1.The expression and significance of CDX2 and Ki-67 in colorectal tumor

ObjectiveTo investigate the expression and significance of CDX2,Ki-67 in adenoma and colorectal adenocarcinoma,and the correlation between CDX2 and Ki-67.MethodsThe expression of CDX2 and Ki-67 was evaluated in the colorectal adenocarcinoma,adenoma and normal colorectal mucosa as control subjects by immunohistochemistry.ResultsThe magnitude of CDX2 were negatively correlated with the level of dysplasia in adenoma(rs=-0.38,P =0.02),and negatively correlated with differentiation,lymph node metastasis,diversion and Dukes stage of cancer in colorectal adenocarcinoma(rs=-0.49,rs=-0.37,rs =- 0.38,rs =-0.37,P =0.01).The expression and relativity of Ki-67 in the samples were contrary to the CDX2.The levels of CDX2 and Ki-67 expression had significant difference between the adenoma and adenocarcinoma,and showed negatively correlation in this two type tissues (rs=-0.69,P =0.00; rs =-0.40,P =0.00).ConclusionsCDX2 may be used as an effective marker to evaluate malignancy degree and prognosis of large bowel neoplasm patient.The levels of Ki-67 expression in colorectal adenocarcinoma are correlated with malignancy degree and clinical stage.The expression of CDX2 is negatively correlated with Ki-67.Combined detection of CDX2 and Ki-67 may be helpful for the judgment of colorectal tumor biological characters.
3.Microtox Technology for the Evaluation of Comprehensive Toxicity of Shen Mai Injection

In this research,we chiefly explored a new fast test method--microtox technology to evaluate the comprehensive toxicity of Shen Mai injection.We investigated characteristic parameters of vibrio fischeri (CS234) under different conditions in the pursuit of the optimum test parameters of the reliable method;and then locked the best parameters for Shen Mai injection assay with the products from different manufacturers by microtox fast test.As a result,the optimum reaction condition included 0.9 mL recovery liquid in each freeze-dried vial and 50 μL bacteria suspension of each sample within 2 mL solution in aggregate with 10-mins detection after adding bacteria suspension which pH value ranging from 5 to 10 and luminous intensity from 0.8 to 1.2 million in 10 mins.The relative standard deviation values of replication experiment and precision test were all below 15%.Under this optimum detection condition,it was found that the EC50 values were 35.60%,92.34% and 146.57%,differing among the samples from three representative drug manufacturers,respectively (P < 0.01).In conclusion,the concentration-effect relationship of Shen Mai injection existed in the toxicity assessment of vibrio fischeri with various ECs0 values from the three manufacturers.These results suggested that biological detection standards of Shen Mai injection presented great growth potential.Microtox-based fast test in the evaluation of comprehensive toxicity of ShenMai injection showed favorable application prospects over controlling the quality of different sources of products.
4.Neoglycoprotein preparation of YCP, a polysaccharide from Phoma herbarum YS4108 and immunogenicity evaluation in mice

Aim:To characterize the immune response to YCP, a polysaccharide (PS) isolated from marine filamentous fungus Phoma herbarum YS4108. Methods:YCP was coupled to bovine serum albumin (BSA) to construct three neoglycoproteins which are different in their degrees of substitution (DSs; ≈ 3, ≈ 6, or ≈ 10 mol of BSA carrier/mol of YCP hapten, respectively), and their immunogenicities were evaluated in mice following subcutaneous immunization,respectively. IgG subclasses against the PS and the carrier protein were measured in addition to the total IgG and IgM antibodies for YCP induced by the neoglycoproteins. Results: While unconjugated PS was weakly immunogenic, theneoglycoprotein induced vigorous primary IgM and booster IgG responses to PS and the carrier protein. Interestingly,the IgG subclass distribution was different between PS and the carrier protein; for PS, the IgG response was predominant of IgG2a subclass with approximately low levels of IgG2b and IgG1, while the response to the carrier protein was mainly of the IgG1 subclass with relatively low levels of IgG2a and IgG2b, and the lowest of IgG3. Conclusion:YCP has the potential to elicit preferentially IgG2a as the dominant isotype antibody in mice.
5.Effect of Wuzhi capsules on whole blood concentration of Tacrolimus in renal transplantation recipients

BACKGROUND: It has reported that the combination of Tacrolimus and Wuzhi capsules can increase the peak concentration and area under curve of Tacrolimus recipients, however, the effects of this combination on recipients is poorly understood. OBJECTIVE: To investigate the effects of Wuzhicapsules on the blood concentration of Tacrolimus recipients. METHODS: A total of 38 renal transplantation recipients receiving triple therapy regime (Tacrolimus+mycophenolate +Prednisolone) were involved in the study and were divided into the experimental group (n=21) and control group (n=17). Recipients in the experimental group were taking Wuzhi capsules simultaneously, and those in the control group were not taking Wuzhicapsules. The Tacrolimus dosage to therapeutic concentration window and renal function were compared between the two groups at months 1,3 and 6 after transplantation. The incidence rates of acute rejection and diabetes mellitus, and renal and liver functions were observed at 6 months after transplantation. RESULTS AND CONCLUSION: The Tacrolimus dosage to therapeutic concentration window of the experimental group was significantly smaller than that of the control group (P < 0.05). However, incidence rate of acute rejection, diabetes mellitus, and renal and liver functions had no dramatically difference in 2 groups (P > 0.05). Results suggest that Wuzhi capsules may improve the immunosuppressive efficacy of Tacrolimus in renal recipients, therefore, can decrease the toxic effect of Tacrolimus with reduction dosage.
6.Significance of CD20-positive lymphocytes infiltrating in renal allograft biopsies with chronic allograft nephropathy

ObjectiveTo investigate the action mechanism of CD20 lymphocyte infiltration in the renal allograft biopsy with chronic allograft nephropathy (CAN).MethodsCAN cases confirmed by renal biopsy within 2 years after renal transplantation served as study subjects. By using immunohistochemistry,the deposition of C4d and the CD20-positive lymphocytes infiltration in the renal grafts were examined.The clinical follow-up data were analyzed.ResultsForty-four cases of CAN were enrolled in the study, including 13 cases (29.5% ) of CD20-positive lymphocytes infiltration,and 31cases (70.5% )of CD20-negative lymphocytes infiltration. CD20-positive lymphocytes in biopsy showed nodular and scattered lymphocytes infiltration.There were 5 (26.3%)cases of CAN Ⅰ,4 cases (25.0%) of CAN Ⅱ,and 4 (44.4%) of CAN Ⅲ in CD20-positive group.There was no statistically significant difference between the only CAN group and CAN with AR group in CD20-positive rate.Immunohistochemical staining showed there were 12 cases (27.3%) with C4d linear deposition in peritubular capillary endothelial cells (PTC).C4d positive rate had no significant difference among the CAN classifications. There was no significant relationship between C4d deposition and CD20-positive lymphocytic infiltration.The average serum creatinine in CD20-negtive group and CD20-posigtive group was 140.8 ± 22.0 and 183.5 ± 25.5μmol/L before biopsy,and 165.6 ± 37.6 and 242.2 ± 59.1 μmol/L one year after biopsy.The average serum creatinine level in CD20-positive group was higher than in CD20-negtive group before and after biopsy.ConclusionProgressive chronic humoral immunity is high risk in the process of CAN. The CD20-positive lymphocyte infiltration has no relevance with CAN grade and C4d deposition in PTC,but is associated with circulating antibody PRA and allograft long-term outcome. Pathogenetic mechanism may not contribute to chronic humoral immune,but B cells presenting donor antigens,are recognized and activated by T cells as antigen-presenting cells.
7.Expression of TSLC1 in human gastric cancer and its clinical significance

Purpose To explore the relationship between tumor suppressor in lung cancer 1 (TSLC1) protein expression and the carci-nogenesis and progression of human gastric carcinoma. Methods Expression of TSLC1 protein in 20 normal gastric mucosa, 30 intra-epithelial neoplasias ( IN) and 50 gastric cancers was examined by immunohistochemistry. Results The expression of TSLC1 in gas-tric cancer was 14. 00% which was lower than that in IN (46. 67%) and normal gastric mucosa (95. 00%, P<0. 05). TSLC1 ex-pression in high-grade IN was lower than that in low-grade IN and normal gastric mucosa (P<0. 05). TSLC1 expression in high-grade IN and gastric cancer was of no significant difference ( P>0. 05 ) . Expression of TSLC1 was significantly associated with lymph node metastasis and TNM in gastric cancer (P<0. 05). Conclusion The expression of TSLC1 is closely related to carcinogenesis, lymph node metastasis and clinical stage in gastric cancer, which suggest that TSLC1 may be a new target for the prevention and treatment of gastric cancer.

9.Practice of human dermal lymphatic endothelial cell isolation and cryopreserving technique

0.05). Conclusion Enough high-purified LECs can be isolated by collagenase digestion procedure followed by immunomagnetic beads sorting. Post-thawed endothelial cells are proved to have high vitality and growth potential in vitro without significant morphological changes. Cryopreserved LECs may serve as a cell choice for research of lymphangiogenesis and lymphatic patterning.
10.Effects of Munc13-1 and Munc18-1 on dopamine secretion dysfunction in manganese-exposed SH-SY5Y cells

Background Neurotransmitter secretion disorder induced by chronic manganese poisoning has always been one of the important causes of body injury, but the mechanism of neurotransmitter secretion disorder caused by manganese is not clear at present. Objective To investigate the effects of presynaptic membrane intracellular protein 13-1 (Munc13-1) and synapse fusion protein binding protein 18-1 (Munc18-1) on dopamine secretion dysfunction induced by manganese chloride (MnCl2) in human neuroblastoma (SH-SY5Y) cells. Methods A SH-SY5Y cell model induced by MnCl2 was established. Cell viability was measured by MTT assay. Four experimental groups were set up: control group and low-, medium-, and high-dose manganese groups (0, 100, 200, and 400 μmol·L−1 MnCl2). They were treated with corresponding doses of MnCl2 for 24 h. The secretion of dopamine was measured by enzyme-linked immunosorbent assay. The mRNA expression of Syntaxin-1 was detected by real-time quantitaive PCR. Total cell proteins were extracted, and the protein expression levels of Munc13-1, Munc18-1, and Syntaxin-1 were detected by Western blotting. The correlations of MnCl2 exposure and dopamine secretion with the protein expressions of Munc13-1 and Munc18-1 were also analyzed by Pearson correlation. Results Compared with the control group, the cell viability rate decreased gradually with the increase of manganese exposure concentration, and the difference between the medium- and the high-dose manganese groups was statistically significant (P<0.05). The concentration of dopamine in cell culture medium of all manganese exposure groups decreased with the increase of manganese concentration, and compared with the control group and the low-dose manganese group, the medium- and the high-dose manganese groups were statistically significant (P<0.05). The expression of Syntaxin-1 at mRNA or protein level did not change significantly among groups (P>0.05). Compared with the control group, the protein expression of Munc13-1 decreased and that of Munc18-1 increased with the increase of manganese concentration (P<0.05). Compared with the low-dose manganese group, the changes of Munc13-1 protein in the high-dose manganese group and Munc18-1 protein in the medium- and high-dose manganese groups had statistical significance (P<0.05). Compared with the medium-dose manganese group, the protein changes of Munc18-1 in the high-dose manganese group were statistically significant (P<0.05). The correlation analysis showed that MnCl2 dose was negatively correlated with Munc13-1 protein expression (r=−0.898, P<0.05), and positively correlated with Munc18-1 protein expression (r=0.678, P<0.05). Dopamine secretion was positively correlated with Munc13-1 protein expression (r=0.932, P<0.05), and negatively correlated with Munc18-1 protein expression (r=−0.817, P<0.05). Conclusion The inhibition of dopamine secretion in SH-SY5Y cells induced by manganese exposure is related to up-regulation of Munc18-1 and down-regulation of Munc13-1 expression levels, which may be one of the reasons for nerve injury caused by manganese.
