2.Detection of β2-microglobulin and fibronection contents in patients with nasopharyngeal carcinoma and their clinical significance

Objective:To study the clinical significance of β2-microglobin(β2-MG)and fibronection (Fn) contents in patients with nasopharyngeal carcinoma.Methods:The contents of serum β2-MG and Fn were detected by radioimmunoassay and agar-diffusion methods in 60 patients with nasopharyngeal carcinoma before and after radiotherapy,also in 60 normal subjects as control group.Results:The contents of serum β2-MG and Fn were 2.99±1.11mg/L(β2-MG)and 134.60±28.93mg/L(Fn) in 60 patients with nasopharyngeal carcinoma,2.16±0.50mg/L(β2-MG)and 196.16±34.65mg/L(Fn)in 60 health subjects,respectively.Those results showed that the content of β2-MG in patients group was higher than that in control group (P<0.05),and the content of Fn in patients group was lower than that in control group(P<0.05).After radiotherapy of patients group,the content of β2-MG were lower(2.16±0.55mg/L)than that before radiotherapy(P<0.05).Conversely,the content of Fn was higher(180.53±34.66mg/L)than after radiotherapy(P<0.05).Conclusions:The detection of serum β2-MG and Fn have clinical significance on the evaluation prognosis of patients with nasopharyngeal carcinoma.
3.Rs-fMRI changes of posterior cingulate cortex in type 2 diabetes mellitus patients with cognitive im-pairment

Objective To investigate the resting?state functional alteration in posterior cingulate cortex ( PCC) in Type 2 diabetes ( T2DM) patients with cognitive impairment and to determine the relation?ship of rs?fMRI changes with cognitive decline. Methods Resting?state functional magnetic resonance ima?ging was performed T2DM patients with impaired cognition ( n=19) and healthy control subjects ( n=20) . The amplitude of low frequency fluctuations ( ALFF) and regional homogeneity ( ReHo) values were calcu?lated in left PCC to represent the spontaneous brain activity. Using left PCC as a seed region,the functional connectivity of the whole brain was mapped. In addition,correlation analysis was conducted among ALFF,Re?Ho,and neuropsychological test scores. Results The ALFF values of left PCC (0.72±0.37) was decreased compared with the control group(1.09±0.46),but the ReHo value(1.12±0.10)was not significantly changed compared with control group (1.14±0.11). The functional connectivity was decreased with the left medial temporal lobe,left superior and middle temporal gyrus,right superior frontal gyrus,and right supplementary motor area ( SMA) ,while it was increased with the left middle frontal gyrus,inferior frontal gyrus and right cerebella. MoCA scores were positively correlated with the ALFF values of left PCC. Conclusion In resting state,the spontaneous activity and functional connectivity of PCC is altered in T2DM patients with cognitive impairement;and the function of PCC contributes to the cognitive decline associated with T2DM.The decrease of ALFF value of PCC may help to detect the cognitive decline of T2DM.

6.Comparative study in two preservation methods of the sputum aspirator nozzle

Objective To explore a simple and safe method for preservation of the sputum aspirator nozzle. Methods Using self-controlled method, the traditional method was to place the nozzle in the glass bottle of disinfectant liquid, the modified method was to fix a sputum tube to a suitable location of the sputum aspirator or bedside with the joint free, after sputum aspiration,the nozzle was connected to the joint. One patient used the same sputum aspirator. The two methods of preservation were alternated every 24 hours. The slippage times of the nozzle were compared, and samples from glass nozzle and joints of disinfectant liquid bottles/spu-tuna tube joints were underwent bacterial culture. Results Significant difference existed in slippage times of the nozzle, the positive rate of bacteria colony in the.glass nozzle and disinfectant liquid bottles/sputum tube joints between the two methods. Conclusions Using sputum aspiration tubes to fix the nozzle of aputum aspi-rator is better than the traditional method of putting the nozzle into the glass bottle of disinfectant liquid,this method proves to be easy, safe,portable and economical and is worthy of clinical application.
7.Disease characteristics of obese patients with polycystic ovary syndrome described by the obesity component of the International Classification of Functioning, Disability and Health comprehensive core set

Objective To describe the disease characteristics of obese patients with polycystic ovary syndrome (PCOS) using the obesity core set of the obesity International Classification of Functioning,Disability and Health (ICF) to reveal the related dysfunction domains.Methods Sixty obese patients with PCOS were evaluated by using the ICF's obesity comprehensive core set.Categories reported as a problem by at least 30% of the patients were considered as a relevant functional profile for PCOS patients.Results In obese 60 patients with PCOS,20 items were selected from 109 items of obesity ICF comprehensive core set and considered as the relevant functional characteristics for PCOS among the obese,in which 6 items of body functions (30% of all selected items),1 item of body structures (5% of all selected items),and 13 items of environmental factors (65% of all selected items) were included.Conclusions The obesity core set of the ICF can be applied to describe the disease characteristics and dysfunction of obese patients with PCOS,which offers the possibility of clinic application of the ICF core set for PCOS.
8.Effects of Homocysteine on Neural Stem Cell Proliferation and Genes Related Notch Signaling in Vitro

Objective To explore the effects of homocysteine (Hcy) on neural stem cell (NSC) proliferation and the mRNA expression level of Notch1 and Hes1 related Notch signaling pathway from neonatal rats in vitro. Methods NSCs from neonatal rats were cultured by serum-free culture method in vitro. Cells were divided into four groups: control group (Hcy-C), low dose Hcy (Hcy-L) group, middle dose Hcy (Hcy-M) group and high dose Hcy (Hcy-H) group. NSCs were iden- tified by immunofluorescent staining using the antibodies against Nestin, β-tubulin Ⅲ and GFAP. The proliferation ability of NSCs was detected by MTT. The mRNA expressions of Notch1 and Hes1 were detected by Real-time PCR. Results In the serum free suspension medium, neurospheres that consisted of a great number of nestin-positive cells were found. β-tu- bulin Ⅲ positive neurons and GFAP positive astrocytes were detected by immunofluorescence staining on the 6 th day of cell induction. MTT assay showed that the cell viability was significantly lower in three Hcy treatment groups than those of con- trol group (P < 0.05). And the effect of concentration-dependent was observed. The results of RT-PCR showed that mRNA expression of Hes1 was significantly lower in three Hcy treatment groups than that in control group (P < 0.05). The mRNA ex- pression of Notch1 was significantly lower in Hcy-H group than that of other three groups (P < 0.05). The mRNA expression of Notch1 was significantly lower in Hcy-M group than that of Hcy-L group and control group (P < 0.05). Conclusion Hcy could inhibit the proliferation of NSCs by down-regulating mRNA expression levels of Notch1 and Hes1 genes related to Notch signal pathway.
9.Study on the injury mechanism of Ang-Ⅱ to mice podocyte in the high glucose environment

Objective To cultivate mice glomerular podocyte with high glucose and high Ang-Ⅱ,observe the morphocytology of podocyte,and investigate protein kinase C (PKC) activity,the expressions of nephrin and CD2-associated protein (CD2AP),and apoptosis of podocyte with injuries cultivations.Methods The conditions immortalization mice podocyte cell lines was cultivated,after passage and differential induce,the cell lines was divided into six groups:normal group,high glucose group,high Ang-Ⅱ] group,the above two mixed group,intervened group of PKC inhibitor,and hypertonic group.After 72 hours,cell apoptosis rate was detected with flow cytometer; PKC activity was detected with enzyme linked immunosorbent assay (ELISA) in 24 hours and 48 hours ; The expressions of nephrin and CD2AP mRNA were detected by real time-polymerase chain reaction (RT-PCR).Results (1)After 24 hours,the PKC of high glucose group and high Ang-Ⅱ group were more activated than normal group [(47.09 ± 1.19) pmol/L,(42.93 ±0.71) pmol/L,P <0.05].The activated phenomenon was more obvious in the mixed group.Compared to 24 hours [(58.75 ±0.71) pmol/ L,P < 0.01],PKC was more activated in 48 hours (P < 0.05).(2) Nephrin mRNA was reduced in podocyte exposed to high glucose and high Ang-Ⅱ at least 24 hours (56.87 ± 0.74,48.54 ± 0.86,P < 0.05),and was reduced more in mixed group (11.7 ± 1.54,P < 0.01).(3) CD2AP mRNA was reduced in podocyte exposed to high glucose,high Ang-Ⅱ,and mixed environment at least 48 hours (56.09 ± 1.46,67.68 ±2.58,and 54.08 ±2.74,P <0.05).The decrease trend of nephrin and CD2AP mRNA was restrained by PKC inhibitor(90.75 ± 1.33,P <0.05).(4) After 48 hours and 72 hours,the apoptosis rates of high glucose group,high Ang-Ⅱ group,and mixed group were risen compared to normal group (P < 0.05).The increase of apoptosis rate can be restrained by PKC inhibitor.Conclusions High glucose,and high Ang-Ⅱ can activate the podocytes PKC activity,inhibit the expression of nephrin and CD2AP mRNAs,and induce podocyte apoptosis.PKC signaling pathway plays an important role in the injury mechanism of Ang-Ⅱ and high glucose to mice podocyte.
10.The Reproduction of Nuclear Polyhedrosis Virus in the Midgut Epithelial Cell and the Other Sensitive Cell of Helicoverpa armigera

The reproduction of Helicoverpa armigera nucleopolyhedrovirus in the midgut epithelia cells and the other sensitive tissues was observed by electron microscopy. The reproducing viruses in the midgut epithelia cells were mostly without envelopes, and thte polyhedrons were seldom formed. The reproduciing viruses in the other sensitive cells were with envelopes, and packed in polyhedrons.
