2.Response of astrocytes and neurons in rat medullary visceral zone to the spared nerve injury

Objective To investigate the response of astrocytes and neurons in the medullary visceral zone (MVZ) to neuropathic pain induced by the spared nerve injury (SNI, the tibial nerve and the common peroneal nerve were sectioned, while the sural nerve was intacted). Methods The SNI operation and sham operation (only incised the skin of thigh, but the tibial nerve and the common peroneal nerve did not cut) were performed in adult male Sprague-Dawley rats. The paw withdrawal mechanical threshold (PWMT) was measured at 10,20,30 days after operation. By using single or double immunofluorescent staining method,we investigated and measured that the mean fluorescent intensity (MFI) of anti-glial fibrillary acidic protein (GFAP, a marker of astrocytes) signal in the astrocytes, the mean member of Fos/ GFAP double labeled astrocytes and Fos / tyrosine hydorxylase (TH) double labeled neurons in MVZ after operation. Results As compared with control or sham groups, SNI induced that the PWMT became significant sensitivity and peaked at 20 days after SNI. The activated astrocytes revealed an activated morphology, the MFI of anti-GFAP signal markedly strengthened, the mean number of Fos/GFAP double labeled astrocytes and Fos/TH double labeled neurons in the nucleus of the tract solitarius (NTS) and the ventral lateral medulla (VLM) increased significantly and peaked at 20 days after SNI. Conclusion The neurons and astrocytes in MVZ were sensitively activated by SNI.

4.Clinical and Pathological Analysis on 286 Cases of Seborrheic Keratosis

Objective To explore the clinical and histopathological characteristics and the diagnostic status of seborrheic keratosis (SK) Methods The clinical and pathological data of 286 cases were analyzed retrospectively.Results Among the 286 patients,the elderly patients accounted for the majority and more males were diagnosed with SK than females.The lesions were found among 46.2% of patients in the exposure sites,such as face and neck.The most frequent types were hyperkeratosis and acanthosis.The clinical diagnosis consisted with pathologic diagnosis in 152 cases.The clinical misdiagnosis rate was 36.7%.Conclusion SK is related to gender,age,and sunlight.Histopathological examination is important in the diagnosis and differential diagnosis of SK.
5.Determination of the Contents of the Related Substances in Metoclopramide by HPLC

OBJECTIVE: To establish an HPLC method for the determination of the contents of the related substances in metoclopramide.METHODS: The sample was separated on C18 column with the mobile phase consisted of acetonitrile-0.02 mol?L-1 phosphonic acid solution(19∶81) at a flow rate of 1.0 mL?min-1.The detection wavelength was set at 275 nm;the column temperature was kept under room temperature and the injected volume was 20 ?L.The contents of the related substances in metoclopramide were computed by self-control method of main constituent.RESULTS: Under the above described chromatographic conditions,metoclopramide was completely separated from its impurities.A good linearity between impurities' peak area and metoclopramide(contrast solution) concentration was achieved when the concentrations of impurities were over the range of 0.05%~4.0%.The lowest detectable limit of metoclopramide was 0.3 ng,and the contents of the related substances were all less than 0.26%.CONCLUSION: The method is convenient,accurate,sensitive and,specific,and it can be used for the determination of the related substances in metoclopramide.
6.Effect of combined electrical stimulation and nursing interventions on female stress urinary incontinence

Objective To investigate the effectiveness of the combined electrical stimulation and nursing interventions for female stress urinary incontinence.Methods The study is qusi-experimental design.48 patients with stress urinary incontinence were allocated to the intervention group and the control group with 24 patients in each group.The control group was given electrical stimulation,the intervention group was given 12-week electrical stimulation and comprehensive nursing interventions.The outcome indicators were 1-hour pad test urine loss,pelvic floor muscle (PFM) strength,the grade of subjective urinary incontinence,quality of life (I-QOL).Results Compared with the control group,no significant subjective urinary incontinence score was seen,but pelvic floor muscle (PFM ) strength and the score of the QOL evidently improved and 1-hour pad test urine loss decreased in the intervention group.Conclusions Combined electrical stimulation and nursing interventions for female stress urinary incontinence is effective treatment.
8.Expression and clinical significance of miR-34b in children with acute leukemia

Objective To research the expression,methylated regulation and clinical significances of miR-34b in chidren with acute leukemia(AL).Methods The methylation status of miR-34b promoter CpG islands were detected with methylation-specific polymerase chain reaction (MSP) in patients with AL.Then the expression of miR-34b was compared,which was detected by Taqman real-time fluorescence quantitative polymerase chain reaction (qRT-PCR),between the AL patients and normal group,in order to analyze their relationship with the clinical indicators.Resuits In 8 leukemia cell lines (U937,HL-60,MV4-11,M2R,K562,Raji,CCRF,DAMI) showed methylation,the positive rate of the methyl was 100%.Thirty-one acute lymphocytic leukemia(ALL) pediatric patients were newly diagnosed,and 24 cases showed methylation,the methylation positive rate 77.42% (24/31).Nineteen acute myeloid leukemia(AML) patients were newly diagnosed,and 8 cases showed methylation,the methylation positive rate was 42.11% (8/19 cases).There was no methylation in the 23 cases of normal children.The relative expression levels of miR-34b in the normal group,the group of leukemia cell lines,the group of ALL pediatric patients with newly diagnosed,the AML group and the group with mixed lineage leukemia gene rearrangement MLL+ were 5.22 ± 1.15,0.03 ± 0.03,1.65 ± 0.69,0.18 ± 0.06,0.64 ± 0.34,respectively.The findings indicated that there were significant differences in the relative expression levels of miR-34b between the normal group and the group of leukemia cell lines,the ALL group,the AML group,and the MLL+ group.The relative expression and methylated level of miR-34b had no statistically difference in gender,age at diagnosis,WBC count,chromosome,fusion gene,MLL gene rearrangement,and the minimal residual disease(LDH) levels in newly diagnosed AML patients(all P > 0.05).And the relative expression and methylated level of miR-34b had no statistically difference in gender,age at diagnosis,WBC count,immunophenotype,chromosome,fusion gene,MLL gene rearrangement,TEL/AML1 gene,risk stratification,the minimal residual disease (MRD) in thirty-three days and the LDH levels in newly diagnosed ALL patients(all P > 0.05).But as for the response to prednisone experiment,there was a significant difference between the sensitive group and the non sensitive group(P < 0.05).Conclusions The expression level of miR-34b in AL was significantly lower and it was regulated by methylation mechanism,which implies that miR-34b may play a role of a tumor suppressor gene in the pathogenesis of leukemia.MiR-34b may affect the early treatment response of ALL patients,and it may be an indicator of risk stratification and poor prognosis in pediatric ALL.
9.HLA-DRB1 allele polymorphosm associated with susceptibility to leukemia in Han nationality of Gansu.

The study was aimed to explore the possible association between HLA-DRB1 allele polymorphism and the susceptibility to leukemia in Gansu Chinese Han nationality and to find the genes susceptible to leukemia. HLA-DRB1 genes in 74 patients with leukemia from northwestern China and 82 healthy Chinese controls were determined by polymerase chain reaction and sequence-specific oligonucleotide probe hybridizations (PCR/SSO) DNA analysis. The results showed that as compared with control, the allele frequencies of HLA-DRB1*03 (chi(2) = 8.125, P = 0.004), HLA-DRB1*07 (chi(2) = 13.526, P = 0.000), HLA-DRB1*08 (chi(2) = 18.855, P = 0.000), HLA-DRB1*13 (chi(2) = 7.039, P = 0.008) significantly increased in AML group. The allele frequencies of HLA-DRB1*07 (chi(2) = 5.689, P = 0.017), HLA-DRB1*11 (chi(2) = 7.73, P = 0.005), HLA-DRB1*12 (chi(2) = 4.234, P = 0.040), HLA-DRB1*13 (chi(2) = 38.333, P = 0.000) significantly increased in CML group. The allele frequency of HLA-DRB1*01 (chi(2) = 5.294, P = 0.021) significantly increased in ALL group. It is concluded that the susceptibility to AML in Gansu Han nationality is positively related to HLA-DRB1*03. 1*07.1*08.1*13. CML positively correlates with HLA-DRB1*07. 1*11.1*12.1*13, and ALL may be positively in relation with HLA-DRB1*01. Allele polymorphism is associated with the leukemia occurrence.

10.Significance of cerebrospinal fluid lactate level in diagnosing neonatal bacterial meningitis

Objective To study the significance of the cerebrospinal fluid(CSF)lactate level in diagnosing neonatal bacterial meningitis(BM). Methods The CSF samples were collected from neonates admitted to Neonatal Ward of Children's Hospital of Fudan University between January 2014 and March 2015. CSF lactate and glucose con-centrations were measured with blood - gas analyzer. CSF and serum glucose levels were measured with glucometer. The enrolled neonates were divided into 2 groups based on CSF culture,CSF white blood cells(WBCs)and clinical presenta-tion:observation group(neonates with BM)and control group(neonates without BM). Statistical analysis of data was performed with Stata 12. 0. Results A total of 93 infants(16 assigned to observation group and 77 assigned to control group)met the inclusion criteria. Neonates in observation group had higher median CSF lactate level(4. 2 mmol/ L)and CSF lactate/ glucose ratio(L/ Gcsf)(2. 32 mmol/ L),than those in control group(1. 3 mmol/ L,0. 52),and there were significant differences(Z = - 6. 19,- 5. 92,all P ﹤ 0. 05). CSF glucose levels were lower in observation group(me-dian,1. 25 mmol/ L)than those in control group(median,2. 5 mmol/ L),and the difference was significant(Z = 4. 97, P ﹤ 0. 05);CSF/ serum glucose ratio(CSF/ Sglu)were lower in observation group(median,0. 44 vs 0. 81 in control group),and the difference was significant(Z = 4. 43,P ﹤ 0. 05). The optimal CSF lactate cutoff point of 2. 2 mmol/ L had a positive predictive value(PPV)of 72. 7% and negative predictive value(NPV)of 100. 0% for bacterial meningi-tis. The optimal L/ Gcsf cutoff point of 1. 24 had a PPV of 94. 1% and NPV of 100. 0% . The optimal CSF glucose cutoff point of 2. 0 mmol/ L had a PPV of 65. 0% and NPV of 95. 9% . The optimal CSF/ Sglu cutoff point of 0. 6 had a PPV of 60. 0% and a NPV of 96. 9% . Conclusion CSF lactate may be an excellent biomarker for early diagnosis of neo-natal BM.
