduced. The protein array proifling found that both interleukin-11 (IL-11) and ifbroblast growth factor-9 (FGF-9) were enhanced extensively in the supernatant collected from LMO2-overexpressed WPMY-1 cells. Conclusion:The expression of LMO2 in prostate stromal cells could be responsible for development of prostate cancer. Paracrine of cytokines, such as IL-11 and FGF-9, from LMO2-overexpressed stromal cells had effects on the proliferation and invasion of prostate cancer cells.

7.Self-made Kirschner wire hook for the treatment of severe displaced proximal humeral fractures in children.

OBJECTIVETo explore the clinical efficacy of self-made Kirschner wire hook in the treatment of severely displaced proximal humerus fractures in children.
METHODSFrom January 2007 to February 2012,35 children with severely displaced proximal humerus fractures were treated with self-made Kirschner wire hook fixation,including 25 boys and 10 girls with an average age of 13.2 years old ranging from 5 to 17. The duration from injury to operation was 1 to 10 days with an average of 4.5 days. Preoperative diagnoses were confirmed by X-ray films as type III in 26 cases and type IV in 9 according to the Neer-Horwitz classification of the proximal humerus fractures. All fractures were close fracture without nerve or vascular injuries. Anatomical reduction was achieved by open reduction and the fractures were fixed by self-made Kirschner wire hook. Intra-operative and postoperative complications, postoperative radiographic examination, upper extremity length and range of shoulder motion were observed and recorded. Neer score system was used to evaluate shoulder function.
RESULTSAll patients were followed up from 6 to 18 months with an average of 12.1 months. The mean Neer score of the injured side was 94.2 +/- 4.8 (ranged, 84 to 99), 30 patients got an excellent result and 5 good according to the Neer scores. The X-ray showed all the fractures healed without shortening deformity or epiphyseal arrest at early stage. There were no complications related to the implant including loosening or breakage of the self-made Kirschner wire hook. The range of shoulder motion in the injuried side was similar to the normal side, except in abductor lift. All the patients could participate in normal physical activities.
CONCLUSIONThe method of open reduction and fixed with self-made Kirschner wire hook is a safe,effective,convenient treatment for severely displaced proximal humeral fractures in children.

2', 3', 5'-Tri-O-acetyl-N6-(3-hydroxylaniline)adenosine (WS070117) is a derivative compound of natural product cordycepin. It has significant lipids regulating activity and low toxicity which has been proved by in vitro and in vivo experiments. In this study, 1H NMR-based metabolomics was used to investigate the dose-related effects of WS070117 on hyperlipidemia of high-fat-fed hamsters. The hyperlipidemic hamsters were administrated with six different doses of WS070117, including 3, 12, 50, 100, 200 and 400 mg x kg(-1) x d(-1). 1H NMR spectra of hamster serum were visually and statistically analyzed using two multivariate analyses: principal component analysis (PCA) and orthogonal partial least squares-discriminant analysis (OPLS-DA). As a result, WS070117-treated groups showed dose-related regulation of metabolites associated with lipid metabolism, choline metabolism and glucose metabolism. The dose of 3 mg x kg(-1) x d(-1) of WS070117 only exhibited a little lipids regulating activity. However, the doses of 12 and 50 mg x kg(-1) x d(-1) of WS070117 both regulated the contents of metabolites to reverse significantly toward normal levels. When the dose of WS070117 reached 100 mg x kg(-1) x d(-1), it was more effective than positive control drugs. The work suggested that NMR-based metabolomics might be a valuable approach to evaluate dose-related effects of lipids regulating compounds.

9.Medical students' cognition on laboratory biosafety

Objective To investigate cognitive status of medical students of a medical university on laboratory biosafety,and provide basic data for laboratory biosafety management in Chinese universities.Methods 900 full-time undergraduate medical students were chosen by cluster random sampling,questionnaires were filled out in by them.Results 900 questionnaires were distributed,877 (98.21%) valid questionnaires were obtained,49.03% (n =430) were from sophomores,50.97 % (n =447) from juniors,148 (16.88 %) students have ever participated in students'scientific research.The overall awareness rate of laboratory biosafety was 58.72%,only 32.16% of students understood the detailed contents of laboratory biosafety regulations,only 8.21% of students have received training in laboratory biosafety;the awareness rate of laboratory biosafety cabinet was only 14.14%,only 7.75% of students knew which operation should be performed in biosafety cabinet;28.28% of students could deal with waste according to the rules,68.19% of students were able to identify warning signs of biological hazard;92.82% of the students thought that laboratory biosafety-related courses should be set up.The overall awareness rate of laboratory biosafety knowledge and safety behavior was low,which were 42.65% and 41.96% respectively,juniors was higher than that of sophomores(P<0.05);in the aspect of chemical hazards and biological hazards,students with scientific research experience scored higher than those who did not participate in scientific research(all P<0.05).Conclusion Medical students' cognition on knowledge of laboratory biosafety is not optimistic,it is imperative to strengthen the management of education and publicity of laboratory biosafety.
10.Effects of penehyclidine hydrochloride on acute lung injury induced by blunt chest trauma-hemorrhagic shock and resuscitation in rats

Objective To investigate the effects of penehyclidine hydrochloride (PHC) on acute lung injury induced by blunt chest trauma-hemorrhagic shock and resuscitation in rats.Methods Forty male SpragueDawley rats,aged 8 weeks,weighing 250-300 g,were randomly assigned into 4 equal groups (n =10 each) using a random number table:sham operation group (S group),blunt chest trauma combined with hemorrhagic shock and resuscitation group (group THSR),PHC for prevention group (group P1)and PHC for treatment group (group P2).ALI was induced by dropping a 300 g weight onto a precordial protective shield to direct the impact force away from the heart and toward the lungs in anesthetized rats in THSR,P1 and P2 groups.Blood was withdrawn via the femoral artery 5 min later until MAP was decreased to 35-45 mmHg within 15 min and maintained at this level for 60 min,followed by resuscitation.In P1 group,PHC 2 mg/kg was injected intravenously at 30 min before blunt chest trauma.In P2 group,PHC 2 mg/kg was injected intravenously at 60 min after hemorrhagic shock.At 6 h after the model was established,arterial blood samples were obtained for blood gas analysis and for measurement of concentrations of interleukin-6 (IL-6) and interleukin-1β (IL-1β) in serum by ELISA.Oxygenation index (OI) was calculated.The animals were sacrificed and bronchoalveolar lavage fluid (BALF) was collected for determination of white blood cell count and protein concentrations.Lungs were removed for examination of pathological changes and ultrastructure and for determination of Toll-like receptor (TLR4) and phosphor-p38 mitogen activated protein kinase (p-p38MAPK) expression (by Western blot).Results Compared with group S,PaO2 and OI were significantly decreased,PaCO2,protein concentrations in BALF,white blood cell count,and IL-6 and IL-1β concentrations in serum were increased,and TLR4 and p-p38MAPK expression was up-regulated in THSR,P1 and P2 groups.Compared with group THSR,PaO2 and OI were significantly increased,PaCO2,protein concentrations in BALF,white blood cell count,and IL-6 and IL-lβ concentrations in serum were decreased,TLR4 and p-p38MAPK expression was down-regulated in P1 and P2 groups.No significant differences were found in the parameters mentioned above between P1 and P2 groups.Conclusion PHC can mitigate acute lung injury induced by blunt chest trauma-hemorrhagic shock and resuscitation in rats,and inhibited activation of TLR4/ p38MAPK signaling pathway and attenuated inflammatory responses are involved in the mechanism.
