1 cases of pleomorphic hyalinizing vascular tumor(PHAT) at the right side of neck soft tissue is reported and related literatures are reviewed.The clinical symptoms of PHAT are mild and non-characteristic,imaging examination and postoperative pathological examination may show specificity of the lesion.It is a kind of soft tissue tumor with malignant tendency.Surgical excision is the main treatment,long-term follow-up is recommended.
2.Culture construction in college and university medical library

3. Extension and dentin differentiation potential of dental pulp stem cells from human deciduous teeth on the stiff matrix surface

BACKGROUND: Dental pulp stem cells can differentiate into dentin under appropriate induction conditions, which are important seed cells i n dental tissue engineering. However, the commonly used inducers are chemical agents, which are not available for in vivo application. Mesenchymal stem cells can differentiate with the material hardness, and the physical property-induced cell differentiation is little reported. OBJECTIVE: To observe the extension characteristics and dentin differentiation potential of dental pulp stem cells from human deciduous teeth on the stiff matrix surface. METHODS: Dental pulp stem cells from naturally shed deciduous teeth were isolated, cultured and identified. Four solid gel matrixes with elasticity modulus of (9.12±0.94), (27.18±3.55), (59.37±4.05) and (86.45±5.33) kPa were made using low melting point agarose. The extension ability of passage 4 dental pulp stem cells on the surface of the above solid matrixes was detected by two-dimensional clone formation and cell scratch tests. The protein expression levels of dentin matrix protein-1, dentin phosphoprotein and dentin sialoprotein were detected by western blot assay. RESULTS AND CONCLUSION: Dental pulp stem cells from human deciduous teeth seeded on the gel matrix with extremely low and low hardness almost existed as cell clones with neat edges, and cell spreading and extension were rare. When seeded on the gel matrix with moderate and high hardness, the cloned edge of deciduous dental pulp stem cells spread and extended obviously. The cell body became large and the cell edge extended significantly. The cell scratch test revealed the similar phenomenon. When seeded on the gel matrix with moderate and high hardness, dental pulp stem cells from human deciduous teeth exhibited high expression levels of of dentin matrix protein-1, dentin phosphoprotein and dentin sialoprotein. In summary, with the increase of matrix hardness, the abilities of extension and differentiation into dentin of dental pulp stem cells from human deciduous teeth are increased gradually, which provides a method for dental tissue engineering.
4.Survey on the knowledge and willingness of genetic counseling and testing in blood relatives of breast cancer patients

Objective To investigate the knowledge and willingness of genetic counseling and testing in blood relatives of breast cancer patients.Methods A total of 922 blood relatives of breast cancer patients finished our questionnaire.Data were devided into different groups according to age,family history of tumor for statistical analysis.Results Most of the respondents were unaware of genetic counseling and genetic testing.However,after a brief introduction,major of them were willing to accept genetic counseling,breast cancer risk evaluation and screening.Specifically,79.8% of them were willing to accept genetic counseling,and 62.3% were willing to accept genetic testing.Most of the respondents would accept inexpensive early genetic screening.For the genetic testing with higher prices,only 37.9% of them would accept it.Supposing a positive genetic testing result,most of them were willing to perform prevention through close follow-upscreening,31.3% of them would choose prophylactic surgery or drugs.Despite being told the confidentiality of the test results,32.9% of them worried about the adverse effects of genetic test.Conclusions Most of the blood relatives of breast cancer patients were unaware of counseling and genetic testing,but had apparent willingness to accept them.Misunderstanding of genetic characteristics,costs and concerning about discrimination are obstacles for the respondents to accept genetic counseling,genetic testing and related screening prevention.
5.Association between cardiorespiratory fitness and muscle strength in maintenance hemodialysis patients

Objective:To investigate the association between cardiorespiratory fitness (CRF) and muscle strength, and the influencing factors of CRF in maintenance hemodialysis (MHD) patients.Methods:It was a single-center cross-sectional study. Patients who were treated with regular MHD from September 1, 2020 to December 31, 2020 were recruited. Baseline data of MHD patients including body circumference (upper arm circumference of non-fistula, triceps skinfold thickness, and calf circumference), exercise capacity [6-min walking test (6MWT), timed up and go test (TUGT)] and upper and lower limb muscle strength (grip strength, knee extension strength) as well as clinical and biochemical data, Charlson comorbidity index were collected or measured. Cardiopulmonary exercise test (CPET) was used to test peak oxygen consumption per kg body weight (peakVO 2/kg) for reflecting CRF. Pearson correlation or Spearman correlation was used to analyze the correlation between peakVO 2/kg and clinical parameters. Multiple linear regression analysis (stepwise) was performed to analyze the influencing factors of peakVO 2/kg. Results:A total of 48 patients were enrolled in the study, of whom 34 were males (70.8%). The age was (60.31±10.44) years old. The median dialysis duration was 48(15, 84) months, and the peakVO 2/kg was (12.37±2.71) ml·kg -1·min -1, which was well below the normal value of 30-50 ml·kg -1·min -1. Furthermore, peakVO 2/kg was positively correlated with lower extremity muscle strength ( r=0.322, P=0.026) and 6MWT ( r=0.307, P=0.034), and negatively correlated with age ( r=-0.300, P=0.038). Multiple linear regression analysis demonstrated that lower extremity muscle strength ( β=0.241, P=0.009) and age ( β=-0.235, P=0.022) were the influencing factor of peakVO 2/kg. Conclusions:CRF of MHD patients is significantly decreased. The decrease of lower extremity muscle strength is an influencing factor of CRF reduction.
6.Comparison of mesenchymal stem cells from human placenta and bone marrow.

BACKGROUNDNowadays bone marrow represents the main source of mesenchymal stem cells (MSCs). We identified a new population of MSCs derived from human placenta and compared its biological characterization with bone marrow derived MSCs.
METHODSMononucleated cells (MNC) were isolated from the human placenta tissue perfusate by density gradient fractionation. Individual colonies were selected and cultured in tissue dishes. At the same time, human bone marrow derived MSCs were identified. Culture-expanded cells were characterized by immune phenotyping and cultured under conditions promoting differentiation to osteoblasts or adipocytes. The hematopoietic cytokines were assayed using reverse transcriptase polymerase chain reaction (RT-PCR).
RESULTSHuman placental MSCs exhibited fibroblastoid morphology. Flow cytometric analyses showed that the placental MSC were CD29, CD44, CD73, CD105, CD166, HLA-ABC positive; but were negative for CD34, CD45, and HLA-DR. Functionally, they could be induced into adipocytes or osteocytes. Moreover, several hematopoietic cytokine mRNA was found in placenta-derived MSCs by RT-PCR analysis, including IL-6, M-CSF, Flt3-ligand and SCF. These results were consistent with the properties of bone marrow derived MSCs.
CONCLUSIONThese observations implicate the postpartum human placenta as an important and novel source of multipotent stem cells that could potentially be used for investigating mesenchymal differentiation and regulation of hematopoiesis.

7.Engineering a folic acid-decorated ultrasmall gemcitabine nanocarrier for breast cancer therapy: Dual targeting of tumor cells and tumor-associated macrophages.

Combination of passive targeting with active targeting is a promising approach to improve the therapeutic efficacy of nanotherapy. However, most reported polymeric systems have sizes above 100 nm, which limits effective extravasation into tumors that are poorly vascularized and have dense stroma. This will, in turn, limit the overall effectiveness of the subsequent uptake by tumor cells via active targeting. In this study, we combined the passive targeting via ultra-small-sized gemcitabine (GEM)-based nanoparticles (NPs) with the active targeting provided by folic acid (FA) conjugation for enhanced dual targeted delivery to tumor cells and tumor-associated macrophages (TAMs). We developed an FA-modified prodrug carrier based on GEM (PGEM) to load doxorubicin (DOX), for co-delivery of GEM and DOX to tumors. The co-delivery system showed small particle size of ∼10 nm in diameter. The ligand-free and FA-targeted micelles showed comparable drug loading efficiency and a sustained DOX release profile. The FA-conjugated micelles effectively increased DOX uptake in cultured KB cancer cells that express a high level of folate receptor (FR), but no obvious increase was observed in 4T1.2 breast cancer cells that have a low-level expression of FR. Interestingly, in vivo, systemic delivery of FA-PGEM/DOX led to enhanced accumulation of the NPs in tumor and drastic reduction of tumor growth in a murine 4T1.2 breast cancer model. Mechanistic study showed that 4T1.2 tumor grown in mice expressed a significantly higher level of FOLR2, which was selectively expressed on TAMs. Thus, targeting of TAM may also contribute to the improved in vivo targeted delivery and therapeutic efficacy.
