Chinese Journal of Tissue Engineering Research 2015;(42):6735-6739
BACKGROUND:In recent years, calcitonin has been reported to have better clinical efficacy in the treatment of osteoarthritis, but its mechanism of action for osteoarthritis is rarely reported. OBJECTIVE:To investigate the effect of calcitonin on the cartilage morphology and proteoglycan expression in rats with osteoarthritis. METHODS:Thirty Sprague Dawley rats were randomly divided into three groups: model group, treatment group, and sham group. Anterior cruciate ligament transaction of the right limbs was implemented in the model and treatment groups, and only the joint cavity was exposed in the sham group. At 2 days after modeling, the treatment group received a daily subcutaneous injection of salmon calcitonin, 15 IU/(kg?d), and the model group and sham group were administered with normal saline at the same dose. The injection lasted for 6 weeks. At 10 weeks after modeling, the articular surface of the tibia of rats in each group was generaly observed; bone mineral density of the distal femoral bone and subchondral bone of the lateral and medial ankle were detected using X-ray test; bone morphology and proteoglycan secretion were measured by hematoxylin-eosin staining and toluidine blue staining, respectively. RESULTS AND CONCLUSION:The tibial articular surface was smooth and glossy in the sham group, but oxblood in the model group with large-area ulcers; the treatment group showed rough and local ulceration. Compared with the sham group, the bone mineral density of the subchondral bone of medial and lateral ankle were increased significantly in the model group (P < 0.05), but trabecular separation and proteoglycan content were decreased (P < 0.05). Compared with the model group, the bone mineral density of the subchondral bone of medial and lateral ankle were decreased significantly in the treatment group (P < 0.05), while the trabecular separation and proteoglycan content were increased (P < 0.05). These findings indicate that calcitonin has better protection on the cartilage of rats with osteoarthritis, and can promote bone the secretion of proteoglycan.

Objective To verify the antibacterial activity of the conserved domain derived from the novel human antimicrobial peptide-hGlyrichin .Methods Bioinformative analysis was performed and two peptides derived from hGlyrichin were synthesized which contained the conserved domain .Results Analysis of antimicrobial activities showed that these two peptides exhibited strong antibacterial activity which was inversely proportional to the length of the peptide within an eligible range.Despite the effective inhibition and killing of bacteria , the synthetic peptide segments had no hemolytic effect on human red blood cells .Conclusion These results indicate that a conserved domain exists in hGlyrichin , and that the peptides which contain this domain have strong antibacterial activity but are not toxic to human somatic cells .
9.Effect of Hyperbaric Oxygen on p38 MAPK Signal Transduction Pathway in Neuropathic Rats
Journal of China Medical University 2016;45(8):723-727
Objective To investigate the effect of hyperbaric oxygen(HBO)on p38 MAPK signal pathway in neuropathic pain, and explore its mechanism. Methods The experiment was divided into two parts with thirty SD rats in each part. Each part was then randomly divided into 3 groups:sham operation group(S group),chronic constriction group(CCI group)and hyperbaric oxygen group(HBO group),with 10 rats in each group. Part one:Pain?related behavior were detected on 3 day,7 day,14 day,28 day after operation. Phosphorylative p38 was detected by Western blot method and the P2X4 receptor was detected by immunohistochemistry method on the 28th day. Part two:All the rats were treated with SB203580. Pain?related behavior were detected on 3 day,7 day,14 day,28 day after operation. P2X4 receptor was detected by immunohistochem?istry method on the 28th day. Results CCI and HBO groups were significantly lower than S group in pain?related behaviors(P<0.05). CCI group was significantly lower than that in HBO group(P<0.05). The content of phosphorylative p38 and P2X4 receptor in CCI and HBO group were increased(P<0.05),and the expression of phosphorylative p38 and P2X4 in CCI group was significantly increased than that in HBO group (P<0.05). When p38 MAPK was inhibited, pain?related behavior in HBO and CCI group increased significantly than the pain?related behavior of HBO and CCI group in Part 1(P<0.05). But the difference between group CCI and HBO was not significant(P>0.05). Though the expression of P2X4 receptor in CCI group and HBO group was significantly higher than that in S group(P<0.05),the difference between group CCI and HBO was not significant(P>0.05). Conclusion Hyperbaric oxygen therapy can affect the expression of p38 MAPK through P2X4 receptor in rats of neuropathic pain.
