1.Influence of nerve growth factor on the expression of nestin and neural stem cell type in rats with focal cerebral ischemia

AIM: To investigate the effect of nerve growth factor (NGF) on the proliferation of neural stem cell in adult rats with focal cerebral ischemia. METHODS: The experiment was conducted in the Center of Science and Experiment, Jinzhou Medical College from January to July 2006. Totally 72 healthy SD rats were randomly divided into sham-operation group, model group and NGF treatment group with 24 rats in each group. Focal cerebral ischemia models were prepared by Logna et al modified thread occlusion method, and the functional evaluation was performed after the animals were awake for 2 hours to select the rats whose nervous function reached grade 2 or above. The sham-operation group was treated like the model group except thread occlusion. 1 000 ?g/kg NGF was administered in the NGF treatment group immediately after ischemia, once a day. After the animals were executed at days 1, 3, 7 and 14 days, immunohistochemical and double immunofluorescence labeling were adopted to observe the effect of NGF on the expression of nestin and the cell type after ischemia. RESULTS: All 72 rats were involved in the result analysis. ①Round or ellipse nestin positive cells were found in both the model group and NGF treatment group. The number of nestin positive cells in the NGF treatment group was remarkably more than in model group except 1 day after ischemia, moreover, the number of the cells in the two groups was more than in the sham operation group [model group: (3.47?0.5), (5.13?1.14), (13.95?3.56), (8.97?2.08); NGF group: (3.81?0.66), (9.88?2.08), (19.87?3.86), (26.17?2.90); sham-operation group: 0, P
2.Treatment of severe intestinal flora alteration induced by the Cefoperazone with the filtrate of fresh human dejecta

Objective To observe the effect of filtrate of normal human fresh dejecta on intestinal flora alteration.Methods 5 severe patients who treated with Cefoperazone suffered in intestinal flora alteration, 4 cases with candida albicans ans, 1 with Welch's basillus. After diagnosing, preserving clyster was done with filtrate of normal human fresh dejecta.Results 1 or 2 days after the treatment, the intestinal flora alteration disappeared.Conclusion The filtrate of normal human fresh dejecta is effective on the treatment of intestinal flora alternation.
3.Studies on Establishment of Co-expression System and Function of Human Complement Regulatory Proteins MCP and CD59

Recombinant expression vector pcDNA3-MCPCD59DP containing human membrane complement regulatory proteins(hCRPs) MCP and CD59 cDNA was constructed successfully by using two independent promoters.After transfected into NIH3T3 cells with calcium phosphate-DNA precipitate method,NIH3T3 pcDNA3-MCPCD59-DP transfectants were obtained by G418 selection.Extraneous genes integration was identified by PCR.The co-expression of human CD59 and MCP at both mRNA and protein levels was confirmed by using RT-PCR and Western blot analysis.Human MCP and CD59 cDNA were integrated in NIH3T3 pcDNA3-MCPCD59-DP genomic DNA after continuous 30 times passages,indicating that NIH3T3 pcDNA3-MCPCD59-DP were stable cell lines.Human complement-mediated cytolysis assays showed that NIH3T3 cells transfected stably with pcDNA3-CD59,pcDNA3-MCP,and pcDNA3-MCPCD59-DP were protected from C-mediated damage and co-expressed human MCP and CD59 provided more excellent protection against C-mediated attack as compared with either CD59 or MCP expressed alone.The dicistronic vector represents an effective and efficacy strategy to overcome C-mediated damage and has potential therapeutic value for effectively controlling complement activation and finally for preventing hyperacute rejection in clinical gene therapy.
4.Coastal situation analysis of hypertension among rural population and its correlation with prevalence of metabolic syndrome

Objective To explore the coastal state of hypertension among rural population and its relationship between the prevalence of metabolic syndrome(MS).Methods Lianyungang coastal rural residents 11089 people for primary hypertension epidemic learn research,age qualified for 45~75age,measurement height,body quality,waist,blood pressure,lipid and fasting blood glucose,respectively in accordance with international diabetes Union(IDF)and CDS of MS diagnosis standard calculation illness rate,and on gender,age,and hypertension level(Ⅰ,and Ⅱ,andⅢ,level),effects factors for correlation analysis.Results 11089 people(male 3985 people,female 7104 people)in the IDF standard MS total illness rate for 44.3%(male,and female MS illness rate respectively for 25.4%,and 54.9%),CDS standard MS total illness rate for 37%,two species diagnosis standard of meet rate for 79.9%;except blood glucose outside,female residents waist,TG,and HDL-C,indicators exception occurs rate obvious than male(x2=1399.198,554.801,652.110,all P<0.01).Conclusion Lianyungang coastal MS prevalence in rural areas is higher than the general average in the middle-aged;female patients with hypertension and obesity are very high risk population of MS;hypertension pathogenesis is longer,higher prevalence of MS.

Objective To optimize the particle size of Huoxue powder by contents comparison of emodin,phellodendrine,berberine and jatrorrhizine before and after permeabilized skin absorption of Huoxue powder in different particle size of 0.150,0.075,0.048,0.038 mm.Methods The contents of emodin,phellodendrine,berberine and jatrorrhizine in transparent absorbent fluid of Huoxue power in different particle size within 24 hours were measured by ultra performance liquid chromatography tandem mass spectrometry,and optimize its particle size by contents comparison of the effective components.Results The contents of the active components in Huoxue power with the particle size of 0.075 mm were high before and after percutaneous absorption.Conclusion Particle size of 0.075 mm is best for Huoxue powder.
8.Evaluation of Statistical Methods for Articles Published in《Chinese Circulation Journal》

Objective: To evaluate the applications and problems of statistical methods for articles published in《Chinese Circulation Journal》. Methods: We reviewed the statistical methods for original articles published in《Chinese Circulation Journal》in 2010, 2011 and 2015; general status with problems was analyzed and the situations were compared among different years. Results: A total of 404 articles were enrolled, statistical hypothesis testing was performed in 366 of them, 32 had descriptive statistics only and 6 articles were meta-analysis. The most frequently used statistical methods were student'st-test (57.4%), followed by chi-squared analysis (56.7%), analysis of variance (35.2%) and regression analysis (23.0%). The main statistical problems in Methods included in described method un-matched to really used method and insufifcient or wrong description. The proportion of insufifcient or wrong description in 2010 and 2011 was 36.6% (71/194), while in 2015 was 26.2% (55/210),P=0.02. The major wrong application of statistical approach was ignoring design with improper method and using unsatisifed precondition for speciifc analysis. The proportion of improper statistical method application in 2010 and 2011 was 19.1% (33/173), in 2015 was 19.7% (38/193),P=0.88. Conclusion: The quality of statistics was improved in 2015 than 2010 and 2011 for articles published in《Chinese Circulation Journal》; while there were still problems at certain degree, enhanced statistical review should be conducted in medical journals.
9.Study on the Quality Standard of Huoxuesan

OBJECTIVE:To establish the quality standard of Huoxuesan. METHODS:Microscopic identification method was adopted for the qualitative identification of Rhizoma pinelliae,Eupolyphaga seu,Phellodendron amurense and Fructus kochiae in the preparation. HPLC was adopted for the contents determination of phellodendrine,polydatin,jatrorrhizine,berberine and emo-din:the column was ZORBAX Eclipse Plus C18 with methanol-0.1% phosphoric acid(gradient elution)at a flow rate of 0.4 ml/min,the detection wavelength was 240 nm,column temperature was 30 ℃,and the injection volume was 5 μl. RESULTS:Micro-scopic identification map of R. pinelliae,E. seu,P. amurense and F. kochiae was clear. The linear range was 0.159-5.073 μg(r=0.997 4)for phellodendrine、0.149-4.761 μg(r=0.999 9)for polydatin、0.239-7.649 μg(r=0.995 5)for jatrorrhizine、0.165-5.268 μg (r=0.997 2)for berberine、0.012-0.382 μg(r=0.999 9)for emodin;RSDs of precision,stability and reproducibility tests were low-er than 3.0%;recoveries were 98.9%-104.1%(RSD=1.9%,n=6),96.1%-101.7%(RSD=2.5%,n=6),99.6%-105.1%(RSD=2.6%,n=6),90.3%-98.2%(RSD=2.9%,n=6)and 92.9%-96.4%(RSD=2.0%,n=6)respectively. CONCLUSIONS:The es-tablished standard can be used for the quality control of Huoxuesan.
10.Radiofrequency hyperthermia in combination with continuous intra-arterial infusion chemotherapy in the treatment of advanced colorectal cancer

Objective To observe the therapeutic efficacy of continuous arterial infusion chemotherapy combined with radiofrequency hyperthermia in the treatment of patients with advanced colorectal cancer. Methods A total of 21 advanced colorectal cancer patients with various distant metastasis were treated by the combined continuous intra arterial infusion chemotherapy and radiofrequency hyperthermia. For the chemotherapy, a dose of 200mg/m 2 surface body area of calcium folinate (CF) was given for 1 to 3 days. 80mg/m 2 of cisplation was infused intravenously for the first day and replaced by etoposide Vp 16, 60mg/m 2 for 1~3days in patients with renal dysfunction. For intra arterial infusion, a dose of 500mg/m 2 5 FU 2 was given for 72 hours. For patients with liver metastases, chemoembolization(ADM 30mg/m 2+MMC 6mg/m 2 mixed with ultra liquefied lipiodol) was carried out. Radiofrequency hyperthermia with a frequency of 41MHz was performed on the second day after chemotherapy. Results Response rates were assessed by CT scan and ultrasonography. The overall response rate of the cases was 66.67%. No serious side effect or complication was found in the course of chemotherapy. Local pain and lipid nodule were occasionally observed in some patients after hyperthermia. Conclusion Continuous intra arterial infusion chemotherapy in combination with radiofrequency hyperthermia is an useful and safe method for the treatment of patients with advanced colorectal cancer.
