5.Temporal and spatial distribution of TGF-β in corneal wound healing
Recent Advances in Ophthalmology 2017;37(2):184-188
Fibrosis is the major cause of corneal scarring.Transforming growth factor-beta (TGF-β) plays a key role in corneal homeostasis and repair.Corneal epithelial basement membrane is thought to be the important barrier of corneal epitheliumstroma interaction. In different stages of corneal wound healing, the isoforms of TGF-β have different temporal and spatial expression. The integrity of basement membrane is a critical factor of these procedures. The temporal and spatial distributions of TGF-β isoforms play the crucial roles in cell migration, proliferation, phenotype changes and deposition of extracellular matrix in corneal wound healing. It is the mechanism of corneal scarring and scar-free healing. This article reviews recent articles to elucidate the biological functions of TGF-β and the temporal and spatial distribution of its isoforms in corneal wound healing.
6.Application of cyclosporine A in noninfectious uveitis
Recent Advances in Ophthalmology 2017;37(3):293-297
Uveitis is a kind of refractory blinding eye diseases.Noninfectious uveitis is an important part of uveitis, which is thought to be an autoimmune disease, and systemic immunosuppressive therapy is usually required to prevent visual loss.As a selective T-cell inhibitor, cyclosporine A is one of the most widely used immunosuppressants, and has obvious therapeutic effect on many kinds of uveitis.Cyclosporine A combined with glucocorticoid, other immunosuppressants or biological agents can improve the therapeutic effect and reduce the side effects.Recently, the inhibitory effect on glucocorticoid-resistant Th17 cells and topical application of cyclosporine A,show its advantage and potential in the treatment of noninfectious uveitis.

9.Factors related to duration of serum HBsAg in patients with acute hepatitis B
Journal of Clinical Hepatology 2014;30(11):1173-1175
Objective To investigate the factors related to negative conversion of HBsAg in patients with acute hepatitis B (AHB).Meth-ods A total of 106 AHB patients who were admitted to the Department of Infectious Diseases,Zhoukou Central Hospital from February 2007 to February 2013 were recruited.Liver function, five serological markers of HBV infection (HBsAg,HBeAg,anti-HBs,anti-HBe,and anti-HBc),and HBV-DNA were measured every three months. All patients were followed up for 12 months. The major HBV genotypes (A through D)were determined by type-specific primer nested PCR.Categorical data were expressed as rates and compared with χ2 test;continuous data were expressed as mean ±SD and analyzed by t-test.Results When just admitted to our hospital, compared with the pa-tients with negative conversion of HBsAg,AHB patients with HBsAg persisting for more than 6 or 12 months had a significantly lower peak level of alanine aminotransferase (ALT)(718 ±696 vs 1282 ±913 U/L,622 ±514 vs 1203 ±924 U/L,P<0.05)and a significantly high-er level of HBV-DNA (6.8 ±1.4 vs 5.2 ±1.5 log10 copies/ml,7.3 ±1.6 vs 5.4 ±1.5 log10 copies/ml,P<0.05).The HBsAg clear-ance rate in patients with HBV genotype A was significantly lower than that in patients with HBV genotype B or D (75.0% vs 89.5% or 100%,83.3% vs 97.4% or 100%,P<0.05).Conclusion The HBsAg clearance rate in AHB patients may be associated with HBV genotypes, peak level of ALT,and HBV-DNA.
10.Effect of AngⅡ on Insulin Gene Expression in RIN-m Cells and Its Molecular Mechanism
China Pharmacist 2015;18(12):2025-2029
Objective:To discuss the effect of AngⅡ on insulin gene expression IN RIN-m cells and its molecular mechanism. Methods:RIN-m cells were cultured and divided into three groups, including the control group, 100 nmol·L-1 AngⅡ group and losartan pretreatment group. After 24-hour incubation, insulin gene expression in RIN-m cells was detected by RT-PCR, the mean flu-orescent intensity of 2', 7'-dichlorofluorescein ( DCF) was detected by flow cytometry, PDX-1 and MafA mRNA expression were detec-ted by RT-PCR and the protein expression was detected by Western-blot. Results: Insulin expression in RIN-m cells, cellular ROS level, PDX-1 expression and MafA expression in 100 nmol · L-1 AngⅡ group were significantly different from those in the control group and losartan pretreatment group (P<0. 05). The later two groups had no significant differentces in those indices (P>0. 05). Conclusion:AngⅡ can down-regulate PDX-1 and MafA expression inβ-cells through oxidative stress pathway, and then inhibit insu-lin gene expression. Pretreatment with losartan can antagonize the effect of AngⅡ, and has protective effect onβ-cells in the aspect of insulin gene expression.
