1.Analysis of 5-HT_3 Receptor Antagonists Used in 25 Hospitals of Wuhan During the Period of 2003-2005

Objective:To evaluate the current situation and the developing trend of 5-HT3 receptor antagonists used in 25 hospital of Wuhan during the period of 2003~2005 in order to provide reference for clinical doctors and pharmacists. Method:The 5-HT3 receptor antagonists were statistically analyzed in respect to the purchase sum of money,DDDs and daily costs. Result:During the 3 years, purchase sum of money of 5-HT3 receptor antagonists were yearly increasing. The share of the market of domestic drugs showed an increasing tendency,while the imported drugs decreased year by year. Granisetron, ondansetron and tropisetron were much more frequently used than others. Conclusion:The market of the oral agents of 5-HT3 receptor antagonists will further expand in China.
2.Clinical evaluation of coronary angiography by prospective ECG gated sequence scanning using second generation dual source CT:no heart rate control

Objective To analyze the effects of image quality and radiation dose of the patients with no heart rate control on different acquisition window settings.Methods A total of 75 patients underwent CTCA scan,with a median age of 55 years.All patients had no heart rate control.HR≤65 beats per minute acquisition phase setting was from 65%to 75%,65 beats per minute
80 beats per minute acquisition phase setting was from 35%to 45%.Evaluated patients radiation dose and subjective image quality of coronary artery.Results 75 patients with ED was (2.44±1.21)mSv,group A was (1.54±0.41)mSv, group B was (3.57±0.53)mSv,group C was (1.64±0.27)mSv,group A vs group B(P=0.000),group A vs group C(P=0.854),group B vs group C(P=0.000).A total of 1 043 coronary artery segments of 75 patients were evaluated,with an average of 13.9 segments showed by every case,(3.66±0.51)scale.Group A was (3.75±0.44)scale,group B was (3.60±0.54)scale,group C was (3.38±0.67) scale,group A vs group B(P=0.000),group A vs group C(P=0.004),group B vs group C(P=0.032).Conclusion The prospective ECG gated sequence scan could obtain the image quality and avoid unnecessary radiation dose by adapting to the different heart rate.
3.Analysis and Countermeasures of Microbial Contamination in Methadone Hydrochloride Oral Solution

Objective:To analyze the causes of microbial contamination in methadone hydrochloride oral solution used for the ma-intenance treatment of heroin addict in Guangxi and propose the relative countermeasures. Methods:The samples of methadone hydro-chloride oral solution were obtained from 10 maintenance treatment clinics, and then the microbial limit test and identification were per-formed,the content of preservative was detected and the relationship between the preservative content and microbial contamination was studied. Results:The total number of aerobe in the samples of methadone hydrochloride oral solution from the 10 clinics was all over the standard limit value of 100 cfu·ml-1 ,and the main contaminating germ was Burkholderia cepacia. The contents of sorbic acid were 78. 2%-98. 3%,the four batches with the lowest concentration of preservative were detected with higher total number of aerobe. Con-clusion:The microbial contamination in methadone hydrochloride oral solution from the 10 maintenance treatment clinics is severe,and the environment of medicine packaging, content of preservatives, packaging machine and storage conditions are the primary causes for the microbial contamination. To ensure drug quality safety, it is suggested that the environment cleanliness of medicine packaging be improved, suitable content of preservatives be added, packaging machine be clean, the products be stored in shade and the quality be tested regularly.
4.Effect of Annonaceous Acetogenin on Proliferation and Apoptosis of Raji Cell

Objective To investigate the effects of annonaceous acetogenin on proliferation and apoptosis in Raji cells and its mechanism.Methods Raji cells cultured in vitro were divided into control group,annonaceous acetogenin group and adriamycin group.Raji cells were effected by 6.25,12.5,25,50 ?g/mL annonaceous acetogenin.Proliferation of Raji cells were evaluated by MTT assays,apoptosis percentage was assessed by flow cytometry.Caspase-9 protein was detected by immunohistochemistry.Results The Raji cell proliferation rate of annonaceous acetogenin decreased compared with the control,that of 25,50 ?g/mL group were lower than adriamycin group,and it was related to the concentration,relying on the incubating time.The apoptosis rate was higher than control,that of 25,50 ?g/mL group were higher than adriamycin group,and it was related to the concentration and the incubating time.The expression of caspase-9 protein of annonaceous acetogenin group was higher than control,and it had a positively relationship with the concentration and incubating time.Conclusions Annonaceous acetogenin could inhibit cell proliferation in a dose-dependent and time-dependent manner in Raji cells,and it may induce Raji cells apoptosis by up-regulating caspase-9 expression.
5.Chemical constituents of medicinal fungus Shiraia bambusicola

Objective To study the chemical constituents of the medicinal fungus Shiraia bambusicola.Methods Chromatography on silica column and preparative HPLC were used to purify the compounds;Spectroscopic methods including 1H-NMR,13CNMR,and MS were used to elucidate their structures.Results Nine compounds were isolated and identified as 3,6,8-trihydroxy-l-methylxanthone(1),3,8-dihydroxy-6-methoxy-1-methylxanthone(2),2,3,6,8-tetrahydroxy-l-methylxanthone(3),3,4,6,8-tetrahydroxy-l-methylxanthone(4),pregn5(10)-en-3?,17?,20?-triol(5),macrosphelide A(6),(+)-griseofulvin(7),griseophenone A(8),and 11,11′-dideoxyverticillin A(9).Conclusion Compounds 1-9 are obtained from the fungus S.bambusicola for the first time.
6.Artificial amniotic fluid replacement using amniocentesis improves intrauterine hypoxia of fetal rats during the treatment of intrahepatic cholestasis of pregnancy

BACKGROUND: Drug therapy can improve the clinical symptoms and extend the gestational age through the maternal way to treat intrahepatic cholestasis of pregnancy (ICP), however, fetal hypoxia and meconium-stained amniotic fluid can not clearly improved. OBJECTIVE: To explore the effect of replacement of amniotic fluid that contains high concentrations of bile acids by artificial amniotic fluid through amniocentesis in the rat model of ICP. DESIGN, TIME AND SETTING: Random control animal experiments were completed in the Laboratory (first-degree laboratory) of Animal Experimental Center in Chongqing Medical University from March to September 2008. MATERIALS: Forty SD pregnant rats with 15 days of pregnancy were randomly divided into intracavitary injection of S-Adenosyl-L-methionine (SAMe) (IUS), intracavitary injection of sodium chloride (IUN), intravenous injection of SAMe (IVS) and blank control (IC) groups, with 10 rats in each group. METHODS: On day 15 of pregnancy, ICP rat models were induced by daily injection of estradiol benzoate. On day 17 of pregnancy, 1 mg SAMe and 0.3 mL sodium chloride were injected into amniotic cavity of in the IUS and IUN groups through amniocentasis. Meanwhile, 30 mg SAMe were injected though tail veins in IVS group. No treatment was performed in the IC group. MAIN OUTCOME MEASURES: The meconium-stained amniotic fluid rate and the still birth rate were calculated. Total bile acid (TBA) level in amniotic fluid was measured. In addition, the expression of hypoxia-inducible factor 1 ?(HIF-1?) mRNA in the placenta of rats were examined by immunohistochemistry and reverse transcription PCR at the days 20 of pregnancy. 8.Research of Access Control Mechanism in Hospital Information System

Objective To present a solution of the hospital information management access control for more secure information sharing. Methods Several commonly used access control models were compared. Some problems of the RBAC model were analyzed in the hospital information circumstances. Results A solution of space-based data access control was presented on the base of the role-based access control (RBAC) module. Conclusion The space-based data access control method can reduce space consumption and maintain data consistency, which is more suitable for the use of the hospital management information system.
9.Thematic Research on Therapeutic Manipulating Process of 4 Systemic Family Therapists

Object:To describe Chinese systemic family therapy process through analysis on the therapeutic experience of 4 Chinese systemic family therapists.Method:Four well-certificated systemic family therapists in China were selected as research sample.A semi-structure interview under the guidance of a self-compiled interview outline was made with every therapist.Transferred raw data was analyzed with deductive thematic analysis following 5 steps:coding,themes searching,themes reviewing,themes defining and naming,themes merging and revision.Results:(1)Respectively 102,93,97,and 95 codes were got from each piece of interview raw data,which generated 57,52,50,and 33 primary themes.Five main themes emerged from the merging of the primary themes.(2)Themes and sub-themes constructed the theoretical hypothesis on the change process of Chinese systemic family therapy.Conclusion:There is an experiential manipulating model in Chinese systemic family therapy process under the guidance of systemic therapeutic philosophy,which is integrated with some therapeutic technique from other psychotherapy models.This model can be studied with deductive thematic analysis.
10.Effects of microRNA-483-3p on human glioma cells

AIM: To investigate the effects of microRNA (miRNA)-483-3p on the growth and migration of human glioma cell line A172 and its potential mechanisms.METHODS: The abundance of miRNA-483-3p in human embryonic kidney 293 cells and different human glioma cell lines (A172, U251 and SHG44) was measured by RT-qPCR.After down-regulation of miRNA-483-3p by transfection of inhibitor in the A172 cells, the cell viability, cell cycle distribution and cell migration were detected by CCK-8 assay, flow cytometry and Transwell assay, respectively.Furthermore, the protein levels of cell cycle-related molecules and epithelial-mesenchymal transition markers were measured by Western blot.Luciferase reporter assay was used to predict and verify the target gene of miRNA-483-3p.RESULTS: miRNA-483-3p was highly expressed in human glioma cells.Knockdown of miRNA-483-3p inhibited A172 cell viability, arrested cell cycle and decreased cell migration rate.Furthermore, the protein levels of cyclin D1, cyclin-dependent kinase 4, phoshorylated retinoblastoma protein, N-cadherin and vimentin were significantly decreased after knockdown of miRNA-483-3p, accompanied with the up-regulation of E-cadherin and β-catenin protein expression.Luciferase reporter assay demonstrated that Smad4 was a potential target gene of miRNA-483-3p.Down-regulation of Smad4 in the A172 cells transfected with miRNA-483-3p inhibitor partially reversed the effect of miRNA-483-3p on cell viability and migration.CONCLUSION: Knockdown of miRNA-483-3p restrains the growth and migration of A172 cells by targeting Smad4.
