Objective To study the DNA damage effect of the organic extracts from Chlorinated domestic sewage coming from three different places in vivo and in vitro. Methods The domestic sewage samples were collected from different places:university A, university B and a hotel. The DNA damage effect was tested in the bacteria, cells and BALB/C mouse by single cell gel electrophoresis (comet assay), the micronucleus test, Ames test. Results Comet assay indicators:all three samples, before chlorinated, at the dosages of 1.0, 5.0 mg/ml, after chlorinated, at the dosages of 0.04, 0.2, 1.0, 5.0 mg/ml for university A, 0.2, 1.0, 5.0 mg/ml for a hotel, 1.0, 5.0 mg/ml for university B, were significantly higher than those in the negative group(P

Objective:To explore antitumor mechanism of a recombinant vaccinia virus expressing human carcinomembryonic antigen(rV CEA).Methods:C57bl/6 mice were immunized three times with rV CEA.Six weeks later, the splenocytes from rV CEA immunized donors were adoptively tranfered in sublethally irradiated CEA + HePa bearing recipients, meanwhile the antiumor effect of the donor splencytes in virtro was detected. Results:Splenocytes from rV CEA immunized donors expressed strong antiumor activity in CEA positive tumorbearing recipients, whereas W VV immunized and nonimmune donor cell did not.The antiumor activity was abolished when immune T cells were depleted before splencytes transfer. Conclusion:It was demonstrated that antiumor acivity induced by rV CEA was essentially mediated by immune T cells which were activated by CEA specific immune response.Adoptive immunotherapy strategies may have implications for both the study of rV CEA and the development of potential clinical applications for tumor immunotherapy.

Journal of Applied Clinical Pediatrics 2004;0(08):-
Objective To observe the expression of resistin mRNA and protein in adipose tissues of obese rats, and to explore the correlations between resistin and obesity, insulin resistance. Methods Thirty SD rats were divided into control group(n=15) and high-fat diet group(n=15).The rats in control group recieved common forage. The obese and insulin resistance models were induced with high-fat diet in SD rats. The plasma insulin level was determined by double antibody radioinimunity analysis, and automatic biochemistry analyzer in plement was used to detect the plasma free fatty acid. After 11 weeks, glucose tolerance test was carried out to determine blood glucose levels at intervals(0,30,60,90,120 min).Resistin mRNA from fat pads tissue was extracted by RT-PCR,and then its protein was detected by Western blot. The data were analyzed statistically by SPSS 11.5 software. Results After 11 weeks, the rats′ weight in high-fat diet group increased obviously than that of control group,as well as fasting blood glucose, free fat acid homeostasis model assessment of insulin resistance(HOMAIR) and the glucose tolerance in high-fat diet group reduced greatly. The resistin mRNA and protein in white adipose tissues were significantly higher in the obese mice than those of control group(Pa

To analyze the prescription and medication regularities of traditional Chinese medicines in the treatment of melancholia in the Chinese journal full text database (CNKI), Wanfang Data knowledge service platform, VIP, Chinese biomedical literature database (CBM) in based on the traditional Chinese medicine inheritance support platform software, in order to provide reference for further mining traditional Chinese medicines for the treatment of melancholia and new drug development. The traditional Chinese medicine inheritance support platform software V2.0 was used to establish the prescription database of traditional Chinese medicines for treating melancholia. The software integrated data mining method was adopted to analyze four Qis, five flavors, meridian distribution, frequency statistics, syndrome distribution, composition regularity and new prescriptions. Totally 358 prescriptions for treating melancholia were analyzed to determine the frequency of prescription drugs, commonly used drug pairs and combinations and develop 22 new prescriptions. According to this study, prescriptions for treating depression collected in modern literature databases mainly have the effects in soothing liver and resolving melancholia, strengthening spleen and eliminating phlegm, activating and replenishing blood, regulating liver qi, tonifying spleen qi, clearing heat and purging heat, soothing the mind, nourishing yin and tonifying kidney, with neutral drug property and sweet or bitter flavor, and follow the melancholia treatment principle of "regulating qi and opening the mind, regulating qi and empathy".

The Journal of Practical Medicine 2017;33(10):1568-1572
Objective To investigate the relationship between diabetes and bone metabolism and the potential epigenetic mechanisms. Methods BMSCs were cultured for 7 and 15 days in cell culture medium with different concentrations of glucose. The mRNA and protein expression of HDACs and osteogenesis-related genes were detected by RT-PCR and Western blot assay ,respectively. Moreover ,the combination of HDAC to the promoter region of Runx2 was tested by the chromatin immunoprecipitation (ChIP) assay. Results ThemRNA expression of osteogenesis-related genes ,incuding OCN(P < 0.05)and Col1(P < 0.05),in the bone marrow of diabetic mice was significantly reduced compared with the control mice. The mRNA and protein expression of ALP ,OCN ,Runx2 and OSX was gradually reduced with the increasing concentration of glucose ,while HDAC2 mRNA and protein expression was increased. The binding activity of HDAC2 to the upstream and downstream of Runx2 promoter region in 25mM glucose-treated BMSCs was higher than the control group(P < 0.05). Conclusoins Diabetes might repress osteogenesis of BMSCs via inhibiting the activity of Runx2 through upregu-lating the expression of HDAC2.

AIM: To study the effect of genistein on activation and proliferation of T cells, and explore the molecular mechanism of genistein. METHODS: Fluorescence conjugated monoclonal antibodies and flow cytometry were used to detect the express of CD69 and CD25 by activated T cells in vitro in response to Concanavalin (ConA )and Phorbol 12,13-dibutyrate(PDB) or T cell proliferation stained by CFSE in response to PDB / Ionomycin or ConA. RESULTS: Genistein inhibited the expression of CD69 and CD25 in activated T cells in response to Con A in a concentration-dependent manner and in response to PDB in a high concentration. Genistein inhibited proliferation of T cells in both groups in a concentration-dependent manner. CONCLUSION: Genistein inhibited activation and proliferation of T cells in vitro in response to polyclonal stimulus, and it may hold potential as a new immunosuppressant.

Objective To evaluate the clinical value of bedside echocardiography in full term infants in NICU.Methods The bedside echocardiography was performed for 313 full term infants, and then they were ana-lyzed.Results The main clinical characteristics of totally 313 cases of neonates were tachypnea, heart murmur and cyanosis.Patent foramen ovale and patent ductus arteriosus were common in the no cyanotic blood shunt,no cyanotic congenital heart disease(CHD)was mainly made of atrial septum defect(31 cases),followed by ventricular septal defect (20 cases).Cyanotic CHD was made of pulmonary atresia(6 cases),complete transposition of great artery (5 cases),tetralogy of fallot(5 cases),total anomalous pulmonary venous drainage(5 cases).Tricuspid regurgitation was common in abnormal blood flow, moderate to severe pulmonary hypertension was common for full term infants. Conclusion There were many abnormal echocardiography examination results, and atypical noise was common in neonatal CHD,routine examination of bedside echocardiography for full term infants in NICU was important for early detection of CHD and may help treatment timely to improve the prognosis.
