1.The cloning of human interleukin-10 gene and its expression in eukaryotic cells

Objective: To clone human interleukin 10(hIL-10) gene full-length cDNA sequence and to construct its eukaryotic expression vector in Chinese hamster ovary(CHO) cells. Methods: cDNA fragment encoding hIL-10 gene was amplified from human peripheral normal white blood cells by RT-PCR and confirmed by DNA sequencing.The ds-DNA was inserted into pcDNA3 vector.This recombinant expressing vector was transfected into CHO cell.The IL-10 molecules expressed were detected by ELISA and the role of inhibition of IL-10 was determined with MTT. Results: The cDNA encoding hIL-10 was cloned by RT-PCR and inserted into T-easy vector and sequence analysis verified that the cloned fragment was hIL-10 cDNA.The IL-10 was expressed in the CHO and it blocked the lymphocyte transformation.Conclusion: The eukaryotic expression plasmid was constructed successfully,which will contribute to further studies on the role of hIL-10 in autoimmunity,transplantation immunity and inflammatory disease.
2.Evaluation of the value of corticospinal tract injury in patients with cerebral infarction with magnetic resonance diffusion tensor and diffusion tensor teactography imaging

Objective To evaluate the relationship between the degree of corticospinal tract (CST) injury and motor function prognosis in patients with cerebral infarction using magnetic resonance diffusion tensor imaging (DTI)and diffusion tensor teactography (DTT)imaging. Methods Sixty-eight patients with cerebral infarction were selected prospectively. They underwent conventional MRI,DWI,and DTI examinations,and the DTT technique was used to perform CST 3D reconstruction. The white matter fractional anisotropy (FA)of the infarct zones and contralateral corresponding regions was measured within 7 days and at 3 months after onset. The Fugl-Meyer scale was used to perform motor function defect score. The CST involvement degrees were divided into four grades. The patients were divided into Group 1 (96-99,roughly return to normal;n=16),Group 2 (51-95,partial recovery;n=34),and Group 3 (<50,poor recovery;n=18)according to the Fugl-Meyer scale scores after treatment. The analysis of variance,Pearson,and Spearman correlation analysis were used to analyze the relationship between FA values,CST involvement degrees and motor function recovery. Results There were significant differences in the amplitude of variation of FA within 7 d (0. 05 ± 0. 06,0. 08 ± 0. 07,and 0. 18 ± 0. 02)and at 3 months after onset (0. 11 ± 0. 02,0. 21 ± 0. 06,and 0. 39 ± 0. 03)(within 7 d F=32. 821,at 3 months F=192. 372,all P<0. 05). DTT showed that the lesion sides of CST were presented as compression,deformation,displacement,and interruption. The CST grades were negatively correlated with the neurological functional rehabilitation of the patients within 7 d after onset (r= -0. 682,P <0. 01). The CST grades were negatively correlated with the neurological functional rehabilitation of the patients at 3 months (r= -0. 728,P<0. 01). Conclusion The decreased FA values in infarct areas and the degrees of CST involvement were associated with the motor function prognosis. MR DTT can directly display the degrees of CST involvement of cerebral infarction. It can provide an important basis for early identifying the prognosis of cerebral infarction.
3.Clinical characteristics of 1 006 Chinese patients with lupus erythematosus: results from the Lupus Erythematosus Multicenter Case-control Study in Chinese populations (LEMCSC)

Objective To explore clinical characteristics of Chinese patients with lupus erythematosus (LE).Methods Data were obtained from the Lupus Erythematosus Multicenter Case-control Study in Chinese populations (LEMCSC).A unified standard was used to recruit patients and collect clinical information.The EpiData 3.1 and SPSS 18 softwares were utilized to input and analyze data respectively.Results One thousand and six patients (87.6% female) with lupus erythematosus (LE) were included in this analysis,of whom,887 (89.9% female) had systemic LE (SLE),and 119 (70.6% female) had isolated cutaneous LE (CLE).The most common involved system in SLE patients was skin (72.7%),followed by joints (69.2%),hematological system (60.8%),kidney (48.5%),serosa (18.2%),and nervous system (5.7%).The appearance of LE-specific skin manifestations was associated with an increased risk of arthritis (odds ratio [OR] =1.612,95% confidence interval [CI]:1.181-2.200),but with a decreased risk of nephritis (OR =0.218,95% CI:0.157-0.303) and serositis (OR =0.311,95% CI:0.218-0.443).The presence of acute CLE (ACLE) lesions was a risk factor for systemic involvement (OR =4.931,95% CI:3.232-7.524),while that of chronic CLE (CCLE) lesions was a protective factor for systemic involvement (OR =0.355,95% CI:0.234-0.541).The appearance of LE-nonspecific skin manifestations was closely correlated with the involvement of internal organs in patients with LE.Conclusion This study revealed main characteristics of LE patients in China and the relationship between LE-related skin lesions and internal organ involvement.

6.Construction of Gadd45a expression plasmid and its expression in human T cells

Objective To construct Gadd45a expression plasmid and induce its expression in human T cells.Methods Gadd45a was amplified by reverse transcription PCR from human embryonic stem cells,and cloned into the pcDNA3.1 vector.The recombinant plasmid or blank plasmid was transfected into Jurkat cells or normal human CD4+T cells using electroporation,and the expression of Gadd45a was detected by quantitative RT-PCR and Western blot.Results Human Gadd45a expression plasmid was constructed successfully.Gadd45a was overexpressed both in Jurkat cells and normal human CD4+T cells after these cells were transfected with pcDNA3.1-Gadd45a.Conclusion The construction of Gadd45a expression plasmid and induction of Gadd45a overexpression in human T cells lay the foundation for further research on the role of Gadd45a in the epigenetic mechanism.
7.Effects of ultraviolet B (UVB) on the expression of Gadd45a gene and DNA methylation level in Jurkat cells

Objective To explore the effects of UVB on the expression of Gadd45a gene and DNA methylation levels in Jurkat cells. Methods Jurkat cells were irradiated with UVB of 1.0 J/cm2 and 1.5 J/cm2 respectively, and collected at 6, 12, 24 and 48 hours after the irradiation. Real-time RT-PCR was used to detect the mRNA expression of Gadd45a gene and methylation-sensitive genes CD11a and CD70. Global methylation level was also measured by MethylAmp global DNA methylation quantification kit. Results After irradiation with UVB at 1.0 J/cm2, the mRNA level of Gadd45a increased but global methylation level decreased at 6, 12, 24 and 48 hours, and significant changes were observed at 6 and 12 hours for the level of both Gadd45a mRNA expression and global methylation (P < 0.01 or 0.05). Elevated mRNA expressions of CD11 a and CD70 were also noted in Jurkat cells after irradiation with UVB of 1.0 J/cm2, and significant elevation was observed at 12 hours (both P < 0.05 ). After irradiation with UVB of 1.5 J/cm2, there was a statistical increase in the mRNA expressions of Gadd45a, CD11 a, CD70, together with a statistical decrease in global methylation level in Jurkat cells, at 6, 12, 24 and 48 hours (P < 0.01 or 0.05). The mRNA expression of Gadd45a negatively correlated with the global level of DNA methylation in Jurkat cells (r = -0.395, P < 0.05). Conclusion UVB irradiation can upregulate the expression of Gadd45a, but downregulate the global DNA methylation level in Jurkat cells.
8.DNA methylation status of miR-126 and its host gene EGFL7 in CD4+T cells from patients with systemic lupus erythematosus

Objective:To explore the mechanisms by which DNA methylation regulates miR-126 and its host gene EGFL7 in CD4+T cells from patients with systemic lupus erythematosus (SLE).
Methods:We analyzed the expression and the DNA methylation status within promoter region of EGFL7 and miR-126 by real-time qPCR and bisulifte genomic sequencing analysis.
Results:miR-126 and EGFL7 mRNA expression was upregulated in CD4+T cells from SLE compared with that from healthy controls (P<0.01). EGFL7 mRNA level was positively correlated with miR-126 expression in CD4+T cells from SLE (r=0.538, P=0.015). The average methylation level of EGFL7 promoter in CD4+T cells from SLE was lower than that from healthy controls (P<0.05).
Conclusion:hTe upregulation of miR-126 and its host gene EGFL7 expression in CD4+T cells from SLE is associated with the hypomethylation of the EGFL7 promoter.
9.Treatment of radial neck fracture in children with elastic stable intramedullary nails.

OBJECTIVETo investigate effects of elastic stable intramedullary nails for the treatment of radial neck fracture in children.
METHODSFrom July 2006 to December 2011, 25 children with radical neck fractures, which included 16 males and 9 females aged from 7 to 15 years old (means 10.7), were treated with elastic stable intramedullary nails. According to Judet classification, 6 cases were type II, 17 cases were type III and 2 cases were type IV (including 1 case with type IVa and 1 case with type IVb). The fracture healing, pain, deformity and range of motion of elbow were recorded.
RESULTSAll patients were followed up for 6 to 24 months with an average of 14 months. Twenty-five patients were obtained bone healing. According to Tibone and Stoltz evaluation standard, 18 cases got excellet results, 4 cases in good and 3 cases in moderate.
CONCLUSIONElastic stable intramedullary nails for the treatment of radial neck fracture in children has advantages of simple operation,less trauma and good results.
