1.Clinical study on Treatment of Active Ankylosing spondylitis with Total panax Notoginseng Saponins

Objective To evaluate the effectiveness and safety of treating active ankylosing spondylitis with total panax notoginseng saponins (PNS). Methods sixty-two patients were randomly recruited into a control group and a treatment group, with thirty-one patients in each group. All patients were treated with the routine therapy of NSAIDs and DMARDs. The treatment group was given PNS additionally, with 400mg PNS joined with 200ml normal saline being intravenously dripped daily. The therapeutic course was 14 days for both groups. The observation items for evaluation included symptoms, signs, and side effects. Results Compared with previous state before treatment, the lumbosacral portion pain and the count of joint pain was reduce, the time of morning stiffness was shortening, erythrocyte sedimentation rate (ESR), and C-reaction protein (CRP) were all significantly reduced (P<0.05) ; There were significant differences in the total effectiveness rates between the two groups (P<0.05) ; while no notable difference was found in adverse drug reaction between the groups (P>0.05). Conclusion It is effective that treating active ankylosing spondylitis with PNS plus NSAIDs.
2.Rosiglitazone Inhibitory Effect on Mesangial Cell Proliferation and Extracellular Matrix Expression Induced by Angiotensin Ⅱ

ObjectiveTo investigate the inhibitory effect of peroxisome proliferator-activated receptor-?(PPAR?) agonist on mesangial cell(MC) proliferation and extracellular matrix expression induced by angiotensin Ⅱ(Ang Ⅱ).MethodsThe incorporation of 3H-thymidine(3H-TdR) and cell count were used as the measurement of MC proliferation.MC cell-cycle was analyzed by flow cytometry.Mouse primary MC was treated with various concentration of Ang Ⅱ(1,10,100 nmol/L) in the presence or Absence of N-acytosistin(NAC) or rosiglitazone.Transforming growth factor-?1(TGF-?1),plasminogen activator inhibitor-1(PAI-1),and fibronectin(FN) mRNA expression were determined by real time-PCR.Reactive oxygen species(ROS) production was measured by 2,7-dichlorofluorescein diacetate(DCFDA) fluorescence.Results1.One hundred nmol/L Ang Ⅱ increased 3H-TdR incorporation and cell number by 2.14 and 2.32 fold,respectively.Ang Ⅱ-induced MC proliferation was inhibited by PPAR? agonist rosiglitazone with dose-dependent manner in mouse MC.2.One hundred nmol/L Ang Ⅱ stimulation for 24 h induced 48% MC processed to S and G2/M phase.Rosiglitazone significantly blocked Ang Ⅱ increased cell number in S and G2/M phase.3.Rosiglitazone reduced Ang Ⅱ-induced TGF-?1,PAI-1,and FN mRNA expression with dose-dependent manner.4.One hundred nmol/L Ang Ⅱ stimulation for 60 min increased ROS production by 3.85 folds.Rosiglitazone significantly inhibited Ang Ⅱ-induced ROS production.Ten ?mol/L rosiglitazone almost completely blocked Ang Ⅱ-induced ROS production.ConclusionPPAR? agonist rosiglitazone could block Ang Ⅱ-induced MC proliferation and extracellular matrix expression via inhibition of ROS production.
3.The basic functions of inosine 5'-monophosphate dehydrogenase and its application in drug discovery.

Inosine 5'-monophosphate dehydrogenase (IMPDH) is a key enzyme of de novo GMP biosynthesis. The expression and activity of IMPDH can be affected by diseases and physiological process. It is the drug target for anticancer, antiviral, antimicrobial and immunosuppressive therapeutics. Not only catalytic action but the other biological functions of IMPDH also play an important role in diseases. The basic functions, mechanism of catalysis, classification of inhibitors, biological functions and the latest advances to IMPDH will be illustrated in this review. It is expected to be helpful to the discovery of new inhibitors and biological functions of IMPDH.

6.Primary bone marrow diffuse large B-cell lymphoma:One case report and literature review

Objective:To analyze the diagnosis and treatment of 1 case of primary bone marrow diffuse large B cell lymphoma (DLBCL),and to provide the reference for its dignosis and treatment. Methods:Combined the medical history and other auxiliary examination results, the possibility of diagnosis of primary bone marrow lymphoma (PBML)was suspected. Bone marrow biopsy and immunohistochemistry were carried out for diagnosis, and R-CHOP (rituximab, cyclophosphamide,doxorubicin,vincristine,and prednisolone) was selected as the chemotherapy scheme. Results:The diagnostic basis of the case was in accordance with the diagnostic standard of PBML.According to the results of immunohistochemical staining,it was classified as the DLBCL subtype. Then the patient received 6 cycles of R-CHOP immune chemotherapy. Complete remission of bone marrow cell morphology was gained after the first cycle of R-CHOP treatment,and the patient was treated for another five cycles and the complete remission was retained in 3 months of follow up after drug withdrawal. Conclusion:Bone marrow biospy and immunohistochemistry have the unique diagnostic values for primary bone marrow DLBCL,and the regimens containing rituximab may provide a ideal efficacy during short term observation.
7.Methylprednisolone's influence on the plasma plasminogen activator inhibitor-1 in lupus nephritis patients

Objective:To investigate the effect of methylprednisolone that decrease plasminogen activator inhibitor-1(PAI-1) in plasma through mediating the PA/plasmin system.Methods:Employed immunohistochemical technique-ELISA to determine PAI-1 level in plasma.Results:①PAI-1 in LN patients is significantly higher than that in the control group (P0.05).Conclusion:It is thus proved that methylprednisolone is capable of decreasing PAI-1 in plasma through mediating the PA/plasmin system and two shocks is the minimum for effective cure. 8.Treatment of submucous hysteromyoma under hysteroscope

Objective To inquire clinical effects of hysteroscopic electroresection in the treatment of submucous hysteromyoma. Methods A retrospective analysis regarding curative effects of hysteroscopic electroresection in 72 cases of submucous hysteromyoma was carried out. Results The resected myoma was 1.5~7 cm in diameter, and the number of resected myoma was 1~3. The operation time was 10~90 min (mean, 30 min). The operation was accomplished on one session in all the 72 cases. No uterine perforation, bleeding and bowel injuries took place. Postoperative follow-up for 4~20 months in the 72 cases reported a satisfaction rate of 97 2% (70/72). Out of 6 patients who had pregnancy desire, 2 were pregnant with child and 1 of them had a delivery. Conclusions Hysteroscopic electroresection in the treatment of submucous hysteromyoma is safe and effective. It can markedly improve the menstruation and elevate the pregnancy rate.
