1.Dendritic cell vaccines in glioma treatment

Dendritic cell ( DC)-based tumor vaccine is applied to the gliomas treatment. Target selection heightens the specificity of DC vaccine, while obtaining of highly immunogenic long-lived and more effective DC vaccines by genetic engineering to antiapoptotic protein gene transfer. With the accumulation of clinical experience, the clinical curative effect of DC vaccine has been materially improved.
2.Recent advance in Pygopus

Pygopus(Pygo)is a new discovered component of Wnt signaling,which is located in the downstream of its core protein β-catenin and can mediate β-catenin into nucleus and activate target gene tran-scription. Pygo plays an important role in mammalian embryonic development and tumor formation. Recent studies show that Pygo regulates stem cells,tumor cells and tumor stem cells by epigenetic mechanisms such as histone interpretations and modifications. Further insights into Pygo′s functions and mechanisms will extend our knowledge of the Pygo-related tumors.
3.STUDY OF TWO-PEAK FEATURE OF c-fos EXPRESSION IN NEURONS AFTER SEVERE BRAIN INJURY AND ITS MECHANISM IN MICE

To investigate the time dependent features and mechanisms of c fos expression in neurons after brain injury in mice. Using the mice model of severe traumatic brain injury, c fos expression in neurons was observed at different time points after trauma with the molecular hybridization and immunohistochemistry staining. A c fos expression process in neurons occurred after severe brain injury, peaking at 1 hour. Another expression peak appeared 24 hours later. Fos positive neurons were observed in the cerebral cortex of injured hemisphere, especially in Ⅱ Ⅳ layers. The c fos gene expression after brain injury in neuron has two peaks. c fos gene expression may be caused by Leao’s spreading depression, and may be associated with cell signal transduction and neuron apoptosis.
4.Correlation between HLA gene and Takayasu arteritis in the Chinese

Objective To determine the correlation between HLA gene and Takayasu arteritis(TA) in the Chinese. Methods Forty four TA patients and 135 healthy controls were examined for HLA DQA 1 and HLA DQB 1 locus alleles by PCR/SSOP typing and mass genetics techniques. Results The frequency of DQA 1 0301 allele(14.8%) in TA patients was significantly decreased compared with the controls( 30.7% )(P0.05). Conclusions In Chinese, people with HLAⅡ DQB 1 0601 gene may be susceptible to TA, while people with HLAⅡ DQA 1 0301 gene may be resistant to TA.
5.Expression of EGFL7 in human glioma and its relationship with FAKpY397 and MVD

ObjectiveTo test the expression of epidermal growth factor-like domain 7 (EGFL7),microvessel density (MVD) and foeal adhesion kinase pY397 (FAKpY397) in human glioma tissues,and to evaluate their relationship.MethodsThe expression of EGFL7 and FAKpY397 in 56 cases of human glioma and 8 cases of normal brain tissues were detected by immunohistochemistry test,and MVD was detected by CD34 staining.ResultsThere was a significant difference of the positive rates of EGFL7 between normal brain tissue (0) and gliomas (75%),χ2 =17.45,P <0.01.With the increased pathological grade,the expression level of EGFL7 increased (χ2 =26.24,P < 0.01 ).There was a significant difference of the positive rates of FAKpY397 between normal brain tissue ( 12.5% ) and gliomas (73.2%),χ2 =6.23,P < 0.05.With the increased pathological grade,the expression level of FAKpY397 increased (χ2 =6.71,P < 0.01 ). MVD on normal brain was( 15 ± 4 )/HP,on Ⅰ - Ⅱ grade and Ⅲ -Ⅳ grade gliomas was ( 27 ± 3 )/HPand ( 60 ± 4 )/HP respectively,there was a significant difference on MVD between normal brain tissue and gliomas (P < 0.01 ).Higher level of MVD was found in gliomas with higher grade ( P < 0.01 ).There was a positive correlation between EGFL7 and FAKpY397 expressions in gliomas (r =0.314,P <0.01 ).There was a significant difference on MVD between positive and negative expression of EGFL7 ( t =26.55,P < 0.01 ). MVD was (56 ± 4 )/HP and (25 ± 3 )/HP respectively.ConclusionThe expression of EGFL7 of human gliomas has a favorable positive correlation with the degree of malignancy,MVD and FAKpY397.It is indicated that EGFL7not only palys an important regulative role in glioma neovascularization,but also it may participate directly in glioma occurrence and invasion.
6.DISTRIBUTION OF iNOS IN THE OVARY,OVIDUCT AND UTERUS OF EARLY PREGNANT MICE

Objective To understand the distribution of iNOS in the ovary, oviduct and uterus of mouse(2 5 days of pregnancy). Methods Immunohistochemistry(LSAB) was used in this study. Results iNOS occured in the luteal cells of all detected ovaries. In the oviduct tissues, iNOS was only found in the mucosa. It also appeared in the epithelia of endometrium and the metrial glands of all detected uterus. In the decidua of both day 4 and day 5 pregnant mice and the day 5 embryo surface, there are the presence of iNOS. Conclusion iNOS can be found in the ovaries,oviducts and uterus during 2 5days of pregnant mice, which suggests that iNOS plays a role in the development of early mouse embryos and the implantation.
7.IMMUNE RESPONSES IN PREGNANT MOUSE UTERUS INDUCED BY A SPERM ANTIGEN SA-30

Objective To detect the changes of immune cells in mouse uterus induced by SA 30(a Con A binding glycoprotein isolated from bovine sperms), and then reveal the influence of SA 30 immunization on the development of early mouse embryos. Methods The CD4 +、CD8 + and CD57 + cells in uterus of SA 30 immunized or control female Kunming mice were detected by immunohistochemical method(LSAB). Results On 3 day of pregnancy, much more CD4 +、CD8 + and CD57 + cells occurred in the uterus of SA 30 immunized mice than those of the control mice. On the other hand, the positive cells were found both in the endometrium and myometrium of immunized mice, but only in myometrium of control mice. Conclusion SA 30 can induce the cell mediated immune responses in mouse uterus, which may be toxical to the early embryos.
8.Mutation analysis of GJB2 gene in a family with Vohwinkel syndrome

ObjectiveTo detect the mutation of GJB2 gene in a Chinese family with Vohwinkel syndrome.MethodsClinical data were collected from 5 patients with Vohwinkel syndrome in a family,and blood samples were obtained from the 5 patients and 4 unaffected individuals in the family as well as from 100 normal human controls.Genomic DNA was extracted and subjected to PCR for the amplification of the entire encoding and flanking sequences of GJB2 gene(1015 bp) followed by bidirectional sequencing with the ABI PRISM 3730 automatic DNA sequencer.Finally,sequence alignment was carried out by using the software Sequencher 4.10.1 Demo.ResultsA heterozygous missense mutation 196G→C in the GJB2 gene,which resulted in the substitution of aspartic acid by histidine at codon 66 (D66H) in the first extracellular domain of the protein,was observed in all the patients of this family,but in none of the 4 unaffected individuals in this family or the 100 normal human controls.ConclusionThe D66H missense mutation in the GJB2 gene may contribute to the occurrence of Vohwinkel syndrome in Chinese Han population.
9.Surgical treatment of Takayasu arteritis accompanied with aneurysms

Objective To discuss the diagnosis and surgical treatment of Takayasu arteritis accompanied with aneurysms. Methods The clinical features and the effect of operation types in 14 patients were reviewed.Results All the 14 patients had history of Takayasu arteritis and had steroid therapy before the formation of aneurysms.Among the 14 patients,aortic aneurysms were found in 13(including 9 thoracoabdominal aneurysms),carotid aneurysm with subclavian aneurysm in 1 and multiple aneurysms in 5.All the 14 patients underwent operations ,including replacement of aorta in 11 cases,bypass of aorta in 3 cases,reconstruction of visceral vessels in 8, renal autotransplatation in 1 and replacement of carotid in 1.1case had heart failure and cured ;1 died postoperatively. Eight cases were followed up for 4 months - 18 years,of which, 1 died of heart failure,2 were excellent, 3 of 5 cases with hypertension preoperatively showed normal blood pressure,and the other 2 were controlled by medicines.Conclusions Operation should be performed as early as possible if the aneurysm is found in patients with Takayasu arteritis.The stenosis of renal artery should be resolved during the operation.
10.Effect Evaluation on Association Rules of Symptoms in TCM Treatment of Viral Pneumonia

This study was aimed to explore an effect evaluation method of traditional Chinese medicine (TCM) in the treatment of viral pneumonia. Association rules were used to analyze the therapeutic effect of heat, stagnation, and phlegm related syndromes from 297 respiratory syncytial virus (RSV) pneumonia children before and after treatment. The results showed that through the comparison of the frequent itemset of heat, stagnation, and phlegm related syn-dromes, in part of the time, the support degree of the experimental group was obviously lower than that in the con-trol group (P < 0.05). The frequent itemset of the experimental group disappeared 1~3 d earlier than the control group, which indicated the effect in the experimental group was better than the control group. It was concluded that association rules can be used for evaluating therapeutic effect of TCM in treatment of viral pneumonia. The effect evaluation method based on association rules of symptoms is more in line with the syndrome differentiation of TCM.
