1.Effects of platelet-derived growth factor, basic fibroblast growth factor and epidermal growth factor on the proliferation, migration and phenotypic modulation of airway smooth muscle cells

Objective To investigate the effects of platelet-derived growth factor(POGF),basic fibroblast growth factor(bFGF)and epidermal growth factor(EGF)on the proliferation,migration and phenotypie modulation of airway smooth muscle cells(ASMCs)in vitro.Methods Cultured human ASMCs were stimulated with PDGF.bFGF,EGF or DMEM(control).Cell pmliferation was detected by BrdU incorporation and cell count.Cell migration was measured using Boyden's chamber.Reverse transcription-poly-merase chain reaction(RT-PCR)and Western blot analysis were used for the detection of sm-α-actin and sm-MHC mRNA and protein expression in ASMCs.Resuits The mitotic index(%BrdU-positive per total number of cells)of ASMCs stimulated with PDGF.bFGF and EGF were significantly higher than those in control(P<0.001 or P<0.05).And the cell number of ASMCs stimulated with PDGF,bFGF and EGFwere also significantly higher than those in control(P<0.05).The number of migrating cells treated with PDGF,bFGF and EGF were higher than that in eontrol(P<0.05).The expressions of sm-α-actin mRNA treated with bFGF and EGF were significantly lower than those in control.but it was higher than control when treated with PDGF.The expressions of sm-MHC mRNA treated with bFGF and EGF were significantly lower than those in control,but it was higher than control when treated with PDGF(P<0.01 orP<0.05).Theexpressions of sm-α-actin protein treated with bFGF and EGF were significantly lower than those in control,but it was higher than control when treated with PDGF(P<0.05).The expressions of sm-MHC protein treated with bFGF and EGF were significantly lower than those in control.but it was higher than control when treated with PDGF(P<0.05).Conclusion PDGF,bFGF and EGF can directly induce ASMCs,proliferation and migration in vitro;and meanwhile,the expressions of cells,contractive phenotype increased treated with PDGF and decreased treated with bFGF and EGF.
2.Applications of IVUS in Modern PCI Procedure

Intravascular ultrasound (IVUS) is a commonly used technology in percutaneous coronary intervention (PCI) procedure. It has higher resolution and repeatability, and more importantly, it can make the vascular structure visiable which shows the plaque morphology and component, compared with traditional coronary angiography. Most researches indicated that IVUS can help doctors find out the optimized stent size, stent malapposition,stent disexpansion,and procedure complication, thus reduce the incidence of restenosis and stent thrombosis. IVUS has been widely applied in research about the predictors of PCI complication, these results benefit us to make better decision on PCI procedure. In this article, we reviewed the studies based on IVUS technology during the last 10 years and summrized according to restenosiss and stent thrombosis with BMS (bare metal stent) and DES (drug eluting stent) .
3.Anatomic measurements of bony orbit of Chinese Han-nationality adults

Objective: To measure the distances among anatomic points of bony orbit of Chinese Han-nationality adults. Methods: Bony orbits were measured with the special measuring tools in 86 Chinese adults (62 males and 24 famales), and the results were statistically analysed. Results: Difference between the left and right obits was observed in orbital width, medial distance of superior orbital foramen a and inferior foramen a and c in males. Difference between bilateral orbits was also observed in orbital width and anterior distance of superior orbital fissure in females. Normal values of the distances of anatomic points of adult orbits were then calculated. Conclusion: Normal values of the distances of anatomic points of orbit of male and female adults are concluded.

Objective To evaluate solitary pulmonary nodules (SPNs) using quantitative dynamic contrast enhanced functional computed tomography (CT), and to illustrate its clinical efficacy in differential diagnosis between benign and malignant pulmonary nodules.Methods Eighty patients with non-calcified SPNs (diameter, 5-30 mm) were studied with dynamic contrast enhanced CT.Patterns of time-density curve (T-DC) were assessed.The precontrast density, peak height in density (PH: the maximum value of the T-DC), and S/A ratio (the ratio of the PH of SPN over aorta) were recorded.Enhancement patterns of SPNs were also recorded.The perfusion of SPNs was calculated.Results Malignant, benign, and inflammatory nodules showed quite different patterns in T-DC.PH and S/A ratio of the malignant and inflammatory nodules were significantly higher than those of the benign nodules (P

OBJECTIVE:To establish a method for the determination of antibacterial effect of Ofloxacin eye drop. METH-ODS:According to the requirements in Chinese Pharmacopoeia(2015 edition)antibacterial effect test,using Staphylococcus au-reus,Pseudomonas aeruginosa,Escherichia coli,Aspergillus niger and Candida albicans as test bacteria,colonycount method suit-ability test was conducted for Ofloxacin eye drop samples,the verified method was used to determine the number of viable bacteria at each time point,calculate the bacteria number of 1 mL in the sample and the number of bacteria at each time point and convert to lg value. RESULTS:Ofloxacin eye drop sample can reach the standard of antibacterial effectiveness of theB. CONCLU-SIONS:The method can effectively determine whether the antibacterial effect of Ofloxacin eye drop fits the standard.

Picture archiving and communication system ( PACS ) based on digital teaching platform of medical imaging consists of web teaching system and teaching resource library of medical imaging.The web teaching system includes four function modules of classroom management,work management,teaching management and test management.The teaching resource library includes script,pictures,and audio,video and animation materials of theoretical books,typical cases,examination techniques and medical imaging equipments.Utilized in theoretical and practical classes,the teaching characteristic of medical imaging is made prominent,and the subjective motility of the students is brought into play.
8.Development of a Software for Automatically Generated Contours in Eclipse TPS.

OBJECTIVEThe automatic generation of planning targets and auxiliary contours have achieved in Eclipse TPS 11.0.
METHODSThe scripting language autohotkey was used to develop a software for automatically generated contours in Eclipse TPS. This software is named Contour Auto Margin (CAM), which is composed of operational functions of contours, script generated visualization and script file operations. RESULTS Ten cases in different cancers have separately selected, in Eclipse TPS 11.0 scripts generated by the software could not only automatically generate contours but also do contour post-processing. For different cancers, there was no difference between automatically generated contours and manually created contours.
CONCLUSIONThe CAM is a user-friendly and powerful software, and can automatically generated contours fast in Eclipse TPS 11.0. With the help of CAM, it greatly save plan preparation time and improve working efficiency of radiation therapy physicists.

9.Oxidative damage of human retinal pigment epithelium cells induced by blue light irradiation and mitochondria-participated mechanism

Background Researches showed that mitochondria and oxidative stress play a crucial role in retinal photochemical injury,but the relationship between the damage of human retinal pigment epithelium (RPE) cell-induced by blue light and light-irradiated time is less studied.Objective The aim of this study was to research the possible mechanism of RPE oxidative damage induced by blue light in vitro.Methods Human RPE cells were isolated from healthy donors and cultured.The cells were divided into the normal control group and the light exposure group.The cells of light exposure group were irradiated using the blue light of (4.0±0.5) mW/cm2 for 0.5,1,2,3,4,5,6,12 and 24 hours,respectively,and the cells of the normal control group were cultured in dark environment.Cellular viability was detected by MTT method,and the ultrastructure change of subcellular organelles in RPE cells was examined under the transmission electron microscope (TEM).The content of reactive oxygen species (ROS) was assayed by flow cytometry for the assessment of oxidative stress reaction.The relative expressions of nicotinamide adenine dinucleotide phosphate (NADPH) mRNA and cyclooxygenase 1 (COX1) mRNA in the cells were detected by real-time fluorescence quantitative PCR to evaluate the mitochondria function.Results The percentages of cellular viability were (100.00±20.00) %,(95.73±0.89) %,(94.67±2.56) %,(84.23±0.16) %,(78.57±3.09)%,(75.43±2.18)%,(66.13±1.42)%,(53.43±1.91)% and (47.97±1.36)% in the normal control group and light exposure for 1-hour,2-hour,3-hour,4-hour,5-hour,6-hour,12-hour and 24-hour groups,respectively,showing a significant difference among the groups (F =172.270,P =0.000),and the percentages of light exposure for the more than 3 hours groups were significantly lower than those of the normal control group (all at P< 0.05).The vacuoles-like degeneration,mitochondrial swelling,decreased microvilli were seen under the TEM.The contents of ROS in RPE cells were (14.75±2.49)%,(19.04± 1.02) %,(22.81 ±3.20)%,(28.75±2.15)%,(33.06±0.96) %,(40.64±2.11) %,(48.25±2.50) % and (60.44±2.68) % in the normal control group and light exposure for 0.5-hour,1-hour,2-hour,3-hour,4-hour,5-hour,6-hour groups,and with significant increases in ROS contents in various light exposure groups compared with the normal control group (all at P<0.05).The relative expression levels of NAPDH mRNA in the cells were gradually elevated 3 hours after light exposure with the increase of time in comparison with the normal control group (all at P<0.05),and the relative expression levels of COX1 mRNA in the cells were higher in the light exposure for 2-hour,3-hour,4-hour and 5-hour group compared with the normal control group (all at P<0.05),and after that the COX1 mRNA levels were gradually declined and were close to the normal level.Conclusions Blue light irradiation for more than 3 hours causes oxidative stress damage of mitochondria in RPE in vitro,and the damage was more obvious after irradiation for 5-6 hours.
10.PARP-1, CK7/20 and p53 in Barrett's esophagus and esophageal adenoeareinoma: expression and its significance

Objective To investigate the expression of poly (ADP-ribose) polymerase-1 (PARP-1 ), cytokeratins (CK) 7/20, and p53 in patients with Barrett's esophagus and esophageal adenocareinoma,and to evaluate their significance. Methods Expression of PARP-1, CK7/20 and p53 were determined by immunohistochemistry in 108 patients (including 40 Barrett's esophagus, 28 esophageal adenocarcinoma and 40 cardiac mucosa). Results The expression of PARP-1 was found in Barrett's esophagus, esophageal ade-nocarcinoma and cardiac epithelium with a significantly higher level in esophageal adenocarcinoma than the other two groups (P <0. 01 ). CK7/20 was expressed in much of intestinal metaplasia, part of cardiac epi-thelium and adenocarcinoma cells. The positive expression of p53 was observed in all three groups, and it was significantly higher in adenocarcinoma group than in other two groups (P < 0. 05 ). PARP-1 expression is highly correlated with that of p53 in Barrett's esophagus ( r= 0.49, P < 0.01 ). Conclusion CK7/20 is a sensitive but less specific indicator for intestinal metaplasia. Both PARP-1 and p53 are involved in the patho-genesis of esophageal adenocarcinoma and might help to determine the risk of Barrett's esophagus developing into esophageal adenocarcinoma.
