2.Effect of helicobacter pylori infection on cell proliferation and apoptosis of gastric cancer SGC-7901

Objective To investigate the effect of Helicobacter pylori infection on cell proliferation and apoptosis of gastric cancer cell line SGC 7901. Method SGC 7901 cells were incubated with 1 × 108,5 × 107,1 × 107,5 × 106 cfu / ml concentration gradient of the standard HP NCTC 11637 strains in vitro. Morphological changes of cells were observed at 24, 48, 72 hour, respectively. Cell proliferation inhibition rate were detected with MTT assay. Apoptosis were observed by flow eytometry and TUNEL analysis, mRNA expression of survivin were detected by RT-PCR, and survivin expression were determined by western blot. Results Cell proliferation inhibition rate was 54.5%, 58.9%, 67.6%, 72.9% with 1 × 108,5 × 107, 1 × 107,5 × 106 cfu / ml concentration gradient of the HP bacteria on the role of SGC 7901 cells at 72h, respectively. The rate of apoptosis after 72h detected by flow cytometry and TUNEL with HP concentration gradient of different bacteria were 42.51%, 45.67%,48.57%, 49.51% and 54.61%, 51.26%, 59.41%, 62.46.% Helieobacter pylori could significantly reduce mRNA and protein expression of survivin. These effects were strengthened as the concentration of Helicobacter pylori increased and the time extended. Conclusion H. Pylori infection may be through reducing the expression of survivin to inhibit cell proliferation and pro-mote apoptosis of SGC 7901 in vitro. Moreover, this effect was positive related with time and dose.
3.Effects of astragaloside IV on myocardial fibrosis and energy metabolism in chronic heart failure rats

AIM:To observe the effects of astragaloside IV (AS-IV) on myocardial fibrosis in chronic heart failure ( CHF) rats and to explore the underlying mechanism preliminarily .METHODS:Chronic heart failure model rats established by abdominal aorta constriction (AAC) were divided into CHF group, valsartan group and AS-IV group.Sham operation group was also established .The rats in valsartan group and AS-IV group received valsartan and AS-IV at 2 and 30 mg· kg-1 · d-1 , respectively.The rats in sham operation group and CHF group received normal saline .After 8 weeks of treatment, the cardiac structure and the hemodynamic parameters were measured .The morphologic changes of myocardial tissue were observed after staining .The expression of long-chain acyl-CoA dehydrogenase ( LCAD) and 6-phosphofructoki-nase-1 (PFK1) at mRNA and protein levels was determined by RT-qPCR and Western blot.RESULTS:Compared with sham operation group , left ventricular mass index ( LVMI) , collagen volume fraction ( CVF) , left ventricular posterior wall depth (LVPWD), and the mRNA and protein of PFK1 in CHF group were increased (P<0.05), while the mRNA and protein levels of LCAD were decreased (P<0.05).Compared with CHF group, the LVMI, CVF, LVPWD, and the mRNA and protein levels of PFK1 in valsartan group and AS-IV group were decreased (P<0.05), while the mRNA and protein levels of LCAD were increased (P<0.05).CONCLUSION:AS-IV inhibits myocardial fibrosis in the CHF rats , the mechanism of which might be associated with up-regulating the expression of LCAD , down-regulating the expression of PFK1 and normalizing the myocardial energy metabolism .
4.Application of Different Routes of Administration of Tranexamic Acid in One Stage Posterior Surgery of Thoracic Tuberculosis

OBJECTIVE:To observe the effects of different routes of administration of tranexamic acid on coagulation function and amount of bleeding in patients with one stage posterior surgery of thoracic tuberculosis. METHODS:40 patients suffered from thoracic tuberculosis in our hospital from Jan. 2011 to Dec. 2013 were randomly divided into intravenous group(5% Glucose injec-tion 100 ml+tranexamic acid 10 mg/kg,through an intravenous drip at 30 min before closing the wound) and topical application group(5% Glucose injection 10 ml and tranexamic acid 10 mg/kg,through soaking the wound before closing the wound)with 20 cases in each group. Other 15 cases suffered from the thoracic tuberculosis in our hospital from Jan. 2009 to Dec. 2010 were includ-ed in control group. 3 groups received one stage posterior surgery of thoracic tuberculosis,interbody fusion and internal fixation. The difference of hemoglobin,coagulation function and the amount of suction drainage were observed before and after surgery, and followed up. Bone graft fusion and therapeutic condition of tuberculosis were observed in the study. RESULTS:There was no statistical significance in postoperative suction drainage between intravenous group and topical application group (P>0.05),but their decrease was more significant than control group,with statistical significance(P<0.05). There was no statistically difference in fiber protease,prothrombin time or activated partial thromboplastin time among 3 groups(P>0.05). The difference value of he-moglobin in control group before and after operation was significantly higher than in intravenous group and topical application group,with statistical significance (P<0.05);there was no statistical significance between intravenous group and topical applica-tion group(P>0.05). 55 patients were all followed up and bone graft of all cases were fused,and all patients were cured and no case recurred. CONCLUSIONS:Tranexamic acid by intravenous application or topical application can reduce hemorrhage and ane-mia after operation of thoracic tuberculosis,and has no effect on blood coagulative system.
5.Impact of Astragaloside on Ventricular Remodeling and Peroxisome Proliferator Activated Receptor a Expression in Pressure-overload Rats

Objective: To study the impact of astragaloside on ventricular remodeling and peroxisome proliferator activated receptor a (PPARa) expression in pressure-overload rats and to preliminarily explore its mechanism. Methods: Pressure-overload rat's model was established by abdominal aorta constriction (AAC) in 8-week old SD rats and the result was conifrmed by echocardiography at 6 weeks later. Pressure-overload rats were divided into 4 groups with different intragastric treatment: Model control (normal saline) group, Benazepril hydrochloride [10mg/(kg.d)] group, Low-dose astragaloside [40mg/(kg·d)] group and High-dose astragaloside [80mg/(kg.d)] group; in addition, Sham operation group, the rats received intragastricnormal normal saline.n=20 in each group and all animals were treated for 8 weeks. Rat's cardiac structure and function indexes were assessed by echocardiography, hemodynamic parameter was examined by left ventricular intubation, myocardium and blood levels of free fatty acid (FFA) were determined, morphological changes of myocardial tissue was observed by HE and Masson staining, mRNA and protein expressions of PPARa were measured by qRT-PCR and Western blot analysis. Results: Compared with Sham operation group, Model control group showed increased left ventricular mass index(LVMI), collagen volume fraction (CVF) and FFA level, allP<0.05, while decreased mRNA and protein expressions of PPARa, bothP<0.05. Compared with Model control group, Low-dose and High-dose astragaloside groups presented reduced LVMI, CVF and FFA level, allP<0.05-0.01, while elevated mRNA and protein expressions of PPARa, bothP<0.01. Conclusion:Astragaloside IV mayinhibit myocardial remodeling in pressure-overload rats, which might be via up-regulating mRNA and protein expressions of PPARa, enhance myocardiumFFA utilization, and therefore improve myocardial energy metabolism.
6.Study on oral absorption enhancers of astragalus polysaccharides.

Astragalus polysaccharides was lounded to 4-(2-aminoethylphenol), followed by labeling the APS-Tyr with fluorescein-5-isothiocyanate (FITC) at the secondary amino group. The absorption enhancement effects of low molecular weight chitosan and protamine on astragalus polysaccharides were evaluated via Caco-2 cell culture model. The results show that the fluorecent labeling compound has good stability and high sensitivity. On the other hand low molecular weight chitosan and protamine also can promoted absorption of the astragalus polysaccharides without any cytotoxity, and the absorption increase was more significant with increasing the amount of low molecular weight chitosan and protamine. At the same time, the low molecular weight chitosan has slightly better effect. The transepithelial electric resistance (TEER) of Caco-2 cells show that absorption enhancers could improve its membrane transport permeability by opening tight junctions between cells and increasing the cell membrane fluidity.

7.A cross-sectional study of myopia among students from middle and high schools in Shantou, China

AIM: To investigate the status of myopia in students from different kinds of schools in Shantou city, China.
METHODS: In this cross-sectional study, 2 418 students from 4 middle and high schools in Shantou were surveyed through randomized cluster sampling. Ophthalmic examination and auto-refraction were performed for all subjects.
RESULTS: Among all 2 394 students examined. The myopia rates for students from middle and high school were 48. 07% and 68. 28%. In normal middle school, students from rural area had lower rate of myopia than students from the urban area (χ2 = 13. 879, P<0. 01 ). Students from key rural high schools had lower rate of myopia than students from key urban high school (χ2 =175. 231, P<0. 01).
CONCLUSION:The prevalence of myopia in the middle school of urban area in Shantou was higher than rural area. The increasing rate of myopia in youngsters is correlated with study stress. In order to decrease the myopia rate, it is extremely important to improve study environment in students.
8.Clinical value of two-dimensional and real-time three-dimensional transesophageal echocardiography for the guidance of left atrial appendage closure:a comparison study for LAmbre device selection

Objective To explore the clinical value of two‐dimensional transesophageal echocardiography (2D‐TEE) and real‐time three‐dimensional transesophageal echocardiography (RT3D‐TEE) for the left atrial appendage (LAA) closure procedures by the visualization of LAA shape by 2D‐TEE and RT3D‐TEE and the comparison between the measurement of LAA ostium and the sized LAmbreTM device during the procedure .Methods Forty‐one atrial fibrillation patients ,who had undergone 2D‐TEE examination at our hospital ,were enrolled in the study .At the mid‐esophageal ,dimensions of inner and outer ostium and depth of LAA were measured at the 2D‐TEE views of 0 ,45 ,90 and 135 degree respectively . RT3D‐TEE views were acquired and the maximal and the minimal dimensions of LAA inner ostium were measured .The measurement by RT3D‐TEE and 2D‐TEE were compared to find the difference and correlation .Eleven of 41 patients who have complied with the requirements for the LAA closure ,were undergone the procedures ,measured the dimension of LAA inner ostium at selective angiography intraoperative .Sizes of closure disks of the closure device and the measurement at selective angiography were recorded to compare the measurement at RT3D‐TEE and 2D‐TEE .Results Forty‐one atrial fibrillation patients were completed TEE examination successfully .Inner ostial dimension of LAA was (20.0±04.3)cm,(19.7±03.8)cm,(21.2±04.6)cm,(23.0±05.0)cmat2D‐TEEviewsof0,45,90and135 degree ,respectively .The maximum dimensions of LAA inner ostium by RT3D‐TEE was (2 4.9 ± 0 5.2)cm . At 2D‐TEE views ,the maximum dimensions of LAA inner ostium was at 135 degree ,there was a difference between it and the measurement by RT3D‐TEE ( P =0 0.12) .Monitoring by TEE ,LAA closure procedures with LAmbreTM device were successful for all 11 patients ,the landing zone by selective angiography was (2 4.9 ± 0 4.4)cm ,and the appropriate sized closure disk of the LAmbreTM device was 2 4. - 3 6. cm . Correlation between the measurements by RT3D‐TEE and selective angiography and the sized closure disk were r =0 8.16 ,P =0 0.02 and r =0 9.14 ,P =0 0.00 ,respectively .Correlation between the measurements by 2D‐TEE and selective angiography and the sized closure disk were r =0 6.93 ,P =0 0.18 and r =0 6.88 , P=0 0.19 ,respectively .Conclusions There was better correlation among the measurements by RT3D‐TEE and selective angiography and the size of closure device .Therefore ,compared to 2D‐TEE ,the guidance of RT3D‐TEE was more accurate during LAA closure procedures for LAmbreTM device selection .
9.The accuracy of bony resection and component size planning of total knee arthroplasty assisted with a novel designed patient-specific instrumentation

Objective:To investigate the accuracy of bony resection and component size planning of total knee arthroplasty (TKA) assisted with a novel designed patient-specific instrumentation (PSI).Methods:Thirty-eight TKAs assisted with a novel designed PSI of thirty-five patients from March 2018 to April 2019 were retrospectively analyzed. There were 11 males (10 knees) and 27 females (25 knees), aged 67.7±6.9 years (range 49-81 years). Intraoperative bone resection thicknesses at medial distal femur (MDF), lateral distal femur (LDF), medial posterior femur (MPF), lateral posterior femur (LPF), anterior femur (AF), medial tibial (MT), lateral tibial (LT) were measured with vernier caliper and compared with the preoperative planned bone resections as a primary outcome. The femur and tribal component sizes used intraoperatively were recorded and compared the preoperative planning.Results:The difference of thickness was -0.1±1.6 mm at MDF, -0.5±1.6 mm at LDF, 0.8±1.7 mm at MPF, 0.0±1.7 mm at LPF, 0.0±1.4 mm at AF, -0.3±1.4 mm at MT and 0.3±1.5 mm at LT. The ratio of differences <2 mm and <3mm were 87.8% and 93.94% at MDF, 87.8% and 93.94% at LDF, 71.3% and 85.71% at MPF, 77.14% and 91.43% at LPF, 88.46% and 100% at AF, 85.29% and 97.06% at MT, 83.78% and 94.59% at LT. 89.47% of femoral components were the same size with preoperative planning, 10.53% femoral components were within 0.5 size compared to the preoperative planning, no femoral components were over 0.5 size compared to the preoperative planning; 36.84% of tibial components were the same size with preoperative planning, 60.53% tibial components were within 0.5 size compared to the preoperative planning, 2.63% tibial components were within 1 size compared to the preoperative planning, notibial components were over 1 size compared to the preoperative planning.Conclusion:TKA assisted with a novel designed PSI shows good accuracy with bone resection and component size planning. The procedure of TKA could be simplified with this technique.
10.A study on yearly and daily circadian rhythm of cardiovascular events

Objective To investigate the circadian and seasonal distribution of cardiovascular events (CVE) and the relationship between average monthly temperature and the incidence of CVE. Methods A total of 5837 emergency patients with CVE were enrolled from 1997 to 2007. The exact admission time of each patient was registered. The average monthly temperature data from a regional weather station for this time period was supplied by the meteorological office of the city. The relationship between the average monthly temperature and the incidence of CVE was explored and the corresponding curves were plotted. Results The occurrence of CVE has obvious seasonal variation, and its a higher tendency of episodes was found in spring and winter periods. High incidence of acute myocardial infarction, angina pectoris, acute left-sided heart failure, hypertensive crisis, and sudden death occurred in spring and winter as wee as the time for a change of season, that is, January, February, March, April, October, November, and December. There was a significant negative correlation between the incidence and the average monthly temperature (P<0.005-0.001). Circadian rhythm of CVE was present, with a peak in the early morning and forenoon. Conclusion There are circadian and circannian rhythms of the incidence of CVE. Cold weather condition is a risk factor for the occurrence of CVE, which usually peaks in the early morning and forenoon.
