1.Protective Effect and Molecular Mechanism of Hirudin on Kidney of Diabetic Rats

To investigate the protective effect of hirudin on kidneys of diabetic rats and its molecular mechanism. Eighteen Sprague-Dawley rats were randomly divided into control group ( Compared with the control group, the diabetic model group showed significantly elevated random blood glucose and renal function parameters during the 6-week treatment (both Hirudin improved renal pathological changes in diabetic rats, and its mechanism may be related to the decrease of fibrosis-related factors and inhibition of the activation of the JAK2/STAT3 pathway.
2.Current Status and Trend Analysis of Disease Burden of Rheumatoid Arthritis in China from 1990 to 2019

Objective To provide a scientific basis for the prevention and control of rheumatoid arthritis(RA)in China by analyzing the current status and trend of disease burden in China from 1990 to 2019.Methods The incidence,prevalence,mortality,and disability adjusted life years of RA from 1990 to 2019 were analyzed based on GBD data.DALYs and other indicators,and comparative analysis of the annual development trend of the levels in China,the United States,Japan and the world through the scaling rates of the four indicators.Results In 2019,the incidence,prevalence,mortality,DALYs rate and standardized rate of RA in China were higher in females than in males,and the mortality rate increased with age,and peaked in the age group over 95 years old.The incidence rate,prevalence rate and DALYs rate increased firstly and then decreased with theincrease of age,and reached the peak in age group 60~,70~and 85~years old,respectively.From 1990 to 2019,the incidence,prevalence,mortality and DALYs of RA in China showed an increasing trend.After eliminating the differences in population age composition,the standardized incidence and prevalence of RA in China showed an increasing trend.The normalized RA mortality rate and the normalized DALYs rate showed a steady trend in China.In 2019,the normalized incidence,prevalence and DALYs of RA in China were lower than those in Japan,the United States and the world.In 2019,the standardized mortality rate was higher than that of Japan,the United States and the world.Conclusion In the past 30 years,the disease burden of RA in China has been on the rise.It is necessary to strengthen the publicity and education of RA prevention and control,especially the prevention and control of middle-aged and elderly women,and formulate reasonable policies to further reduce the disease burden of RA in China.
3.Targeted metabolomics reveals the aberrant energy status in diabetic peripheral neuropathy and the neuroprotective mechanism of traditional Chinese medicine JinMaiTong

Diabetic peripheral neuropathy(DPN)is a common and devastating complication of diabetes,for which effective therapies are currently lacking.Disturbed energy status plays a crucial role in DPN pathogenesis.However,the integrated profile of energy metabolism,especially the central carbohy-drate metabolism,remains unclear in DPN.Here,we developed a metabolomics approach by targeting 56 metabolites using high-performance ion chromatography-tandem mass spectrometry(HPIC-MS/MS)to illustrate the integrative characteristics of central carbohydrate metabolism in patients with DPN and streptozotocin-induced DPN rats.Furthermore,JinMaiTong(JMT),a traditional Chinese medicine(TCM)formula,was found to be effective for DPN,improving the peripheral neurological function and alleviating the neuropathology of DPN rats even after demyelination and axonal degeneration.JMT ameliorated DPN by regulating the aberrant energy balance and mitochondrial functions,including excessive glycolysis restoration,tricarboxylic acid cycle improvement,and increased adenosine triphosphate(ATP)generation.Bioenergetic profile was aberrant in cultured rat Schwann cells under high-glucose conditions,which was remarkably corrected by JMT treatment.In-vivo and in-vitro studies revealed that these effects of JMT were mainly attributed to the activation of adenosine monophosphate(AMP)-activated protein kinase(AMPK)and downstream peroxisome proliferator-activated receptor gamma coactivator 1-alpha(PGC-1α).Our results expand the thera-peutic framework for DPN and suggest the integrative modulation of energy metabolism using TCMs,such as JMT,as an effective strategy for its treatment.
