1.Effects of mild hypothermia used for protecting neurological function on electrolytes changes in swine model of ventricular fibrillation after cardiopulmonary resuscitation

Objective To study the changes of hemodynamic parameters and electrolytes observed within 72 hours of hypothermic therapy in porcine model of cardiac ventricular fibrillation after cardiopulmonary resuscitation (CPR ) in order to provide clinical basis for safe application of mild hypothermia.Methods After typical ventricular fibrillation (VF)for 8 minutes,the survival animals were randomly (random number)divided into two groups,namely normothermia group and hypothermia group. Upon restoration of spontaneous circulation (ROSC ),swine of the hypothermic group was treated by endovascular cooling device at a rate of 1 ℃/h until 33 ℃ and it was maintained for 12 h,then rewarming was initiated passively at a rate of 0.5 ℃/h until 38 ℃.The neurologic deficit scores (NDS)of swine were used to evaluate neurological function at 24 h and 72 h after recovery.Serum levels of potassium and sodium were measured at 0.5 h,6 h,12 h,24 h and 72 h after recovery.Results ROSC (restoration of spontaneous circulation)rate was 84.2%.The hypothermia group had higher survival rates at 24 h (75%) and 72 h (62.5%)compared to the normothermia group (37.5% and 25%,respectively),(P<0.01 ). There were no significant differences in hemodynamic variables (HR,MAP,CO,PAWP and CVP)between the two groups (P<0.05 ),Serum potassium of the hypothermia group was lower obviously at 24 h after recovery (3.41 ±0.11)(P<0.01),and there were no significant differences in serum sodium at all the intervals,and there were no significant differences in underlying trend of electrolytes changes over time (P>0.05).The mean NDSs at 24 h and 72 h after recovery was 112.5 (98.75 -126.25)and 61 (50-75), respectively,in the hypothermic group,and 230 (225 -235)and 207.5 (165 -250),respectively,in the normothermia group (P <0.01 ).Conclusions Hypothermia has little influence on serum levels of potassium and sodium,and mild hypothermia following resuscitation improves neurological function in the porcine models of cardiac fibrillation.

Recently,with the research progress in the pathogenesis of lung-damaging agents,numerous corresponding experimental studies were carried out and some progress were made. This paper summarizes the progress in the study of lung-damaging agents on the re-search situation,pathogenic mechanism and biomarker,to provide reference for the promotion of ALI prevention,medical antagonis-tic measures and clinical treatment.
3.Biological response modifiers combined with radiotherapy and chemotherapy for stage Ⅲ non-small-cell lung cancer

Purpose:To evaluate the effect of biological response modifiers for Ⅲ stage non-small-cell lung cancer.Methods:Fifty-nine patients were randomized into two groups.Twenty-nine patients were put into radiotherapy and chemotherapy(R+C) groups,thirty patients were into Biological response modifiers and radiotherapy chemotherapy(BRMS+R+C) groups.All patients received routine radiotherapy 2.0 Gy per day,5times a week to a total dose of 60-65 Gy,in 42-45 days.Radiation fields just covered clinical tumors.Chemotherapy consisted of cisplatin 25-30 mg/m 2 ,etoposide 50-70 mg/m 2 on days 1?2and 3 every 4 week,for squamous cell carcinoma.For adenocarcinoma,it was mitemycin 4-6 mg/m 2 on day1 and vindesine 3 mg on day1 and8,and cisplatin25-30 mg/m 2 on day1?2 and 3,every 4 weeks interferon-?(IFN-?)and interleukin-2(IL-2)were used in(BRMS+R+C)groups after the second chemotherapy was over. IFN-? was given on days 1?2?3 and IL-2 on days 2?4?6 to a total of forty-eight day.Results:The overall response rate was 51.7% and 76.6% in(R+C) and(BMRS+R+C) groups.The one.two year survival rates were 62.1% 17 3% and 83 3%,43 3% in (R+C) and (BMRS+R+C) groups respectively.Conclusions:The preliminary results from our study has shown that biological response modifiers combined with radiotherapy and chemotherapy for non -small cell lung cancer has better early response rate and survival rate.
4.The study of the cervical identiifed on CT iflm

Objective Selected the characteristic index of cervical CT tablets, and established stepwise regression equation, to explore the value of its application in forensic medicine identification. Method Each one cervical CT images of 130 cases were collected, 70 subjects were examined for 2 cervical CT images of each. Selected and measured 13 indicators of cervical CT iflms, the difference between the groups was calculated by the same measurement indicators after the random grouping of the inspectors, and the difference between the different measurement indexes of the same person. Using two classification logistic Stepwise regression analysis, establish the regression equation of the index and multiple regression equation of multinomial index, and the test of the equation was carried on blind test. Results Accuracy of 13 equations of the established element is 69.0%~90.4%,accuracy of 4 multiple regression equation is 90.4%~96.7%, blind accuracy rate is 100%. Conclusion 13 feature indicators selected from CT tablets can be used for the same identiifcation, in practice researchers should use multivariate index to get more accurate result.
5.Clinical efficacy of Qingpeng ointment for the treatment of chronic eczema

ObjectiveTo evaluate the efficacy and safety of Qingpeng ointment in the treatment of chronic eczema.MethodsTotally,95 patients with chronic eczema were enrolled and divided into 3 groups to receive topical Qingpeng ointment (n =33),0.2% hydrocortisone cream (n =32),10% urea ointment (n =30) respectively twice a day for 4 weeks.All the patients were followed up once a week for the evaluation of therapeutic and side effects.ResultsAfter 4-week treatment,the response rate was 78.8%,84.4% and 46.7% in the Qingpeng ointment-,hydrocortisone cream- and urea ointment-treated patients respectively.Significant differences were observed in the response rate between the Qingpeng ointment- and urea ointment-treated patients (P < 0.05 ),but not between the hydrocortisone cream- and Qingpeng ointment-treated patients (P >0.05).Skin flushing with a sensation of burning was observed at the application sites of 2 patients treated with Qingpeng ointment,and tingling sensation at the application sites was reported in 1 patient treated with hydrocortisone cream.All of these adverse reactions were tolerable and had no influence on the treatment.Conclusion Topical Qingpeng ointment is effective and safe for the treatment of chronic eczema.
6.The effects of TGF-β1 and Smad2 on liver fibrosis of biliary atresia

Objective To investigate the expression and function of transforming growth factor (TGF)-β1 and Smad2 in liver fibrosis of biliary atresia (BA). Methods Liver biopsy specimens were collected from autopsy (normal group, n=5), congenital biliary dilatation (CBD group, n=10), BA patients underwent Kasai procedure (early hepatic fibrosis group, n=19) and liver transplantation (transplantation group, n=11). The first three groups were collected from January 2010 to July 2014 in Tianjin Children’s Hospital, and the last group was collected from January 2013 to January 2014 in Tianjin First Central Hospital. The hematoxylin and eosin (HE) stain were used to observe the degree of liver fibrosis of four groups. Immunohistochemistry (IHC) was used to observe expressions of TGF-β1 and Smad2 in liver tissues of these samples. Quantitative real-time polymerase chain reaction (qRT-PCR) was used to test the quantitative mRNA of TGF-β1 and Smad2 in these samples. Results Results of HE showed that no fibrosis in autopsy group, mild fiber cell hyperplasia in CBD group, severe fibrosis in Kasai group and significant pseudolobule in transplantation group. Results of IHC showed that TGF-β1 was expressed in the cytoplasm of hepatocytes, bile duct cells, lymphocytes and neutrophils. The average optical density of TGF-β1 was the highest in Kasai group compared with that of other three groups (P < 0.05). There was no significant difference in Smad2 expression in cytoplasm of hepatocytes, bile duct cells and lymphocytes between four groups (P>0.05). Results of qRT-PCR showed that both TGF-β1 mRNA and Smad2 mRNA were the highest in early hepatic fibrosis group than those of CBD group and transplantation group (P<0.017). Conclusion In early stage of BA, TGF-β1 and Smad2 promote liver fibrosis until the formation of P-P,P-C desmosome structure. However, with BA fibrosis becomes more serious, the pro-fibrogenic function of TGF-β1 and Smad2 becomes less.

8.Study on mutations in exons 19, 20 and 21 of epidermal growth factor receptor gene in elderly patients with non-small cell lung cancer

ObjectiveTo investigate mutations in exons 19, 20 and 21 of epidermal growth factor receptor (EGFR) gene in elderly patients with non-small cell lung cancer (NSCLC). MethodsEGFR gene mutations in exons 19, 20 and 21 were detected by nested PCR amplification and DNA sequencing in 46 elderly patients with non-small cell lung cancer. The relationship between mutations and clinical characteristics of these patients was analyzed. ResultsEGFR gene mutations were found in 56.5% (26/46) patients and 41.3% (19/46)were non-silent mutations. Mutation of exon 19 was detected in 6 cases (13.0%), mutation of exon 20 in 13 cases(28.2%) and that of exon 21 in 14 cases (30.4%). Seven patients among them had double mutations and the rest only had a single mutation. The incidence of EGFR gene mutations was higher in non-smokers than in smokers(P< 0.01). Higher EGFR mutation rate in exon 19, 20 and 21 were found in patients with clinical benefit who were treated with tyrosine kinase inhibitor(TKl)(P<0.05). There was no difference in EGFR mutation rate between 60~69 age group and 70~85 age group. ConclusionsThe data suggest that the characteristics of EGFR gene mutations in elderly patients with NSCLC is the same as in the general NSCLC patients. The forecast informations of TKI treatment can be obtained by gene detection in elderly NSCLC patients.
10.Toxicity of glufosfamide in Beagle dogs following intravenous injection

Aim: To evaluate the safety of glufosfamide in Beagle dogs. Methods: The safety of glufosfamide in Beagle dogs following single iv and multiple iv administration for 3 months was observed. Results: The minimum lethal dose of glufosfamide was 100 mg/kg. The maximum non-lethal dose of glufosfamide was 50 mg/kg. The approximate lethal dose of glufosfamide was 75-100 mg/kg. No serious adverse reactions was observed in Beagle dogs following multiple iv administration once a week for 3 months at the dose of 20 mg/kg, 40 mg/kg, and 60 mg/kg. However, organ injury was observed extensively in the high dose-group and mediumt-dose group. The low-dose group showed a light change in individual organs. Conclusion: Under the experimental conditions, 40 mg/kg and 60 mg/kg iv administration once a week for 3 months can lead to extensive organ injury in Beagle dogs, so the dose shouldn't be higher than 20 mg/kg.
