2.Effcts of the panaxadiol saponins on serum 5-HT and platelets in dog with hemorrhagic shock

Experiments were performed on 29 healthy mongrel dogs with average body weight of 13?2.2kg. The animals were randomly divided into three groups:(1) nine dogs in the hemorrhagic shock group(HS); (2) tendogs in the panaxadiol saponin pretreatement group (HSG), which were intravenously injected with 25mg panaxadiol saponin 1 h before bleeding; (3) In the dexamethasone pretreatment group, othet ten dogs were intramuscularly injected with 1 mg/kg body wt of dexamethasone 1 h before bleeding. The mean arterial pressure of 5.3 kPa was kept during experimental period. The morphological changes of platelet were observed with scanning electromicroscope. The results showed that the platelet damage was much lighter in HSP and HSD gnoups than that of the HS group. The seum 5-HT content of HS group gradually increased hemorrhage with time delay. In the HSG group,the 5-HT level was increased from 1 and 3 h after bleeding,then it was decreased at 4 and 5 h. In the HSD group 5-HT level didn′t show any change from prebleeding to postbleeding. However, in the HS group 5-HIAA concentration increased significantly from 3rd to 5th hour after bleeding in the HS group,but was not in the HSG and HSD groups. These findings suggest that the panaxadiol saponins and dexamethsone can effecti vely inhibit the increasing of 5-HT and 5-HIAA levels in the themorrhagic shock.
3.Ovarian endometriosis and the risk of malignancy

Objective To explore the clinical and pathological characteristics of ovarian endometriosis and malignant cases, and to improve our recognition of this disease. Methods A total of 1 289 cases of ovarian endometriosis were divided into two groups: the benign group (1 275 cases)and the malignant group (14 cases). They were analyzed on: dysmenorrhea, infertility, features of ultrasound examination,site of tumour,pathological types and complications. Results Malignancy arising from ovarian endometriosis was 1.09%. As far as degree of dysmenorrhea, size of cyst >10 cm, CA125 >500 U/L, features of ultrasound were concerned, difference between the two groups was statistically significant. Conclusions In the present study, the malignant rate of ovarian endometriosis was about 1%. Malignancy should be considered seriously when the following situations occured: change in the degree of dysmenorrhea, size of cyst larger than 10 cm, CA125 higher than 500 U/L, mamilla observed by ultrasound examination.

6.Preparation of medical polypropylene anti-bacterial mesh coated with triclosan and detection of its nature

Objective To explore the production of medical polypropylene anti-bacterial mesh coated with triclosan,and study the properties of its controlled-release drugs,as well as the efficiency of the mesh coated with triclosan.Methods A medical polypropylene anti-bacterial mesh coated with triclosan was produced by the method of solvent evaporation,with triclosan as antibacterial agent and ethylene-vinyl alcohol copolymer as carrier.The meshes were divided into four groups: A,B,C and D.The meshes in A,B and C(experimental) groups were coated with triclosan,and D group was control group.The ultrastructure of meshes of the 4 groups was observed under electron microscope.The slip diffusion method was used to observe the diameter of bacteriostasis-rings in different time periods.At the same time,we observed the maximum tensile strength of the mesh in transverse and longitudinal axis when they were pulled off before and after the experiment.Results The surfaces of B group were uniform and smooth,and the coating could continuously release drugs during 7 days in sodium chloride.The diameter of bacteriostasis-rings in B group was significantly larger than that in control group(P
7.MRI research progress of spinal stability changes based on lumbar intervertebral disc degeneration

The lumbar intervertebral disc is a disc-shaped fibrocartilaginous structure located between the vertebral bodies and acts as an important anatomical structure in the spine for stability. The spinal stability system consists of three subsystems: passive subsystem, active subsystem, and neural control subsystem. Lumbar intervertebral disc degeneration (IVDD), the leading cause of low back pain, induces pathological changes in other tissues of the three subsystems and interacts to degrade spinal stability. IVDD is commonly accompanied by degeneration of facet joint and ligament, Modic change, decreased vertebral blood flow, increased paraspinal muscle fat infiltration, reduced axial tension injury of the nerve, et al. Magnetic resonance imaging (MRI) is the preferred imaging modality for assessing spinal stability. Conventional MRI could completely demonstrate the morphological changes of the three subsystems in IVDD patients, and functional MRI could quantitatively evaluate the degree of pathophysiological change. We summarized the morphological and functional changes of MRI in lumbar intervertebral disc, facet joint, ligament, vertebral body, paraspinal muscle, and nerve of patients with IVDD, and then analysed the changes in spinal stability caused by IVDD, aiming to provide more imaging information for improving diagnosis accuracy and developing appropriate management of patients with low back pain.
8.Detection of CGRP receptor in rat bone marrow mesenchymal stem cells and its induced osteoblasts

[Objective]To compare the expression of calcitonin gene-related peptide(CGRP) receptor in rat bone marrow mesenchymal stem cells(rBMSCs) and the induced osteoblasts.[Method]The rBMSCs were isolated using whole bone marrow adherence method,and classified into osteoblast-induced group and non-induced group.In the different periods of culturing(1,2,and 3 weeks),identification of osteoblasts was performed by immunohistochemistry and reverse transcriptase-polymerase chain reaction(RT-PCR),and expression of the CGRP receptor was detected by Western Blot and RT-PCR.[Result]RT-PCR test demonstrated that osteoblast-induced group had a higher expression of CGRP receptor than non-induced group at the same time point,expression of CGRP receptor was increased in a time-dependent manner in the osteoblast-induced groups.Western Blot test demonstrated that osteoblast-induced group had a higher expression of CGRP receptor than non-induced group at the same time point.Expression of CGRP receptor was increased in a time-dependent manner in osteoblast-induced groups.[Conclusion]According to the mRNA test and protein test,it is suggested that CGRP receptor exists in the rBMSCs.CGRP receptor expression increased in the progress of osteogenic differentiation.
9.The comparison of genetic susceptibility of type 1A diabetes mellitus between Asian and Caucasian populations

Some human leucocyte antigens(HLA),espeually class Ⅱ genotypes such as HLA DQ/DR are closely linked to type 1A diabetes mellitus(T1ADM).But the genetic susceptibility of HLA DR/DQ are different between Caucasian and Asian populations.The aspartic acid at position 57 of DQ? can not confer resistance to T1ADM in Asians.DQ2 and DQ8 are the well-known susceptible genotypes in Caucasians while DQ4 and DQ9 are in Asians.An unexpected high risk of T1ADM is observed for the linkage disequilibrium between DRB1*0301/ DRB1*0401 and DQ2/DQ8 in both the populations.The DQB1*0302,DQA1*0501,DRB1*0401,*0402,*0407 are associated with T1ADM in Caucasians,but it is not the case in Asians.In contrast,DQB1*0401 shows a positive association to T1ADM in Asians,has a protective effect against T1ADM in Caucasians.Additionally DRB1*09 could be another susceptible gene in Asians.
10.High phosphorus foods combined with 5/6 nephrectomy for establishing rat hyperparathyroidism model secondary to chronic renal failure

Objective:To establish a rat hyperparathyroidism model secondary to chronic renal failure,so as to lay a foundation for studying the mechanism and treatment of secondary hyperparathyroidism.Methods: Thiry-six male Sprague-Dawley rats,weighing 200-250 g,were randomly divided into 6 groups: 5/6 nephrectomy plus high phosphorus food group(STNx + HP),5/6 nephrectomy plus low phosphorus food group(STNx + LP),5/6 nephrectomy plus normal phosphorus food group(STNx + NP),sham operation plus high phosphorus food group(Sham + HP),sham operation plus low phosphorus food group(Sham + LP),and sham operation plus normal phosphorus food group(Sham + NP).Nephrectomy was performed in 2 steps.Serum phosphorus levels(P) and iPTH levels were detected at day 7 pre-operation and day 7,14,and 21 after the second operation.The kidneys,thyroid glands and parathyroid glands complex underwent pathological analysis 4 weeks after operation.Results: Five patients survived in STNx+NP group,4 in STNx+LP group and 4 in STNx+HP group postoperatively.No death occurred in sham operated groups.Serum phosphorus levels of nephrectomy groups at different time points after operation were higher than those before operation and those of sham operation groups(P
