Objective To investigate the feasibility and clinical effect of one-stage total knee arthroplasty for old femoral condyle fractures combined with knee osteoarthritis. Methods From January 2003 to December 2010,11 patients with old femoral condyle fracture combined with knee osteoarthritis, including 4males and 7 females, aged from 38 to 77 years (average,58.3 years),underwent one-stage total knee arthroplasty. There were 8 cases of supracondylar fracture, and 3 cases of intercondylar fracture. Moreover, there were 2 cases of knee fibrous ankylosis with mild amyotrophy,3 cases of collateral ligaments injury of the knee,2 cases of fibrous union, and 2 cases of infective nonunion. Results All patients were followed up for 6 to 72 months (average,41.3 months). The HSS score improved from preoperative 6.65±7.01 (range,0-13)to 88.5±6.18 (range,81-95) at final follow-up.The range of motion of the knee improved from preoperative 32.7°±17.6° (range,0°-50°) to 101.3°±9.8° (range,85°- 115°) at final follow-up.One case underwent knee disarticulation 6 years after operation owing to infection. For other patients, there were no complications such as infection, deep vein thrombosis, knee instability, component loosening and inequality of lower limb. Conclusion One-stage total knee arthroplasty is a technically difficult but effective method for patients with old femoral condyle fracture and knee osteoarthritis.

Retinopathy of prematurity (ROP) is a proliferative vascular retinal disease. Cryotherapy, laser photocoagulation, intravitreal injection of anti-vascular endothelial growth factor, scleral buckling surgery and vitrectomy are the main treatments. Treated with cryotherapy or laser photocoagulation or intravitreal injection of anti-vascular endothelial growth factor, patients with a history of ROP have thicker foveas, and the morphology of the fovea and the development of the retinal vessels in the macular area are affected, resulting in abnormal vision development. However, the specific mechanisms by which different treatments of ROP affecting the development of the macula are not yet clear. It still need further study with large samples to verify and explore, whether changes in the levels of intraocular vascular endothelial growth factor changing the process of normal macular development and how the abnormal development of the macula affects visual function.

Objective To construct markless gene deletion mutant at the clpP loci on the chromosome of Streptococcus mutans(S.mutans).Methods ASp resistance gene was amplified by PCR,to construct the Sp resistance cassette where the Sp resistance gene was flanked with two loxP site. After the clpP gene was cloned into the pGEM-T-Easy TA cloning vector,it was digested and linked with the Sp resistance cassette, yielding homologous recombination vector pIB △ clpP-Sp.The vector was linearized and used for the transformation of S.mutans UA159,with transformants selected on TPY plates containing Sp.The selected strain was transformed with the thermosensitive plasmid pCrePA to excise the Sp resistance gene. The pCre-PA was then easily eliminated at nonpermissive temperature, resulting in a markless mutant strain carrying a deletion at the clpP loci, which was verified by PCR and DNA sequencing. Results The result of the PCR analysis and DNA sequencing indicated that a part of the clpP gene was deleted. There was a loxP at this loci without the Sp resistance gene. Conclusion The markless clpP-deletion mutant of S.mutans was constructed successfully, which laid a foundation for further study of its biological function and its influence on the cariogenicity of S.mutans.

Shanghai district and county health system performance evaluation conceptual frameworkwas proposed based on health services system performance assessment frameworks raised by some countries and international organizations, and combined with Shanghai district and county health system characteristics. This framework contains two first grade indices and seven second grade indices:In the long term goals,'health','satisfaction' and 'disease risk protection' are the county's longterm effects of health system performance assessment dimensions; in the medium-term goals, the'accessibility','cost',' efficiency' and 'quality' are the county' s medium-term effects of health system performance assessment dimensions. This study aimed to evaluate Shanghai county health system's overall performance.Further study is needed in constructing its third grade indices.

Objective · To investigate the correlation between proportion of natural killer (NK) cells and dendritic cells (DCs) and liver injury in patients with chronic hepatitis B virus (HBV) infection. Methods · Peripheral blood samples were collected from 65 patients with chronic HBV infection and 10 healthy volunteers (HCs group). Flow cytometry was used to detect the proportion of peripheral NK cells and DCs. The patient were divided into asymptomatic HBV carrier (AsCs) group, chronic hepatitis B in low or moderate grade (CHB-LM) group, and chronic severe hepatitis B (CSHB)group. The relationship of the proportion of NK cells, the proportion of DCs, and the ratio of NK/DC with the degree of liver injury was analyzed. Results · The proportion of peripheral NK cells was significantly lower in CHB-LM group (9.29%±1.20%) and CSHB group (3.60%±0.79%) than that in HCs group (14.77%±2.90%) and AsCs group (14.05%±2.32%) (P=0.000). And the proportion of peripheral DCs was significantly higher in CHB-LM group (4.10%±1.18%) than that in HCs (1.67%±0.31%), AsCs (2.05%±0.46%) and CSHB groups (2.08%±0.41%) (P=0.000). The ratio of NK/DC was decreased in CSHB and CHB-LM groups compared to that in HCs and AsCs groups (P=0.000). The correlation analysis indicated the proportion of NK cells was negatively correlated with the levels of ALT, AST, TB and DB (r=-0.50, r=-0.49, r=-0.53, r=-0.51, P=0.000 for all) in patients with chronic HBV infection. Conclusion · The proportion of peripheral NK cells and the ratio of NK/DC in patients with chronic hepatitis B were decreased. The decrease of the frequency of NK cells and the ratio of NK/DC were associated with liver injury in patients with chronic HBV infection.

Acta Nutrimenta Sinica 1956;0(02):-
Objective: To observe the effects of carotenoids extracts from Potamogoton crispus L. (CEPC) on proliferation and apoptosis of Hela cell line. Method: The cell growth inhibition was tested in MTT assay and cell cycle arrest was measured by flow cytometry (FCM). Apoptotic cells were observed by fluorescent microscope and Laser scanning confocal microscopy (LSCM). LSCM was also used to determine the Ca2+ concentration in cells. Results: CEPC had strong inhibition effects on the cell growth. The cell cycle progression was arrested at G2/M phase. The typical characteristics of apoptotic cells such as chromatin condensation were observed by the fluorescent microscope and LSCM. The concentration of intracellular Ca2+ was remarkably increased after treatment of CEPC as compared with the control. Conclusion: Apoptosis of Hela cell line is the important anticancer mechanism of CEPC and the increase of intracellular Ca2+ level might participate in the process.

Objective:Analyze the influence of hematopoietic stem cell mobilization on circulating endothelial cells (CECs) and circulating endothelial precursors (CEPs).Methods:CECs and CEPs were enumerated in 68 tumor patients and 15 healthy controls by 3-color flow cytometry. 11 cases underwent PBSC (peripheral blood stem cell) mobilization by combination chemotherapy and G-CSF (granulocyte colony-stimulating factor). Results:CECs and CEPs in tumor patients were 0.378%?0.103% and 0.059%?0.013% respectively, which were higher than that of normal controls.CECs/CEPs in peripheral blood (PB) were increased after G-CSF mobilization. Conclusion:Hematopoietic and endothelial progenitors can be mobilized into the PB by treatment with growth factor G-CSF.
