2.Bladder Pain Syndrome (review)
Chinese Journal of Rehabilitation Theory and Practice 2016;22(5):552-558
Bladder pain syndrome (BPS) is a chronic disease characterized by urinary symptoms and pain in or around the bladder with uncertain reasons. The incidence of BPS is higher in the female and people in American and Europen countries. The BPS patients' quality of life has been damaged and a new disabled group has been formed. The pathogeneses of BPS include pathogens inducing inflammation, chronic ischemia and ischemia-reperfusion injury producing degeneration in bladder, lower urinary epithelium dysfunction, apoptosis in bladder tissue, epigenetic reprogramming, abnormal intracellular signaling pathway, pain regulating dysfunctioning, and autophagy inhibit-ed. There is no identified diagnosis for BPS. Cystoscopy, urodynamia, and biomarks in blood or urine are helpful in diagnosis. Diet, psycho-logical intervention, oral medicine, intravesical therapy, nerve blocking and nerve stimulation, and surgeries have been used for it, but the ef-ficacy remains unsatisfied.

Objective To evaluate the effect of incision of the intrarenal sinus plus postrenal low pole segmental incision for removal of calculus on the renal hemodynamics. Methods The hemodynamic changes of intrarenal and incisional sites in 98 cases with complex calculi were consecutively monitored by color Doppler ultrasonograph before operation and also 1 month,3 months,6 months after operation. The ratio(S/D) of the peak flow-rate in systole(S),the flow-rate at the end of diastole(D),and the resistant index(RI) of the intrarenal chief renal arteries and intersegmental arteries and interlobar arteries were analyzed. The signals of blood stream in segmental incision were divided into four classes(Ⅰ to Ⅳ) and were observed. Results The changes of the intrarenal hemodynamics were related to the degrees of hydronephrosis(P0.05),but it caused the blood stream to be obstructed in the area of incision where the signals of blood stream showed classes Ⅲ to Ⅳ.The patients generally recovered after 3 to 6 months, and the time of recovery was associated with the degrees of hydronephrosis Conclusions It is concluded that the incision of the intrarenal sinus plus postrenal low pole segmental incision for removal of calculus is an effective and safe method for treat-ment of complex renal calculus.

Objective To investigate the proliferation inhibition and apoptosis induction effect of thalidomide(Thal) and its joint effect with 5-Fu on MGC-803 cell line. Methods The morphological changes of MGC-803 cells with AO/EB stain were observed under fluorescence microscope. The proliferation inhibition effeet was evaluated with MTT. The apoptosis induction effect was determined by FAM. Results The results of MTr array were as below: the difference was significant between Thai groups of 25 mg/L or above concentration and the control group (P<0.05), 5-Fu of all testing concentrations showed significant difference compared with the control gToup(P<0.01). For combined groups with the same concentration of 25 mg/L 5-Fu,combined groups showed distinct difference from the corresponding 5-Fu group (P<0.01), but two combined groups showed no distinct difference with each other (P>0.05). For combined groups with the same concentration of 5-Fu 12.5 mg/L, 5-Fu plus 50 mg/L or 100 mg/L Thai showed no distinct difference from 5-Fu group of 25 mg/L(P>0.05). With FAM study, all test groups showed significant difference compared with the control group (P<0.01). There was also significant difference between two test groups arbitrarily (P<0.01).Conclusion Both Thai and 5-Fu could inhibit the proliferation and induce the apoptosis of MGC-803 cells. The effect is reinforced with the combination significantly. Combination with Thai could decrease the concentration of 5-Fu but have the similar effect.

Objective: To isolate small molecular polypeptides which bind to KDR specifically using C7 and 12 peptide libraries. Methods: KDR/IgGFc was coated directly on plates, C7 and 12 peptide libraries are then applied, followed by vigorous washings in washing buffers to remove most non-specifically bound phage and to select specific phage particals by VEGF suspension and elution in acid buffers. The positive phage clones were detected by ELISA, cell-ELISA and competitive binding ELISA. Results: After three washes, 12 positive phage clones were selected and sequenced. Conclusion: A conserved peptide motif was not found in these sequences. The peptide binding to KDR specifically may be helpful for lead drug to improve selectivity and decrease the side effect of anti cancer drug in clinical treatment.
