1.The current status of lipid-lowering therapy in patients with coronary heart disease

Objective To investigate the plasma lipid levels,ratio achieving the optimal goals and its possible influencing factors in patients with coronary heart disease,and to elaborate the current status of lipid-lowering therapy.Methods The incipient plasma lipid level and status of therapy from the case history data of the 101 patients with CHD who were followed up after discharge were collected.The data including blood lipid levels and medications were recorded and analyzed.Results (1)60.6%patients with CHD had elevated LDL-C,but actually 80.7% had taken lipid-lowering drugs in hospital.(2)Lipid-lowering therapy was used in 68.3% of patients during the follow-up period.According to NCEP ATP Ⅲ,the ratio achieving the optimal LDL-C goals was 65.3%.(3)The ratio achieving the optimal goals was related to income and smoking(P0.05).(4)None of the patients had apparent elevated alanine aminotransferase(ALT)or creatine kinase(CK).Conclusion The current status of CHD lipid-lowering therapy remains a big gap.The ratio achieving the optimal goals is related to economy status and life style(smoking),and influenced by the physician quality of care,patient compliance,drug efficacy and cost–benefit.Long-term lipid-lowering therapy is safe when the drug use and dosage are appropriate.
2.The expressions of collagen Ⅰ and Ⅲ during mouse tooth germ development

Objective:To investigate the temporal and spatial distrib ut ions of collagenⅠ and Ⅲ during mouse tooth germ development and their function s during tooth mineralization.Methods:Immunohistochemistry stain ing technique was used to test the expressions of collagen Ⅰ and Ⅲ during mous e tooth germ development. Results:collagen Ⅲ was positive in or al epithelial cells in bud stage,in oral epithelial cells and in stellate reticu lum cells in cap stage. During bell and differentiation stage,collagen Ⅲ was positive in oral epithelial cells, stellate reticulum cells, dental papilla cell s and dental sac cells. During P2-10 d(crown development stage), collagen Ⅲ was expressed possitively in ameloblasts,enamel matrix,odontoblasts,predentin, dental papilla cells,dental sac cells and pulp tissues. During P10-30 d(toot h root development stage),collagen Ⅲ was strongly positive in Hertwig's epithe lial root sheath, cementum, alveolar bone and periodontal ligament cells apart f rom above mentioned cell types. CollagenⅠ was not expressed in bud stage and wa s positive in oral epithelial cells,stellate reticulum cells in cap stage. Durin g bell and differentiation stage,collagen Ⅰ was positive in oral epithelial ce lls, stellate reticulum cells, dental papilla cells and dental sac cells.After P2 d (crown and root development stage), the distribution and expression of co llagen Ⅰ were similar to those of collagen Ⅲ.Conclusions:Coll agenⅠand Ⅲ are involved in tooth germ and tooth tissue development. But the fu nction of collagenⅢ is more extensive than that of collagenⅠ.
3.The spatiotemporal expressions of ADAM28 in mouse tooth germ development

Objective:To investigate spatiotemporal expression of ADAM28 in mouse tooth germ development.Methods:Immunohistochemistry and image analysis technique were used to observe the expressions of ADAM28 at mouse tooth germ development stages.Results:Different expression levels of ADAM28 at tooth germ development stages were observed.At cap stage,ADAM28 was found strongly positive in oral epithelial,stellate reticulum cells of enamel organ,basement membrane,dental papilla cells and dental sac cells.At late bell stage,positive staining was found in ameloblasts,enamel matrix,epithelial root sheath and dental papilla cells.At crown and root development stage,positive staining for ADAM28 was detected in ameloblasts,odontoblasts,cementoblasts,epithelial root sheath,dental papilla cells and dental sac cells.Conclusion:ADAM28 participates in crown and root morphogenesis process ranging from bud stage to late bell stage and from matrix secretion to sclerous tissue formation.It might play an important role in early formation,proliferation and differentiation of odontogenic mesenchymal cells.
4.Mechanical proprieties of four cements at different mixing ratios

To investigate the influence of mixing ratios on the hardness,wear resistance and compressive strength of the cements clinically used,Zinc Oxide Eugenol cement,Glass ionomer cements,Zinc Phosphate Cement,and Dual-curing composite cement were mixed at different ratios separately,then made into test pieces.Vickers hardness,wear resistance and compressive strength of these test pieces were measured by related machines.The results showed that vickers hardness,wear resistance and compressive strength of Zinc Oxide Eugenol cement,Glass ionomer cements and Zinc Phosphate Cement reduced with the decrease of powder/liquid ratios.As to Dual-curing composite cement,Vickers hardness,wear resistance and compressive strength decreased with the difference between A/B ratio and 1∶1 increase.Conclusively,mixing ratios could influence the mechanical proprieties of the cements clinical used,so the use of cements should be under the instructions of the product.
5.Preparation and identification of a polyclonal antibody against tooth development related gene-adam28 product

Objective:To prepare and identify a polyclonal antibody againstadam28 gene product.Methods:Theprotein coding region of ADAM28 was amplified by RT-PCR and cloned into pMD18-T Vector to produce the newconstruct, pMD18-T-adam28. The cloned ADAM28 segment was cut with two restriction enzymes and theadam28fregment was directed into the prokaryotic expression vector, pGEX-4T-1,to produce the expression vector pGEX-4T-adam28. The recombinant plasmid was transformed intoE. coliDH5?and GST-ADAM28 fusion protein was ob-tained after the inducement by IPTG. The fusion protein was extracted and purified by SDS-PAGE,and the newpro-tein band of 35 300 was isolated as antigen, the antigen was injected into rabbits to produce polyclonal antibody a-gainst ADAM28 product.Results:The expression vector pGEX-4T-adam28 was constructed successfully,and GST-ADAM28 fusion protein was obtained. The rabbit serum containing polyclonal antibody against ADAM28 productwas obtained and the antibody was purified by salting out method. Western blot analysis displayed that the antibodyhad high specificity. ELISA analysis confirmed that the titer for the antibody reached 1∶16 000.Conclusion:Thepolyclonal antibody against ADAM28 product with high titer is successfully prepared,it may be used for further studyof the role and expression of ADAM28 during tooth development.
7.A clinical analysis of hemophagocytic syndrome in autoimmune diseases

Objective To analyze the clinical features of patients with hemophagocytic syndrome (HPS) in autoimmune diseases (AID). Methods We collected the data of 11 patients with AID complicated with HPS in Peking Union Medical College Hospital from 2004 to 2009. The underlying diseases, clinical features, laboratory findings and treatment outcomes were retrospectively analyzed. Results Of the 11 patients,3 were male,8 were female. Mean age was (30. 7 ± 18. 3) years. The underlying diseases included Still disease ( n = 4 ), systemic lupus erythematosus ( n = 3 ), and rheumatoid arthritis, primary Sj(o)gren's syndrome, Wegener granulomatosis and Crohn disease in each one case. HPS was associated with the onset of AID ( n = 4), active infection alone ( n = 1 ) and both factors ( n = 6 ). HPS was clinically characterized by high fever ( 100% ), hepatosplenomegaly ( 72. 7% ) , lymphadenopathy ( 63.3% ) and central nervous system involvement (36. 3% ). 4 patients presented with disseminated intravascular coagulation(DIC) (36. 3% ). Laboratory data mainly manifested with cytopenia ( 100% ), liver dysfunction ( 100% ), hypofibrinogenemia ( 62. 5% ), hypertriglyceridemia ( 81.8% ), serum ferritin > 500 μg/L (100%), low NK-cell activity(80% ) and hemophagocytosis in bone marrow( 100% ). Based on treating underlying infections and use of corticosteroids and immunosuppressive agents in combination with intravenous immunoglobulins(IVIG) therapy, 5 patients recovered , 6 patients died. The mortality rate was 54. 5%. DIC were associated with mortality ( r = 0. 69, P = 0. 019 ). Conclusion The episode of HPS always occurs simultaneously with multiple system involvement that was often difficult to distinguish from active AID. The present of DIC on HPS related with poor prognosis and high mortality. Corticosteroids and immunodepressant and IVIG may improve the prognosis of HPS, while anti-infection therapy is very important and necessary for the patients accompany with active infection.
9.Meta-analysis of dosemetric comparision between helical tomotherapy and intensity-modulated radiotherapy for early-stage postoperative breast cancer

Objective To compare the dosemetry between helical tomotherapy (HT) and intensity-modulated radiotherapy (IMRT) for early-stage postoperative breast cancer and provide more valuable evidences to the clinical researches. Methods Clinical trails of dosimetric comparing between HT and IMRT for early-stage breast cancer were obtained from PubMed, Embase, Sciencedirect, CNKI, VIP and Wanfang databases, evaluated and analyzed with the Cochrane Collaboration's RevMan 5.2 software. Results 10 studies were included with a total of 135 patients. Compared to IMRT plans, HT plans provided a significantly better conformity index (P<0.000 1), mean (P<0.000 01) and maximal dose (P=0.003) of the planning target volume (PTV). HT plans had a lower heart maximal dose (P=0.005), V20 (P=0.05), V30 (P=0.003), and ipsilateral lung maximal dose (P=0.003), V20 (P=0.02), as while as had a higher contralateral breast V5 (P=0.01), mean (P=0.05) and maximal dose (P<0.000 01). There was no significantly difference between HT and IMRT plans for homogeneity index of PTV, heart V5, V10, mean dose, ipsilateral lung V5, V10, V30, mean dose, contralateral breast V10, contralateral lung mean and maximal dose (all P >0.05). Conclusion Compared to IMRT plans, HT plans have the dosimetry superiority for early-stage breast cancer with significantly better coverage and dose conformity while maintaining lower doses to high risk organs.
10.Determination of Salicylic Acid and the Related Substances in Salicylic Acid Ointment by HPLC

OBJECTIVE:To determine the contents of salicylic acid and the related substances in salicylic acid ointment by HPLC.METHODS:The determination was performed on Kromasil ODS-C18 chromatographic column with mobile phase consisted of methanol-water(adjust pH=2.5 using acetic acid,50∶50)at a flow rate of 1.0 mL?min-1.The sample size was 20 ?L and the detection wavelength was set at 270 nm.RESULTS:The main peak and the related substances were well-separated.The linear range of salicylic acid was 10.014~100.14 ?g?mL-1(r=0.999 9)with an average recovery of 99.35%(RSD=0.99%)and a lowest detectable limit of 5 ng?mL-1,the labeled amout of related substances all less than 1.01%.CONCLUSION:The method developed in our study is simple in operation,accurate in results,specific and suitable for the content determination of salicylic acid ointment.
