Objective To study the effects of the expression of hypoxia inducible factor-1 alpha (HIF-1?) induced by FasL on the invasiveness of rectal cancer cells. Methods With molecular cloning method, eukaryotic expression vectors of pcDNA3.1 FasL containing total length of FasL cDNA sequence were transfected into human rectal cancer cells HR-8348. The cell invasiveness was examined. A hypoxia model of HR-8348 was reproduced, and its invasiveness was assessed with transwell methodology. Expression of HIF-1? was assayed by Western bloting. Results The number of HR-8348 cells transfected with FasL penetrated transwell filter membrane was larger (12.930?2.434) than that of empty vector transfected cells (7.670?2.093) and control cells (8.133?1.959) with statistically significant differences (P0.05), but it was significantly higher in FasL positive HR-8348 cells than in FasL negative cells at 12h and 24h after hypoxia (P0.05). Conclusions FasL is an effective factor inducing HIF-1? expression, and FasL expression correlates positively with HIF-1? expression. Enhanced FasL expression in rectal cancer cells could induce higher expression of HIF-1?, enable the tumor cells to adapt to hypoxia environment, and enhance the invasiveness of tumor cells.

Objective To discuss the effect of interferon ?-2a on the radiosensitivity and cell cycle for nasopharyngeal carcinoma cell line CNE-I. Methods Interferon ?-2a was given by different concentration. Then the cells were radiated with X-ray (6?MV) and the cell survival rate was calculated. The change of cell cycle dynamics was measured with flow cytometry. Results The cell survival fraction was 0.62, 0.43 and 0.20 respectively after the interferon ?-2a was given by different concentration(0.5?106, 1.0?106 and 1.5?106?IU/L). The radiosensitization ratio was 1.16, 1.57 and 1.93 respectively. Compared with the control group, increasing cell percentage in G_1 and G_2+M phage and decreasing cell percentage in S phage were observed on 24 h after the interferon ?-2a(1?106?IU/L) was given(P

AIM:To investigate the effect of fluvastatin on the expression of serum and glucocorticoid inducible kinase 1 ( SGK1) and connective tissue growth factor ( CTGF) induced by aldosterone ( Ald) in rat mesangial cells (GMCs). METHODS:GMCs were divided into (1) control group; (2) aldosterone group with different concentrations and times; (3) Ald (10 -7 mol/L) + spironolactone (10 -9 mol/L) group; (4) Ald (10 -7 mol/L) + LY294002 (20 ?mol/L) group; (5) Ald (10-7mol/L) +SB203580 (20 mmol/L) group; (6) the group of Ald (10-7mol/L) + fluvas-tatin at different concentrations (10-7,10-6,10-5 mol/L); (7) Ald (10 -7mol/L) + fluvastatin (10 -5mol/L) + mevalonate (10 -4 mol/L) group; (8) Ald (10 -7 mol/L) + fluvastatin (10 -5 mol/L) + FPP (farnesyl pyrophosphate,10-4 mol/L) group; (9) Ald (10 -7mol/L) + fluvastatin (10 -5 mol/L) + GGPP (geranylgerany pyrophosphate,10 -4 mol/L) group. The protein levels of SGK1 and CTGF were determined by Western blotting. The levels of fibronection (FN),monocyte chemoattractant protein-1 (MCP-1) and intercellular adhesion molecule-1 (ICAM-1) in the supernatants were determined by enzymelinked immunosorbant assay (ELISA). RESULTS:Aldosterone stimulated the protein expression of SGK1 and CTGF in cultured mesangial cells in a dose-dependent manner (P

Objective:To observe the clinical efficacy of warm needling moxibustion plus tuina in treating knee osteoarthritis (KOA) due to cold-dampness blocking collaterals.
Methods:Forty-eight KOA patients were randomized into 2 groups by their visiting sequence, 24 cases in each group. The treatment group was intervened by warm needling moxibustion plus tuina, while the control group was treated with acupuncture plus tuina. The two groups were both treated once a day, 30 min for each session, 10 d as a treatment course, totally for 3 treatment courses. The visual analogue scale (VAS) and Lysholm knee scoring scale (LKSS) were observed before and after the treatment to evaluate the clinical efficacy.
Results:After 3 treatment courses, the VAS and LKSS scores were significantly changed in the two groups (P<0.01), and the between-group differences were also statistically significant (P<0.01). The markedly-effective rate was 83.3% in the treatment group versus 66.7% in the control group, and the difference was statistically significant (P<0.05).
Conclusion:Warm needling moxibustion plus tuina can produce a more significant efficacy than acupuncture plus tuina in treating KOA due to cold-dampness blocking collaterals.
