2.A New Subset of CD4~+T cells: Th17 and Its Biological Effects
Progress in Biochemistry and Biophysics 2006;0(04):-
The T help cell 1(Th1) and Th2 cell classification have provided the framework for understanding CD4+ T cell biology and the interplay between innate and adaptive immunity for almost two decades. Recent studies have defined a previously unknown subset of the CD4+ T cell effectors, the Th17 lineage. The uncover on the differentiation of IL-17-producing effector T cells from naive T cell precursors provides insights into mechanisms by which signals from cells of the innate immune system guide alternative pathways of Th1, Th2 or Th17 development. Th17 lineage has an important role in autoimmune and inflammatory diseases. This promises to change our understanding on the immune regulation, immune pathogenesis and host defense. The identification, differentiation and immune function of the new subsets of Th cells, Th17 cells will be reviewed.

The treatment group was intervened by acupuncture at lumbar Jiaji (EX-B 2) points plus botulinum toxin injection, while the control group was by conventional acupuncture plus botulinum toxin injection. After 3 treatment courses, the seated function zone (zone B) of Gross Motor Function Measure (GMFM) was used for evaluation, and the clinical efficacies were compared.Result Respectively after 1, 2, and 3 treatment courses, the total effective rates were 46.7%, 73.3% and 83.3% in the treatment group, versus 33.3%, 46.7% and 60.0%. There were significant differences in comparing the total effective rate between the two groups after 2 and 3 courses (P<0.05). After the first treatment course, the seated function score of GMFM was significantly changed in both groups (P<0.01). After the second course, the seated function score was significantly different from that after the first course in both groups (P<0.01). After the third course, the seated function score was significantly different from that after the second course in both groups (P<0.01). There was a significant between-group difference in comparing the changes of the seated function score during the second treatment course (P<0.01). There was a significant between-group difference in comparing the changes of the seated function score during the second treatment course (P<0.05). Conclusion Acupuncture at lumbar Jiaji (EX-B 2) points plus botulinum toxin injection can produce a more significant efficacy in improving the seated function of patients with spastic cerebral palsy.

Objective To investigate the influence of high-level of glucose on the expression of ghrelin in human umbilical vein endothelial cells (HUVECs). Methods After 12 h glucose free culturing, the effects of different concentrations of glucose and different incubation times on expressions of ghrelin were observed in HUVECs. ( 1) The cells were treated with 0, 5, 10, 15, 20 mmol/L glucose for 2 h, then ghrelin mRNA expression levels were detected by RT-PCR, and the protein levels of ghrelin were detected by ELISA. (2) The cells were treated with 10 mmol/L glucose for 0, 0.5, 1, 2, 6, 12 hours, ghrelin mRNA and protein levels were detected respectively. Results (1) The expression levels of ghrelin mRNA and protein decreased along with increased glucose concentrations, which showed no obvious changes when the glucose was above 15 mmol/L. (2) The expression levels of ghrelin mRNA and protein decreased with the prolonged incubation time. But more than 6 h culturing time showed no further effect on reducing the expression level of ghrelin. Conclusion High levels of glucose can inhibit the expression level of ghrelin, which may be one of the mechanisms of atherosclerosis.
