1.Effect of hyperbaric oxygen preconditioning on expression of MMP-2, MMP-9 and Cx43 in rat abdominal skin flap model of ischemia-reperfusion injury

Objective To study the effect of hyperbaric oxygen preconditioning management on the expression of MMP-2,MMP-9 and Cx43 in a rat abdominal skin flap model of ischemia-reperfusion injury.Methods Eighteen male adult SD rats,weight ranged from 220-250 g,were randomly divided into three groups:sham group (SH),ischemia-reperfusion group (IR) and hyperbaric oxygen preconditioning group (HBO).All the rats in HBO group received hyperbaric oxygen treatment twice each day,apart 12 hours,during the last three days before operation.Abdominal skin flap with superficial epigastric artery as pedicle was established.In HBO and IR group,3 hours of ischemia was performed.On the 3rd postoperative day,samples were taken to assess the expression of MMP-9,MMP-2 and Cx43 by immunohistochemical staining and Western blot.Results Compared with IR group,the expression of Cx43 was significantly increased (IR 15.03±3.66;HBO 36.01±4.12) and the ex pression of MMP-9 and MMP-2 was decreased (MMP 2:IR 12.01±1.23;HBO 5.98±1.48;MMP9:IR 16.77±2.01;HBO 11.48±1.77).Conclusions Hyperbaric oxygen preconditioning for the rat abdominal skin flap model of ischemia-reperfusion injury has inhibitory effect on the expression of MMP-9 and MMP-2,and stimulative effect on the expression of Cx43.
2.Effect of the surgical treatment on the keloid of mons pubis with postoperative radiotherapy

Objective To explore an effective surgical treatment to the keloid of mons pubis.Methods From March 2008 to January 2015,local flap and adjacent flap (including ilioinguinal flap and rectus abdominal musculocutaneous flap) were performed,and used to repair the defect after surgical removal of the keloid in 25 patients,and all the patients were received postoperative local radiotherapy,and then chi square test was applied to evaluate the effect in the six aspects,including perineal form,skin color,pubic hair distribution,satisfactory effect,skin texture and sense.Results The local flap group was better than the adjacent flap group in perineal form,skin color,pubic hair distribution,satisfactory effect,but there was no significant difference in skin texture and sense.Conclusions For the keloid of mons pubis,the first choice is local flap graft,which can maintain the normal mons pubis formation as most as possible,while the wound is so large that the local flap cannot work,and therefore the vascular pedicle axial flap should be used.

Medical quality and safety are essential safeguards for the harmonious society in China, directly affecting people's sense of security and happiness. Continuously improving medical quality and ensuring medical safety are fundamental and essential to implementing the strategies of the Party Central Committee and the State Council, and promoting the construction of a healthy China. In recent years, with the rapid development of China's economy and the people's increasing emphasis on their own health and safety, China's plastic surgery industry has achieved significant progress and yielded outstanding results. However, along with the rapid growth of the industry, medical quality issues and safety accidents have occurred frequently, attracting widespread attention from all sectors of society. This article briefly outlines the history of quality management and quality control for medical quality and safety in China's plastic surgery specialty. It explores the development of quality management from its origin to medical quality management, as well as the responsibilities and contributions of the National Plastic Surgery Quality Control Center in improving industry standards and ensuring patient safety. It aims to provide support for the standardized management of the plastic surgery industry in the future, so as to promote its healthy and sustainable development.
4.Data Development Trend of Public/Private Medical Institutions in Chinese Plastic and Aesthetic Major

To compare and understand the trends in quality control data changes in public and private medical institutions specializing in plastic and aesthetic major in China over the past three years. The national quality control center of plastic and aesthetic major formulated quality control indicators, collected indicator data through the National Clinical Improvement System, conducted feedback verification on the data, organized and summarized the final data, and convened expert committees to analyze the data change trends. In the past three years, the number of both public and private medical institutions specializing in plastic and aesthetic major has shown an upward trend. The number of doctors increased significantly in 2022, with over 90% of doctors in public institutions (about 70% in private institutions) receiving higher education. In recent years, the proportion of doctors from other specialties engaging in plastic and aesthetic activities has increased. The proportion of inpatients seeking aesthetic treatments has also increased. Among outpatient patients, the proportion of injection procedures was relatively low. About one-third of private medical institutions have not yet adopted an electronic medical record system. Data feedback is the cornerstone of medical quality control in Chinese plastic and aesthetic major. Existing data indicates that the related industry is developing steadily, but there is still room for improvement in terms of some indicator data. However, due to the limited data from private medical institutions, it is still necessary to encourage them to improve their reporting enthusiasm in order to conduct a more comprehensive assessment in the future.
5.Construction of Medical Quality Control Indicators System for Chinese Plastic and Aesthetic Major

To construct a quality control indicators system for Chinese plastic and aesthetic major and lay foundation for medical quality control. National Quality Control Center of Plastic and Aesthetic Major established a working group in February 2023. Guided by the "structure-process-outcome" theory, the working group formulated medical quality control indicators for Plastic and Aesthetic major by learning from relevant indicators of other majors, reviewing literature, discussing in meetings, and combining opinions from quality control experts. The quality control indicators system was finally established by Delphi expert consultation. Delphi survey was 100% of 2 rounds. The authorities of 2 rounds of expert consultation were 0.854 and 0.857. The harmonious coefficients were 0.387 of primary indicators and 0.425 of secondary indicators( The quality control indicators system established in this study for Plastic and Aesthetic major in China has a certain degree of scientificity and rationality, which offers reference for medical quality control of Plastic and Aesthetic major in China. However, this system should be improved and ameliorated in practical application.
6.Effects of HOTAIR on epithelial mesenchymal transition in HaCaT cells

Objective:To verify that hypoxia induces epithelial mesenchymal transition in HaCaT cells, and to observe the effect of LncRNA HOTAIR on epithelial mesenchymal transition in HaCaT cells.Methods:From Jan. 2018 to Dec. 2018 in Chinese Academy of Medical Science, HaCaT cells were divided into four groups: group A cultured under normoxia, group B cultured under hypoxia, group C transfected with sh-control cultured under hypoxia, and group D transfected with sh-LncRNA HOTAIR cultured under hypoxia. After 36 hours of culturing, the expression of E-cadherin and vimentin were detected using qPCR and Western blot. The migration of HaCaT cells was evaluated by Transwell assay.Results:The relative quantity of E-cadherin mRNA in the four groups were 3.076±0.271, 1.000±0.089, 1.024±0.222, and 2.595±0.085, while vimentin mRNAs were 1.002±0.183, 4.170±0.279, 4.111±0.477, and 2.412±0.134, respectively. In addition, the Transwll invasion assay showed that numbers of cell migration in the four groups were 32.70±3.93, 125.40±6.26, 120.10±6.79, and 58.24±7.06, respectively.Conclusions:The study suggests that hypoxia promotes epithelial mesenchymal transition in keratinocytes. Furthermore, downregulation of HOTAIR is noted to inhibit epithelial mesenchymal transition of keratinocytes under hypoxia condition.
7.The effect of hyperbaric oxygen preconditioning on the expression of ICAM-1, VCAM-1, NF-κB and flap survival rate during ischemia-reperfusion injury in rat abdominal skin flap

Objective To evaluate the effect of hyperbaric oxygen preconditioning on the expression of intercellular adhesion molecule-1 (ICAM-1),vascular cell adhesion molecule-1 (VCAM-1),NF-κB and flap survival rate during Ischemia-Reperfusion Injury in Rat Abdominal Skin Flap.Methods 18 male adult SD rats were divided randomly into sham group (SH),ischemia-reperfusion group (IR) and hyperbaric oxygen preconditioning group (HBO),6 rats in each group.The rats in HBO group received regular hyperbaric oxygen treatment for three days (twice a day with an interval of 12 h) before operation.An abdominal skin flap pedicled with superficial epigastric artery was established.Ischemia lasted for 3 hours in the IR group and HBO group.On the third postoperative day,samples were taken to evaluate the expression of ICAM-1,VCAM-1 and NF-κB by immunohistochemistry staining and Western Blot.Results In the IR group,the flap survival rate and average blood perfusion were (22.38 ±4.35) % and (32.61 ±5.68) PU,while in the HBO group they were (45.34 ±3.15) % and (61.78 ±3.61) PU,showing significant difference between the two groups (P <0.01).The inflammatory cell infiltration condition in IR group was much higher than that in the HBO group from the HE staining observation.Compared with the HBO group,ICAM-1,VCAM-1 and NF-κB showed (+ + +) with higher positive cells in the IR group (P < 0.01).Results of Western Blot showed similar conclusion.The relative expression of ICAM-1,VCAM-1 and NF-κB was much higher in the IR group than that in the HBO group (P < 0.01).Conclusions Hyperbaric oxygen preconditioning could decrease the expression of ICAM-1,VCAM-1,NF-κB and promote flap survival rate during the process of ischemia-reperfusion injury on a rat abdominal skin flap model.

9.Application effects of PBL teaching method on the chapter teaching of "Chronic diseases nursing in community" for the course of Community Nursing in traditional Chinese medicine colleges

Objective To investigate the effects of problem-based learning (PBL) applied in the chapter of " Chronic diseases nursing in community" in course of Community Nursing in traditional Chiuese medicine colleges.Methods A total of 50 undergraduate students of 201 1 in the nursing specialty in some traditional Chinese medicine colleges were chosen with the convenience sampling method.They were randomly divided into control group and observation group,with 25 students in each group.PBL was applied in the observation group,choosing 2 teaching blocks from the course of Community Nursing.The same blocks were chosen for the method of teaching and learning,which was applied in the control group.The score of California Critical Thinking Disposition Inventory (CCTDI) of students and the teaching effects in two groups were evaluated one week before and after PBL implemented.Results The total score of CCTD1 in the observation group was(275.36 ± 16.41) after intervention,while the score in the control group was(266.28 ± 18.67) (P >0.05).The mean increase scores in the observation group were higher than that in the control group,but the difference was not statistically significant (P > 0.05).The mean scores of PBL teaching evaluation were closed to the median in basic knowledge reserve,study attitude,ability improvement,cooperation,professional quality in the observation group.Conclusions The application of PBL teaching method in the teaching of Community Nursing does not have strongly impact on the ability of students' critical thinking.
10.Role of Estrogen Receptor-alpha in the Regulation of Claudin-6 Expression in Breast Cancer Cells.

PURPOSE: In our previous studies we showed that upregulating claudin-6 (CLDN6) expression may contribute to preventing breast cancer, and that 17beta-estradiol induces a concentration- and time-related effect on CLDN6 mRNA and protein expression in MCF-7 cells. However, the mechanisms of 17beta-estradiol regulation of CLDN6 are still unclear. We determined the role of estrogen receptors in the regulation of CLDN6 expression in human breast cancer tissues and a cell line. METHODS: CLDN6, estrogen receptor alpha (ERalpha) and estrogen receptor beta (ERbeta) expression in breast cancer tissues were examined using immunohistochemistry. The human breast cancer cell line, MCF-7, which expresses ERalpha but not ERbeta was used. CLDN6 and ERalpha expression were measured by reverse transcriptase-PCR, Western blotting and immunofluorescent staining. Treatments with propyl pyrazole triol (PPT) and ICI 182, 780 (ICI) were performed. RESULTS: The results revealed that CLDN6 expression was related to ERalpha in breast cancer tissues (p=0.033). PPT, an ERalpha-selective ligand, upregulated CLDN6 expression at 10-5 mol/L after 24 hours. The effect of PPT on regulating CLDN6 expression in MCF-7 cells was blocked by ICI. CONCLUSION: These findings suggest that Eralpha reulates CLDN6 expression in breast cancer tissues and that 17beta-estradiol induces CLDN6 expression through an ERalpha pathway in MCF-7 cells.
