1.The relationship of the difference of sol precursor protein expression and chemotherapy drug resistance in breast cancer

Objective To detect the change of precursor protein of sol in breast cancer before and after the neoadjuvant chemotherapy,so as to find the serum protein marker of chemotherapy drug resistance in breast cancer.Methods Breast cancer patients with Ⅲ period neoadjuvant chemotherapy were enrolled in the study.The serum specimens of the patients were obtained.Two-way gel electrophoresis was used to get the gel map before and after chemotherapy,then the image map was analyzed by gel analysis software.The differentially expressed proteins point was screened before and after chemotherapy.Serum protein differences were identified with matrix auxiliary laser disi ntegrate series time of flight mass spectrometer (MALDI-TOF-MS),including coagulation sol precursor protein.Western blot was used to test the variation trend of them before and after chemotherapy.Results Coagulation sol precursor protein levels fell after neoadjuvant chemotherapy in resistance group.The variation trend of the results of western blot and that of the proteomics were consistent.Conclusion Coagulation sol precursor protein can be as the candidate serum marker of chemotherapy drug resistance in breast cancer.
2.Consistency study of PowerPlex 21 kit and Goldeneye 20A kit and forensic application.

To ensure the consistency of genotype results for PowerPlex 21 kit and Goldeneye 20A kit.
METHODS:
The STR loci were amplified in DNA samples from 205 unrelated individuals in Beijing Han population. And consistency of 19 overlap STR loci typing were observed. The genetic polymorphism of D1S1656 locus was obtained.
RESULTS:
All 19 overlap loci typing showed consistent. The proportion of peak height of heterozygous loci in two kits showed no statistical difference (P > 0.05). The observed heterozygosis of D1S1656 was 0.878. The discrimination power was 0.949. The excluding probability of paternity of triplet was 0.751. The excluding probability of paternity of diploid was 0.506. The polymorphism information content was 0.810.
CONCLUSION
PowerPlex 21 kit and Goldeneye 20A kit present a good consistency. The primer design is reasonable. The polymorphism of D1S1656 is good. The two kits can be used for human genetic analysis, paternity test, and individual identification in forensic practice.

3.Dual-phase18F-FDG coincidence detection SPECT/CT imaging for differential diagnosis of pulmonary lesions

Background and purpose:Although FDG tumor imaging has been applied in clinic widely, dual-phase imaging can provide much more information about the FDG uptaking of pulmonary lesions. The purpose of the study was to evaluate the usefulness of dual-phase18F-FDG coincidence detection SPECT/CT imaging in the differential diagnosis of the pulmonary lesions.Methods:There were 28 patients with pulmonary lesions which were detected by CT. All the patients undertook the SPECT/CT imaging at 2 time-phases respectively: early imaging at 40-60 min and delayed imaging at 2-3 h after the intravenous injection of FDG. Data processing: calculating the radio of T and N in early and delayed imaging respectively; T: The radioactive count of the lesions; N: The radioactive count of the normal tissue; and the change rate:ΔT/N. ROC was used to ifnd out the threshold of T1/N1, T2/N2及ΔT/N in the differential diagnosis between benign and malignant lesions. AUC was used to evaluate the diagnosis value of the dual-phase and single-phase imaging.Results:The threshold of T1/N1 in early imaging was 2.65, whereas AUC was 0.767. The sensitivity, speciifcity and accuracy were 83.3%, 30% and 64.3%, respectively. The threshold of T2/N2 in delayed imaging was 3.14, whereas AUC was 0.847. The sensitivity, speciifcity and accuracy were 94.4%, 60.0% and 82.1%, respectively. The threshold ofΔT/N in delayed imaging was 16.9%, whereas AUC is 0.950. The sensitivity, speciifcity and accuracy were 88.5%, 71.4% and 86.2%, respectively.Conclusion:Dual-phase18F-FDG coincidence detection SPECT/CT imaging has much higher accuracy and speciifcity. However it still has false positivity, and should be analyzed with CT and clinical history.
4.An Investigation on Nursing Demands of Home Healthcare among Elderly in Yunnan Province

Objective To establish programs of home healthcare service by investigation and analyzing the current status of the elderly requirements towards community nursing in yunnan province.Methods A self-administrated questionnaire which includes functional status,living situations,and home healthcare demands was distributed in 105 community elderly around Yunnan Province by professional staff. Results 76.20% of the community elderly people had different home care nursing service demands. The highest needs for community elderly is daily medical and nursing care.Conclusion Developing home healthcare can effectively improve the self- care consciousness of aged people, and also improving the quality of life in elderly has important significance for healthcare insurance of elderly.
5.Sorting of side population cells from breast cancer MCF-7 cell line and its biological characteristics

Objective To separate the side population cells(SP) from breast cancer MCF-7 cell line,and observe its biological characteristics.Methods Flow cytometry and Hcechst 33342 dye efflux assay were used to isolate SP cells and non-SP cells from the MCF-7 cell line of human breast cancer.Tumorigenicity of the two subpopulations was observed by a soft agar cloning method.Results The results of FACS analysis indicated that (6.5 ± 0.4 ) %of the MCF-7 cells were SP cells;The vitro colony formation rate of SP cells was(38.5 ±9.4)%,and higher than that of non-SP cells ( 8.4 ± 2.6 ) % ( t =5.34,P < 0,05 ).Concluslon The SP cells sorted from MCF-7 cell line enriched tunor stem cells,which exhibited high tumorigenicity.It indicated that SP cells should play a principal role in breast cancer.
6.Significance of brain stem auditory evoked potential and somatosensory evoked potential on prognosis evaluation in patients with acute cerebral infarction

Objective To investigate changes of brainstem auditory evoked potential (BAEP)and somatosensory evoked potential (SEP) in patients with acute cerebral infarction,and discuss their relation with prognosis of the patients.Methods The study involved 60 patients with acute cerebral infarction.Changes of BAEP and SEP in each patient were detected and recorded continuously.Prognosis evaluation was performed by using GCS.Another 60 age-matched and gender-matched healthy human beings were enrolled as controls.Results Incubation period of BAEP wave Ⅰ had no significant difference between the cerebral infarction and control groups (P > 0.05).However,interspike intervals of other BAEP waves in cerebral infarction group were different from those in control group (P < 0.05).A series of waves of SEP (P14-N60) were all significantly prolonged in cerebral infarction group (P <0.05).Conclusion BAEP and SEP can effectively reflect function of brain stem in patients with acute cerebral infarction and have some values in determining their prognosis.
7.Evaluation of computerized image analysis system for quantification of posterior capsular opacification

AIM: To set up and evaluate a computerized image scoring system for quantification of posterior capsular opacification (PCO) that is independent of examinee.METHODS: The PCO model was made in rabbits. Following dilation of the pupil (>6.5mm), standardized digital images of the PCO were obtained using slit-lamp photographic system(digital camera, computer included). PCO was scored by evaluating retroillumination photograghs. The PCO score was calculated by the formula: PCO score=∑ (OD×AI in CR)×2 +∑(OD×AI in OR), in which OD is opacity density (class 0 to 4),AI is the opacity area fraction, CR is the central region (diameter=3mm), OR is the outer region (3-6mm ring region).To evaluate the reliability of the scoring system, 6 examiners scored the pictures of 5 same eyes (interindividual reliability),and one examiner scored the pictures of 5 eyes on five different days (intraindividual reliability).RESULTS: The PCO scoring system was very reliable. With PCO score range from 0 to 1.5, the interindividual reliability showed standard deviation between 0.05-0.087. Kruskal-Wallis analysis showed no significant difference (H=0.314,P=0.9726). The intraindividual reliability showed standard deviation between 0.041-0.067, no significant difference either (H =0.613, P =0.9616).CONCLUSION: This PCO quantification analysis system evaluates the central area (diameter=6mm) of posterior capsule which is larger than visual acuity or other objective testing. The system revealed high reliability and insignificant investigator-dependent difference. With a standardized photographic setup, systematic errors by the photographic technique were not relevant. This system was proved to be an easy and useful tool to accurately evaluate PCO.
8.Programmed death ligand 1 negatively regulates inflammatory response of chronic periodontitis.

OBJECTIVETo investigate whether programmed death ligand 1 (PD-L1) expressed in the periodontal tissue of chronic periodontitis and the correlativity of PD-L1 and different degrees of chronic periodontitis, provide experience for immunoregulation mechanism, clinical treatment and prognosis of chronic periodontitis.
METHODSGingiva and periodontal tissue of healthy people and chronic periodontitis patients were collected. Based on clinical probing, periodontal tissue were classified into three groups: periodontal tissues of healthy people, periodontal tissue of mild chronic periodontitis, periodontal tissue of severe chronic periodontitis. Fluorescent quantitation polymerase chain reaction was applied to explore the expression of PD-L1 mRNA in the periodontal tissue of the different groups. Western blot and immunohistochemistry method were utilized to test the expression of PD-L1 protein in the periodontal tissue of the different groups. Combining with clinical image data, the relationship between differentially expressions of PD-L1 and different degrees of chronic periodontitis was analyzed.
RESULTSThe relative expression quantity of PD-L1 in the periodontal tissue of the mild chronic periodontitis was significantly higher that of the severe chronic periodontitis (P<0.01). The relative expression quantity of PD-L1 in the periodontal tissue of healthy subjects and severe chronic periodontitis had no statistical significance (P>0.05).
CONCLUSIONThe expression of PD-L1 in the periodontal tissue negativelv regulates inflammatory periodontal tissue damage.

9.Experimental study on effects of herbs for nourishing and smoothing the liver in reversing bile lithogenicity of guinea pig

OBJECTIVE: To further probe the mechanisms of herbs for nourishing and smoothing the liver in reversing bile lithogenicity of guinea pig. METHODS: Sixty guinea pigs were divided randomly into control group (fed with normal diet, n=20), model group (fed with lithogenic diet, n=20) and treatment group (fed with lithogenic diet plus herbal medicine, n=20). After four-week feeding, the animals were sacrificed and sampled, the rates of gallstone formation in each group were estimated, and the total bile acid (TBA), total bilirubin (TBIL), conjugated bilirubin (CB), unconjugated bilirubin (UCB), and calcium ion in the bile were determined, and the different bilirubins were analyzed by HPLC. RESULTS: (1) The rate of gallstone formation was 5% in normal group, 81.25% in model group and 31.25% in treatment group (P<0.05). (2) The bile TBIL, CB, UCB and Ca(2+) were higher and the bile TBA was lower significantly in model group than that in the other two groups (P<0.05). (3) HPLC analysis revealed that MCB was higher and DCB was lower significantly in model group (P<0.01), and there were no significant differences of UCB and IPA among the three groups. (4) The percentages of MCB and UCB were much higher and the percentage of DCB was remarkably lower in model group (P<0.05). CONCLUSION: Herbs for nourishing and smoothing the liver can significantly reduce the rate of gallstone formation and has effect of reversing lithogenicity of bile in guinea pigs fed with lithogenic diet.
10.Role of prolyl hydroxylases 2 in the cellular response to hypoxia activated autophagy in human renal epithelial cell model

Objective To test the hypothesis of autophagy that silencing PHD2 gene could increase hypoxia inducible factor (HIF)-1α levels in the renal medulla and attenuate hypoxia injury in cultured human renal proximal tubular epithelial cell (HK-2) under cobalt dichloride (CoCl2) exposure.Methods HK-2 cells were harvested at hour 0,6,12,24,36 and 48 after exposure to CoC12 (200 μmol/L).The role of HIF/PHD pathway in CoCl2-induced cell apoptosis/autophagy was studied by employing small-interfering RNA (siRNA).Dynamic profiles of apoptosis markers (Bax,Bcl-xl) and autophagy marker (LC3) of HK-2 cells within 48 h after exposing to CoCl2 were recorded.Alamar Blue assay was used for quantitative analysis of cellular growth and viability.Electron microscopy analysis was employed to evaluate the changes in autophagic structures.Results The protein expressions of PHD2 were gradually increased after exposing to CoCl2 (200 μmol/L),with statistics significance at 24 h and reached the peak at 48 h (both P < 0.01).PHD2 siRNA reduced PHD2 levels by > 60% and significantly increased HIF-1α protein levels (P < 0.01),but had little effect on HIF-2α.The protein expression of Bcl-xl was significantly up-regulated,while the level of Bax and LC3-Ⅱ/LC3-Ⅰ were down-regulated in PHD2 siRNA group (all P < 0.01),compared with the negative control group.Meanwhile,either 3-Methyladenine (an autophagy inhibitor) treatment or PHD2 knockdown rescued cell death and increased cell viability through autophagy inactivation.The ratio of LC3-Ⅱ/LC3-Ⅰ and the quantity of autophagosomes were decreased,and the cell ultrastructure was also relatively intacter than the negative control group.Of interest,co-administration of HIF-1α siRNA with PHD2 siRNA abrogated renoprotective effect conveyed by PHD2 siRNA alone,suggesting that activation of endogenous HIF-1α-dependent pathways mediated the autophagy inactivation effects of PHD2 silencing.Conclusions Direct inhibition of PHD2 promotes renal epithelia cell survival against CoCl2-induced cell apoptosis/autophagy.Activation of the HIF-1α signaling pathway is required to reduce apoptosis and autophagy via up-regulating the expression of Bcl-xl protein.
