Objective To study the immunoregulation effect of three kinds of polysaccharides extracted from different tissues of Octopus dollfusi on immunosuppression mice. Methods Two kinds of immunosuppression mice models were built by ip,cyclophosphamide(CY) and sc.hydrocortisone(HC),respectively.The spleen and thymus were weighed and the index of the immune organs was calculated. The white blood cells were counted by automated hematologic analyzer. Results In certain concentrations(20~80 mg?kg-1),the three kinds of polysaccharides could antagonize the atrophy of spleen and thymus caused by CY and HC,and increase the weight of spleen and thymus; The polysaccharides could protect against leukocytopenia induced by CY and HC,and especially in low and middle dose. Conclusion The results suggested that three kinds of polysaccharides extracted from Octopus dollfusi have potent enhancement effect on non-specific immunity for the immunosuppression mice.

Objective To search for an effective acupuncture-moxibustion therapy for high fever in common cold. Methods According to the randomized and multi-central trial principle, 276 cases enrolled were randomly assigned to observation group of 138 cases and control group of 138 cases. The observation group were treated by electroacupuncture at Dazhui and the control group were treated by Antondinie injection coordinating pathogeny therapy to observe the prompt effects in 24 hours. Results After treatment, the temperature of every time point of the observation group was lower than that of the control group (P 0.05). Conclusion The method of electroacupuncturing at Dazhui has a definite therapeutic effect on high fever in common cold.

ObjectiveTo analyze the changes of plasma concentration of calcitonin gene-related peptide (CGRP) and substance P (SP) after electrical stimulation on dura mater adjacent to sinus sagittalis superior (SSS) in rats.Methods13 rats were randomly divided into control group(n=6) and stimulation group(n=7) in which the dura matter adjacent to SSS was stimulated electrically with square wave of 20 Hz, 4 mA and 250 μs. The plasma concentrations of CGRP and SP in both groups were compared by means of radioimmunity method.ResultsThere was significant difference in the plasma concentration of CGRP between control and stimulation groups (50.60±7.16 pg/ml vs 42.20±4.82 pg/ml, P=0.029). However, there was no significant difference in the plasma concentration of SP between two groups (308.59±43.07 pg/ml vs 264.60±60.28 pg/ml, P=0.165).ConclusionPlasma concentration of CGRP decreased after electrical stimulation on dura mater in rats but the concentrations of SP unchanged.

Objective To investigate whether valproic acid (VPA) affect spatial learning memory and senile plaques in the APP/PS1 double transgenic AD mouse model of different gender. MethodsTwenty 3-month old APP/PS1 double transgenic AD mice, male and female mouse evenly, were randomly divided into VPA-treated and saline-treated groups ( 10 for each group). 30 mg· kg-1 · d-1 of VPA and the same amount of saline were peritoneally injected into mice for 4 weeks. Morris water maze was conducted to check the effect of VPA on the capability of spatial learning and memory of AD mouse model. Immunohistochemical staining was used to examine the effect of VPA on the morphological changes in the brains of mice. ResultsVisible platform test showed that VPA-treated and saline-treated mice had similar escape latency (P>0.05) and path length (P>0.05) ,the swimming speed between male and female mice had no difference (P>0.05). Hidden platform test showed that VPA treated mice had a significantly shorter latency (P<0.01) and path length (P<0.01) to reach the platform compared with saline-treated mice. Meanwhile, both in VPA-treated and control groups, the male mice had a shorter correlation escape latency and path-length than female mice had(P<0.05 ). Immuohistochemical staining showed that the number (11.23±3.78) of senile plaques (SP) in the cerebral cortex and hippocampus of VPA-treated male mice were notably decreased than that(28.17 ±3.46) in the control group ( t= 14.67, P<0.01 ),furthermore, the number of SP in the cerebral cortex and hippocampus of VPA-treated male mice was significantly reduced,as compared with which (20.36 ±4.21)in the VPA-treated female mice(P<0.05). ConclusionVPA can significantly lower formation of SP, and remarkably improve the capability of spatial learning and memory of APP/PS1 transgenic mice, which have gender difference.

OBJECTIVE:To prepare captopril hydrophilic gel sustained-release tablet and study the influencing factors on its release METHODS:With HPMC as the matrix, the tablets were prepared by direct compression method and influencing factors on release were studied RESULTS & CONCLUSION:The in vitro release of prepared tablets conformed to Higuchi equation The HPMC matrix tablet could release in a sustained way when the proportion of HPMC was at least 15% in weight and the best proportion was 60%;The dissolution of Methocel K was slower than 60RT or 75RT;Taking lactose as the filler was better than starch or CaSO4;When the proportion of lactose increased, the dissolution sharply decreased; The tablet dissolved more rapidly with paddle method than with rotating basket methos, compression force and pH of dissolution medium affected the release very little

Acta Pharmaceutica Sinica 2013;48(5):621-34
Based on the character of the molecular structure, the prodrugs of phosphates and phosphonates were divided into two categories. The first is the drug which contained the phosphate group, introducing protected groups to increase lipophilicity and improve bioavailability. The other one is the drug which had no phosphate group, introducing the phosphate group into molecules to enhance the solubility, regulate the distribution coefficient and enhance the drug-like property. This review focuses on the application of phosphates and phosphonates in drug research and development based on improvement of physico-chemical property, drug safety and the pharmacokinetics.
