1. The protective mechanism of Naoxintong on cerebral ischemic injury in rats

Objective: To observe the influence of Naoxintong on expression of glucose-regulated protein 78(GRP78) and GRP94 in the striatum of rats with focal cerebral ischemic injury, so as to investigate their possible protective mechanism on cerebral ischemic injury. Methods: Ninety rats were equally randomized into 3 groups(n=30): sham operated group, ischemic group (Focal transient cerebral ischemia model was established with intraluminal occlusion of left middle cerebral artery) and Naoxintong pre-treated group (Treated with Naoxintong 5 d before ischemic injury). The expression of GRP78, GRP94 in rats striatum was detected by histological method, immunohistochemistry staining and semiquantitative RT-PCR at the different time points(6,12 and 24 h after ischemic treatment). Results: The focal transient cerebral ischemia model was successfully established in rats. Histological results showed that the degree of focal cerebral ischemic injury in Naoxintong pre-treated group was significantly lower than that in ischemic group. Immunohistochemistry staining and RT-PCR results illustrated that the expression of GRP78 and GRP94 in ischemic group was lower than that in sham operated group at each time point after ischemic treatment (P<0.01). The expression of GRP78 and GRP94 in ischemic group and Naoxintong pre-treated group were the lowest at 6 h and the highest at 12 h, and the expression at 24 h was lower than that at 12 h but higher than that at 6 h. Compared with the ischemic group, the expression of GRP78 and GRP94 was higher in Naoxintong pre-treated group at the same time point (P<0.05,P<0.01). Conclusion: The expression of GRP78 and GRP94 is upregulated within 12 h of cerebral ischemic injury in rats. Naoxintong can promote the expression of GRP78 and GRP94 in ischemic cerebral areas, protect the function of endoplasmic reticulum and relieve the cerebral injury caused by ischemia.
2. The changes of expressions of the endoplasmic reticulum chaperones GRP78, GRP94, and caspase-12 following cerebral ischemia in rats

Objective: To observe the expressions of glucose-regulated protein (GRP) 78, GRP94 and caspase-12 related with endoplasmic reticulum in the striatum during focal cerebral ischemia in rats, so as to investigate the possible effect of endoplasmic reticulum molecular chaperones and apoptosis factor in the ischemic process. Methods: Sixty rats were equally randomized into 2 groups (n = 30): The sham operated group and the ischemic group (focal transient cerebral ischemia model was established with intraluminal occlusion of left meddle cerebral artery). The expression of GRP78, GRP94 and caspase-12 in rats striatum was detected by immunohistochemistry staining and semiquantitative RT-PCR at the different time points (6, 12 and 24 h after ischemia). Results: Immunohistochemistry staining and RT-PCR results illustrated that the mRNAs and protein expressions of GRP78 and GRP94 in ischemic group were lower than that in sham operated group at each time point (P < 0.05). The expression of the GRP78 and GRP94 were highest at 12 h from 6 h to 24 h (P < 0.05) in ischemic group. The expression of caspase-12 in ischemic group upgrade at 6 h and were highest at 12 h and decreased at 24 h. There was no expression in sham operated group. Conclusion: Endoplasmic reticulum-self regulated system was started through the increased expressions of GRP78 and GRP94 after striatum ischemia in rats, so as to attenuated cerebral ischemic injury. Endoplasmic reticulum-self regulated function loss because of severe cerebral ischemia. Initiation of caspase-12 apoptosis pathway is possible one of the mechanisms that priming endoplasmic reticulum apoptosis pathway in cerebral ischemic injury.
3.Differential expression of matrix metalloproteinases and their tissue inhibitors in human primary cultured prostatic cells and malignant prostate cell lines

AIM: To investigate the expression of matrix metalloproteinases (MMPs), membrane type MMPs (MT-MMPs) and their inhibitors (TIMPs) in human primary cultured prostatic cells and malignant prostate cell lines.METHODS: Reverse transcription-polymerase chain reaction-based measurements of the mRNA levels of MMP-2, MMP-7, MMP-9, MT1-MMP, MT3-MMP, TIMP-1 and TIMP-2 in relation to the house-keeping gene glyceraldehydes phosphate dehydrogenase were performed in cancerous and non-cancerous prostatic tissue samples, in primary cell cultures of epithelial cells, in both fibroblasts and smooth-muscle cells as stromal cells, and in the human malignant prostatic cell lines DU-145, LNCaP and PC-3.RESULTS: MMP-2 was mainly expressed in stromal cells. MMP-7 and MMP-9 showed their highest values in epithelial cells. MT1-MMP, MT3-MMP, TIMP-1 and TIMP-2 were found both in stromal and in epithelial cells, but there were some differences between the expression in fibroblasts and smooth-muscle cells. Different expression was also observed between the cells deriving from the primary cell cultures, the benign cell line BPH-1, and the malignant cell lines LNCaP, DU-145, and PC-3.CONCLUSION: These results concerning different expression of MMPs and TIMPs in cells from prostatic tissue suggest that a better insight into changes observed in prostatic tissue needs studies on cells cultured from the tissue.
4.Nursing management in the treatment of large number of infants with urinary calculi

To explore the measures of nursing management in the treatment of large number of infants with urinary calculi. The nursing management measures included launching the preparedness and response project for sudden public health events, formulating scientific and standardized nursing management system,optimizing work flow,strengthening nurse training,focusing on the details in nursing management,implementing disinfection and isolation system seriously,and paying close attention to health education for the parents of minority infants. Scientific nursing management can ensure the treatment effectiveness and nursing safety for the infants with urinary calculi.
5.Screening Value of Color Doppler Flow Imaging for Gastroesophageal Reflux

Objective To study the screening value of color doppler flow imaging (CDFI) for gastroesophageal reflux(GER).Methods Through the window of left lobe liver, the abdominal esophageal length,the phenomenon of GER and the frequency of GER were detected by CDFI in 55 children with GER and 55 control group.Results Abdominal esophagus was identified by CDFI in every children. The abdominal esophageal length was shorter in refluxers than that in control group. A significant correlation was found between its length and the age of control group.To diagnose GER with CDFI ,its accuracy was 98.18%,and its specificity was 76.36%.Conclusions Visualization and measurements of the abdominal esophagus are readily achieved with CDFI in children.Abdominal esophageal length is shorter in refluxers than that in control group. CDFI is a rapid method of screening GER.

10.Expression of matrix metrallproteinase-2 in human tears fluid after LASlK

AlM: To monitor long - term changes of matrix metalloproteinase-2 ( MMP-2 ) in human tears fluid after laser in situ keratomileusis ( LASlK) .
METHODS: Thirty - two myopia cases ( 64 eyes ) underwent uneventful LASlK were enrolled in the study. Tear fluid were collected and MMP-2 expression was analyzed by Western - bolt assay preoperatively and postoperatively on 15d, at 1, 3mo, and 1a.
RESULTS:LASlK increased the concentration of MMP-2 in human tear fluid. At 15d postoperatively, the magnitude of MMP-2 was 1. 4 times that of preoperative, thereafter subsided, but didn't return to preoperative level by 3mo ( P < 0. 05 ). Up to 1a after surgery, the concentration of MMP-2 almost recovered (P>0. 05).
CONCLUSlON: MMP- 2 is significantly expressed in human tear fluid after LASlK, then subsided with time, but didn't return to preoperative level by 3mo and almost recovered up to 1a, indicating wound healing of LASlK would continue up at least 3mo after surgery and almost recovered 1a postoperatively.
