1.Relationship between drug resistance of Candida albicans,gene expression associated with drug resistance and proteinase

Objective To study the relationship between drug resistance and expression of the gene associated with drug resistance in Candida albicans,as well as the secretory proteinase activity of these isolates.Methods The minimum inhibitory concentrations(MIC)to fouconzole and itraconazole in 99 strains of Candida albicans isolated from respiratory tract were measured by broth microdilution method,the M27-A protocol recommended by NCCLS.The gene expressions of CDR1 and MDR1 were semi-quantitatively determined by RT-RCR.The secretory proteinase activity of these isolates was assayed by plate with medium containing bovine serum albumin.Results The rates of drug resistance to fluconzole and itraconazole were 4% and 12.1% respectively.CDR1 expression in the susceptible dose-dependent(S-DD)isolates was higher than those in the susceptible isolates and drug-resistant isolates(P

Mitochondrial dysfunction has been implicated in the aetiology of sporadic Parkinson's disease but its role in the disease mechanism remains unclear.To investigate the effect of synuclein on mitochondrial dysfunction induced by rotenone.The human dopaminergic SH-SY5Y cells were used as a cell model.The cells over-expressed the wild-type ?-synuclein were treated with complex I inhibitor rotenone.The cell viability,complex I activity,Mitochondrial swelling and O2-content were tested at different time point-1w,2w,4w after rotenone treated.CCK-8 test results showed that the cell viability of overexpressed ?-synuclein(SH-SY5Y-Syn)was much lower than the control group(SH-SY5Y-Ctr).After administrating with rotenone about 1w or 2w the cell viability of SH-SY5Y-Syn became higher than that of SH-SY5Y-Ctr.On the 4th week the results were contrary to the first 2 weeks.Similar results were got when test the mitochondrial function.In the first 2 weeks after roteoone administrating,the mitochondrial function of SH-SY5Y-Syn was better than that of SH-SY5Y-Ctr.This suggest that the ?-synuclein could protect the mitochondrial against the injury induced by rotenone in the early stage-1w,2w,while this effect disappeared in the final stage-4w.
4.Therapeutic effect of splenic autologous transplantation on rat liver cancer induced by DEN

Objective To investigate the therapeutic effect of autologous transplantation of spleen on rat liver cancer induced by DEN. Methods Forty rats were randomly divided into A and B group on average. Total spleens of 40 rats were resected, which preserved in liquid nitrogen. All splenectomy rats were feeded with DEN to induce hepatocellular carcinoma. After 12 weeks ,all rats which had were successfully induced liver cancer were transplanted by autologous spleen in group A. Group B were sham operation as control. Results Survival period was significantly higher in group A than that in group B. But transplanted splenic tissue had not recovered to normal morphological structure until these rats died. Conclusion Autologous transplantation of spleen has therapeutic effect for liver cancer induced by DEN.
5.Lipotoxicity of palmitic acid on islets and protecting effect of fenofibrate

Objective To observe the effects of palmitic acid (PA) and fenofibrate (FF) on rat islets. Methods Rat islets were isolated with collagenase digestion and divided into 6 groups: control group, PA0.2 group (with 0.2 mmol/L PA), PA0.4 group(with0.4mmol/LPA),FF group (with 5?10 -6 mol/L fenofibrate), PA0.2+FF group and PA0.4 +FF group. After being cultured 24 hours with PA in the presence or absence FF, baseline insulin secretion (BIS) and glucose stimulated insulin secretion (GSIS) were examined. The mRNA levels of insulin(INS), pancreas-duodenum homeobox-1 (PDX-1), glucose transport protein 2 (GLUT-2) and PPAR? were determined by RT-PCR or real-time PCR. Results (1)In both PA0.2 and PA0.4 groups, BIS was increased and GSIS increase was impaired as compared with control group (P0.05). 6.Application of Data Mining on Selection and Specificity of Acupoints

Objective To explore the feasibility of application of data mining in the study on selection and specificity of acupoints.Methods The ancient and modern prescription data of acupuncture treating migraine and functional dyspepsia were regarded as sample data. Database techniques and natural language processing techniques were introduced to structurize and standardize acupuncture prescription data. Based on association rule data mining, the self-developed Acupuncture Data Mining System was applied to discover the rules of selection of acupoints.Results There were collected 634 and 386 prescriptions of acupuncture treating migraine and functional dyspepsia respectively. In the prescriptions treating migraine, acupoints distributed on Gallbladder meridian and Triple Energizer meridian accounted for 40.5% and 50.6% in ancient time and modern time respectively, and specific points accounted for 84.0% and 82.8% in ancient time and modern time respectively. In the prescriptions treating functional dyspepsia, acupoints distributed on Conception vessel, Bladder meridian and Stomach meridian accounted for 62.1% and 75.7% in ancient time and modern time respectively, and specific points accounted for 70.0% and 83.3% in ancient time and modern time respectively.Conclusion The result of data mining proves that the rule of selection of acupoints is "selecting acupoints along the affected meridian" and "mostly selecting specific points", and the specificity of acupoints is "where the meridian goes, where the qi accumulates".

9.A Novel Single Nucleotide Polymorphism of the Human Thyrotropin Receptor and Evaluation the Effect on Development of Graves Disease

Objective To obtain more information concerning polymorphism of the thyrotropin (TSHR) in Graves diseases(GD). Methods (1)A family of GD was studied (including 3 patients and 9 healthy family members)to examine SNPs of TSHR through direct sequencing of all 10 exons and part of introns. (2)In the current case-control study, 30 patients with familiar GD, 48 sporadic patients and 96 healthy control individuals were used to assess whether SNP of TSHR was associated with GD. Genomic DNA was extracted from peripheral leukocytes isolated from ACD-anticoagulated blood. Ten exons were amplified by PCR, using primers designed by ourselves. After purifying, the products were sequenced. Results Eight polymorphisms were found. There was a novel polymorphism in exon 8. There were no significant differences between patients and controls. Conclusions These findings suggested that the novel and other polymorphisms of the TSHR gene may not be responsible for GD. There are racial differences in the distribution of polymorphisms of TSHR gene.
10.Clinical efficacy of gabapentin or topiramate combined with venlafaxine in the treatment of chronic migraine with anxiety disorder

Objective:To investigate the clinical efficacy and safety of gabapentin or topiramate combined with venlafaxine in the treatment of chronic migraine patients with generalized anxiety disorder.Methods:From June 2018 to February 2020, 127 patients with chronic migraine complicated with generalized anxiety disorder in the Second Affiliated Hospital of Guangxi Medical University were selected. The patients were divided into gabapentin combined with venlafaxine group (observation group, 64 cases) and topiramate combined with venlafaxine group (control group, 63 cases) according to the random number table method, and all patients were treated for 6 months. The headache attack days per month, headache visual analogue scale (VAS), migraine specific quality of life questionnaire V2.1 (MSQ V2.1), headache impact measurement-6 (HIT-6) score, Pittsburgh sleep quality index (PSQI) score were recorded before treatment and 3 and 6 months after treatment.Results:In observation group, 57 cases completed 3 months of treatment, and 53 cases completed 6 months of treatment. In the control group, 56 cases completed 3 months of treatment, and 50 cases completed 6 months of treatment. The headache attack days per month, headache VAS, HIT-6 and PSQI 3 and 6 months after treatment in 2 groups were significantly lower than those before treatment, observation group: (16.31 ± 5.02) and (15.69 ± 6.31) d vs. (22.62 ± 3.27) d, (3.67 ± 1.60) and (1.91±1.05) scores vs. (5.09 ± 1.43) scores, (49.34 ± 11.01) and (47.34 ± 9.05) scores vs. (60.25 ± 11.61) scores, (10.09 ± 2.81) and (9.68 ± 2.74) scores vs. (13.50 ± 2.81) scores; control group: (14.58 ± 7.37) and (9.92 ± 5.07) d vs. (23.05 ± 5.24) d, (4.74 ± 1.15) and (3.16 ± 1.60) scores vs. (5.90 ± 2.06) scores, (42.77 ± 8.02) and (40.09 ± 9.72) scores vs. (59.37 ± 9.08) scores, (9.66 ± 2.71) and (8.62 ± 2.07) scores vs. (14.61 ± 2.79) scores, and there were statistical differences ( P<0.05). The headache VAS 3 and 6 months after treatment in observation group was significantly lower than that in control group, and there was statistical difference ( P<0.05). The functional limitations, function loss, emotional function scores and total score of MSQ V2.1 3 and 6 months after treatment in 2 groups were significantly lower than those before treatment, observation group: (17.62 ± 9.81) and (16.01 ± 5.73) scores vs. (36.96 ± 9.55) scores, (12.17 ± 5.60) and (11.09 ± 3.27) scores vs. (17.06 ± 6.08) scores, (8.42 ± 2.17) and (8.94 ± 1.90) scores vs. (11.40 ± 4.09) scores, (33.24 ± 9.61) and (28.62 ± 5.04) scores vs. (62.75 ± 14.02) scores; control group: (17.08 ± 8.73) and (16.79 ± 5.19) scores vs. (36.82 ± 9.68) scores, (9.04 ± 4.48) and (8.90 ± 3.46) scores vs. (17.26 ±6.01) scores, (6.92 ± 2.61) and (5.15 ± 1.74) scores vs. (11.30 ± 5.47) scores, (31.65 ± 9.17) and (30.66 ± 6.04) scores vs. (62.91 ± 11.18) scores, and there were statistical differences ( P<0.05). There was no significant difference in the effective rate and the incidence of adverse drug reactions 3 and 6 months after treatment between 2 groups ( P>0.05). Conclusions:Gabapentin or topiramate combined with venlafaxine can reduce the degree of headache in chronic migraine patients with generalized anxiety disorder, reduce the number of headache days per month, improve sleep and improve the quality of life. However, the adverse reactions of gabapentin still need to be paid more attention.
