1.Electrophysiological Properties of Frequency Domain in Rats' Skeletal Muscle

Objective To study the electrophysiological properties of rat skeletal muscle within the frequency from 100Hz to 100MHz.Methods The AC impedance of rat sural muscle tissues was measured using the Agilent 4294A impedance analyzer, then the electrophysiological properties of the normal rat sural muscle cells were established in the respect of cell dielectric spectroscopy, Cole-Cole plots, spectrum of loss factor and loss tangent.Results (1) The dielectric response of rat sural muscle cells was frequency-dependent, such as increase in frequency caused decrease in permittivity. At the same time the conductivity trended to arise; (2) The dielectric parameters(?_L,?_h,?_L,?_h,△?″_ max,△tg?_ max,f_ C1,f_ C2) cell fiber parallel to the applied electric field differed from that cell fiber perpendicular to the applied electric field; (3) The effect of alternating current electric field on rat sural muscle cells showed a dielectric dispersion phenomenon, representing two characteristic frequency: the 1 st frequency(f_ C1(∥)=f_ C1(⊥)=1.02 kHz) and 2 nd frequency(f_ C2(∥)=59.31 kHz,f_ C2(⊥)=380.04kHz). Conclusion The method for studying the electrophysiological properties of rat skeletal muscle cells was established in the respect of the frequency domain,providing a possibility for investigating muscle fatigue in the future.
2.Role of TRAIL in the treatment of prostate cancer: An update.

Tumor necrosis factor (TNF)-related apoptosis-inducing ligand (TRAIL) is a member of the TNF super family found in recent years, which widely exists in the body tissues and participates in the immune regulation, immune stability, and immune surveillance of the human body. The TRAIL receptor is expressed in the surface of a variety of cells. Recent studies show that TRAIL induces the apoptosis of tumor cells and has no significant toxic effect on normal cells. Its anti-tumor activity and safety have been widely recognized. The development of prostate cancer is regulated by the mechanisms of cell apoptosis. TRAIL can induce the apoptosis of prostate cancer cells, and therefore has a great application value in the treatment of prostate cancer.

3.Prognosis and its influencing factors of post-encephalitic epilepsy

Objective To investigate the prognosis and its influencing factors of post-encephalitic epilepsy ( PEE) .Methods Fifty viral encephalitis patients with PEE were followed up, which prognosis was observed by whether their no seizures for at least 1 year or not.The clinical data of patients was collected, and the relate risk factors on the poor prognosis of PEE were analyzed.Results The disturbance of consciousness and the seizure presence during acute encephalitis, more than ten seizures in poor prognosis group were much higner than the good one (all P<0.01).The age, sex, seizure type, dysphrenia and neurological defect in acute encephalitis, status epileptisus in acute encephalitis, abnormal EEG and abnormal head CT/MRI had no correlation with poor prognosis ( all P>0.05 ) .Multiple logistic regression analysis demonstrated that disturbance of consciousness during acute encephalitis、seizures presence in acute encephalitis were independent risk factors of poor outcome in PEE ( OR=7.269,95%CI:1.22 -43.35,P=0.029;OR =22.893,95%CI:4.02 -130.43,P=0.000).Conclusion Disturbance of consciousness during acute encephalitis, seizure presence in acute encephalitis both are the single risk factors of poor prognosis in PEE.
4.Effects of Tail Suspension on Dielectric Properties of Cells in Isolated Rat Sural Muscle

Objective To study changes of dielectric properties of gastrocnemius caused by simulated weightlessness.Methods Male Sprague-Dawley rats tail suspended for 10 wk were observed together with its normal control.AC impedance of isolated gastrocnemius was measured with an impedance analyzer(Agilent 4294A).Effects of simulated weightlessness on electric properties of cells in isolated rat sural muscle were also observed by analyzing the dielectric numerical characters of the dielectric spectra,the Cole-Cole plots,spectrum of loss factor and loss tangent,as well as spectrum of conductivity imaginary part.Results It was found that simulated weightlessness could induce certain changes:both relative permittivity at low frequency ?L and relative permittivity increment ??,as well as conductivity at high frequency ?h and conductivity increment ?? were all reduced.The first characteristic frequency fC1 decreased,while the second characteristic frequency fC2 increased.Peak of loss factor,peak of loss tangent and the maximum of conductivity imaginary part were also reduced.Conclusion Changes of dielectric property of gastrocnemius induced by simulated weightlessness can be observed effectively by frequency domain impedance technique.
5.A primary study of evolution of hepatitis B virus based on motif discovery.

Hepatitis B is a serious infectious disease worldwide, and hepatitis B virus (HBV) is the direct cause of this disease. In recent years, as an essential part of its evolutionary process, HBV mutation has been extensively studied domestically and globally. However, the study on the conserved sequences in HBV sequences is still in its infancy. In this study, we applied multiple EM for motif elicitation (MEME) algorithm to discover HBV motif and proposed a new metric, conservative index (CI), to carry out phylogenetic analysis based on HBV sequences. Then, the constructed phylogenetic tree was subjected to reliability assessment. The results demonstrated that the new metric CI combined with the MEME algorithm can effectively help to discover motifs in HBV sequences and construct a phylogenetic tree based on them and to analyze the evolutionary relationship between HBV sequences; in addition, the possible ancestral sequences of samples may be obtained by conservative analysis. The proposed method is valuable for the exploratory study on large HBV sequence data sets.

6.Effects of the synthetic peptide coatings inhibiting the growth of Streptococcus gordonii on titanium surface

Objective To detect the independently designed synthetic peptide adsorbed to the titanium surface and its inhibitory effect on streptococcus gordonii, and to provide a new means for antibiosis reseach on oral implants. Methods The physical and chemical properties of the synthetic peptide and antimicrobial peptide were measured by ExPASy Prot?Param tool, ProtScale analysis, circular dichroism and Zeta potential instrument. The synthetic peptide was anchored on the surface of the titanium specimen through incubation at room temperature. The adsorption of the synthetic peptide to the titani?um surface was examined by X-ray photoelectron spectroscopy (XPS) and the atomic force microscope (AFM). The inhibitory effect on streptococcus gordonii of the synthetic peptide fixed on the titanium surface was viewed by confocal laser scanning microscopy (CLSM). The destructive effects of the synthetic peptide and the antimicrobial peptide on streptococcus gordonii were observed through the transmission electron microscope (TEM). Results The independently designed synthetic peptide still had the physical and chemical properties that the antimicrobial peptide desired. The synthetic peptide had already been detected on the titanium surface after incubated in a 5 g/L synthetic peptide solution. The titanium specimen fixed with the synthetic peptide inhibited the survival and adhesion of streptococcus gordonii. Conclusion It suggests that the indepen?dently designed synthetic peptide might have reached the goal of bacterial inhibition on the titanium surface.
7.Experimental study on electrical impedance properties of human hepatoma cells.

The AC impedance of human hepatoma SMMC-7721 cells were measured in our laboratory by Agilent 4294A impedance analyzer in the frequency range of 0.01-100 MHz. And then the effect of hematocrit on electrical impedance characteristics of hepatoma cells was observed by electrical impedance spectroscopy, Bode diagram, Nyquist diagram and Nichols diagram. The results showed that firstly, there is a frequency dependence, i.e., the increment of real part and the imaginary part of complex electrical impedance (δZ', δZ"), the increment of the amplitude modulus of complex electrical impedance (δ[Z *]) and phase angle (δθ) were all changed with the increasing frequency. Secondly, it showed cell volume fraction (CVF) dependence, i. e. , the increment of low-frequency limit (δZ'0, δ[Z*] 0), peak (δZ"(p), δθ(p)), area and radius (Nyquist diagram, Nichols diagram) were all increased along with the electric field frequency. Thirdly, there was the presence of two characteristic frequencies: the first characteristic frequency (f(c1)) and the second characteristic frequency (f(c2)), which were originated respectively in the polarization effects of two interfaces that the cell membrane and extracellular fluid, cell membrane and cytoplasm. A conclusion can be drawn that the electrical impedance spectroscopy is able to be used to observe the electrical characteristics of human hepatoma cells, and therefore this method can be used to investigate the electrophysiological mechanisms of liver cancer cells, and provide research tools and observation parameters, and it also has important theoretical value and potential applications for screening anticancer drugs.

8.Intracoronary abciximab administration for patients undergoing percutaneous coronary intervention:a meta-analysis of randomized trials

Objective To systematically evaluate the efficacy and safety of intracoronary abciximab administration as compared to intravenous in percutaneous coronary intervention (PCI) .Methods A search was conducted in PubMed ,EMBASE ,OVID ,CBM , CNKI and VIP for the randomized controlled trials (RCTs)of intracoronary abciximab administration versus intravenous in PCI , from the date of their establishment to September 30 ,2013 ,and the domestic relevant papers published in recent 1 year were also searched manually ,the bibliographies of the included studies were searched too .According to the criteria of the cochrane Hand-book ,two reviewers evaluated the quality of the included RCTs and extracted data independently ,and then the extracted data were analyzed by using RevMan 5 .1 software .Results 8 RCTs involving 10 articles with 4 150 patients who treated with PCI were in-cluded .The results of meta-analysis showed that :(1) Compared intravenous administration and intracoronary abciximab administra-tion ,there were no significant differences in the two groups for the major adverse cardiovascular event (MACE) and the mortality (OR=0 .78 ,95% CI 0 .54-1 .14 ,P=0 .20) ,(OR=0 .56 ,95% CI 0 .24-1 .30 ,P=0 .18) .(2)Compared with intravenous adminis-tration ,intracoronary abciximab administration were not associated with any excess of major bleeding complications :(OR=1 .26 , 95% CI 0 .78-2 .02 ,P=0 .35) .Conclusion Compared with standard abciximab regimen of intravenous bolus ,intracoronary admin-istration had equivalent effects in clinical outcomes in patients undergoing PCI ,and did not increase the incidence of bleeding events .
9.Preoperative risk factors analysis of pulmonary hypertension crisis during perioperative period for caesarean section of woman with severe pulmonary hypertension

Objective To analyze preoperative risk factors of perioperative pulmonary hypertension crisis (PHC) for pregnant woman with severe pulmonary artery hypertension (PAH), and approach its clinical value. Methods A retrospective analysis was conducted. The clinical data from 152 pregnant women with severe PAH underwent cesarean delivery admitted to Beijing Anzhen Hospital from January 1st 2008 to December 31st 2016 was collected. The patients were divided into two groups according to with perioperative PHC or not. Through the case management system, age, height, weight, gestational age, pregnancy time, type of PAH, emergency or selective surgery, New York Heart Association (NYHA) cardiac function classification, and preoperative ultrasound left ventricular ejection fraction (LVEF), left ventricular diastolic final diameter (LVEDD), the pulmonary artery systolic pressure (sPAP) estimated by ultrasonic TI method, radial artery systolic blood pressure (SBP) and diastolic blood pressure (DBP), heart rate (HR), pulse oxygen saturation (SpO2) without oxygen, oral sildenafil ingestion, having Swan-Ganz catheter placement or not, and whether used norepinephrine or not, as well as the occurrence of perioperative PHC and clinical outcomes were collected. Possible preoperative risk factors were compared between the two groups by single factor and multiple factors logistic regression analysis. The receiver-operating characteristic curve (ROC) was plotted to assess the diagnostic value of various risk factors.Results A total of 152 patients were screened. Ten patients got heart surgery under general anesthesia at the same time, and 4 patients experiencing cesarean section with general anesthesia were excluded. 138 patients were enrolled finally, 27 patients underwent perioperative PHC (19.57%), and 17 patients died with a mortality of 62.96%. Compared with non-PHC group, the patients in PHC group were older (years: 25.07±3.55 vs. 27.64±4.82), had a poor cardiac function (NYHA cardiac function classification: 3.22±0.64 vs. 2.85±0.53), a smaller LVEDD (mm: 38.78±4.76 vs. 43.91±9.67), lower SpO2 without oxygen (0.83±0.12 vs. 0.92±0.06) and oral sildenafil ingestion rate (29.63% vs. 56.76%), and higher sPAP estimated by ultrasonic TI method [mmHg (1 mmHg = 0.133 kPa): 113.41±24.73 vs. 99.35±21.10] and DBP (mmHg: 79.63±13.23 vs. 75.23±12.14), more having Swan-Ganz catheter placement (85.19% vs. 57.66%), more Eisenmenger syndrome (70.37% vs. 37.84%), and more emergency operation (48.15% vs. 23.42%, allP ≤ 0.1). The variables with statistically significant differences showed by single factor analysis were collected, and it was shown by multiple factors logistic regression analysis that LVEDD [odds ratio (OR) = 0.878, 95% confidence interval (95%CI) = 0.796-0.968,P = 0.009], whether oral taken sildenafil (OR = 0.161, 95%CI = 0.051-0.515,P = 0.002) or not, SpO2 at room air (OR = 0.882, 95%CI = 0.829-0.938,P = 0.000), Swan-Ganz catheter placement or not (OR = 6.186, 95%CI = 1.533-24.964,P = 0.010) were independent risk factors of perioperative PHC in pregnant women with severe PAH. It was shown by ROC curve analysis that the area under the ROC curve (AUC) of four factors mentioned above combined diagnosis for PHC was 0.878 (P = 0.000) with the sensitivity of 88.89% and specificity of 76.58%.Conclusions PHC is very dangerous for gravida with severe PAH, and the mortality rate is very high. LVEDD, oral sildenafil, SpO2 at room air, Swan-Ganz catheter placement or not were independent risk factors of perioperative PHC for severe PAH maternal. Four preoperative factors of perioperative PHC joint diagnosis accuracy were higher.
10.Analysis of literature on therapeutic methods and medicines of traditional Chinese medicine for primary liver cancer

OBJECTIVE: To find out the rules of therapeutic methods, formulas and medicines of traditional Chinese medicine (TCM) for primary liver cancer (PLC). METHODS: The frequencies of treatments and medicines of TCM in 978 pieces of documents concerning PLC were analyzed. RESULTS: Among the 165 therapeutic methods, 277 formulas and 459 kinds of herbs, the methods for activating blood circulation, the formulas for regulating qi-flowing and the herbs for invigorating qi are the most commonly used. CONCLUSION: Therapeutic methods such as invigorating qi, regulating qi-flowing and activating blood circulation are the main choices for PLC, and the methods such as clearing heat, removing dampness and nourishing yin are used as supplements.
