International Journal of Laboratory Medicine 2010;31(5):444-446
Objective To investigate the distribution and the characteristics of HBV genotype in Kunming City,China. Methods The HBV genotypes and HBV-DNA levels of 360 patients in Kunming were detected by real-time quantitative PCR using type-specific primers, the serum-types were also detected in ELISA at the same time. Results Of the 360 specimens,346(96.11%) could be detected directly, among them 196 were genotype C(56.65%),91 were genotype B(26.30%),91 were genotype B(26.30%),37 were in B-C mix-type (10.69%),21 were genotype D(6.07%),1 were genotype A(0.29%).Among them, the genotype C was higer than the others (P<0.01);HBV-DNA levels of the patients with genotype C and B-C mix-type were higer than the genotype B and the others(P<0.01,P<0.05) (beside the positive proportions of genotype B was lower in the serum with"HB-sAg( + ),HBeAg( + ),anti-HBc( + )"(P<0.01),the other genotypes were no relation with their serum-types (P>0.05);the 360 specimens,14(3.89%)could not be detected directly, the average level of them was 4.136X 103IU/ml,it was lower than that of others(P<0.05).Conclusion The HBV-genotypes in Kunming area are abundant, including genotype C,B,D,A and B/C mix-type.The dominant genotype of HBV are C and B,and the HBV-DNA level of genotypes B/C mix-type and C were singnificantly higher than that of others ;The negative relation existed between the genotype B's frequency and the positive proportions of HBeAg;The genotype D frequency of this region is higher than that of lower-altitude inland areas and lower than that of plateau areas around Lahsa, which probably stands for its epidemiologic feature.Meanwhile, those samples without direct identification contain relatively lower level of HBV-DNA,which probably suggests that HBV genotype should be conducted on a basis of a certain HBV-DNA quantity.

Objective:To investigate the clinical effects of health and safety management in elderly patients receiving gastroenteroscopy under sedation. Methods:From February 2018 to November 2019, 126 elderly patients admitted to the cadre medicine and gerontology department of our hospital to undergo gastroenteroscopy under sedation were included as research subjects.According to the digital random number method, they were divided into the control group and the test group(n=63, each group). The control group was given routine management, while the test group was given routine management plus health and safety management.Clinical effects and safety were analyzed and compared between the two groups. Results:There was no significant difference in scores of the Hamilton anxiety scale(HAMA)and the Hamilton depression scale(HAMD)between the two groups before intervention. After intervention, the scores of HAMA and HAMD were lower in the test group than in the control group[(39.2±3.1)scores vs.(48.6±2.1)scores, (40.1±4.1)scores vs.(52.6±5.5)scores, t=19.926 and 14.463, both P=0.000]. The incidence of postoperative complications was lower in the test group than in the control group(7.94% or 5/63 vs.19.05% or 12/65, χ2=14.209, P=0.002). The differences between maximum and minimum values of heart rate(HR), diastolic blood pressure(DBP), respiratory rate(RR)and systolic blood pressure(SBP)were lower in the test group than in the control group(all P<0.05). The length of the procedure was shorter and the anesthetic dose was lower in the test group than in the control group(9.11±2.56 min vs.13.56±3.05 min, 15.35±2.66 ml vs.21.67±6.15 ml, t=8.870 and 7.486, both P=0.000). Patient satisfaction was higher in the test group than in the control group( χ2=16.383, P=0.014). Conclusions:Health and safety management can improve emotional health, stabilize vital signs and enhance satisfaction in elderly patients undergoing gastroenteroscopy under sedation.

Journal of Third Military Medical University 2003;0(20):-
Objective To investigate the effects of GDNF-modified Schwann cells on the repair of sciatic nerve defects in rats. Methods Forty adult Wistar rats were randomly divided into four groups: group A (nerve grafts, fabricated by extracellular gel and PLGA conduits, n=10), group B (Schwann cells and extracellular gel and PLGA conduits, n=10), group C (GDNF-modified Schwann cells and extracellular gel and PLGA conduits, n=10), and group D (autograft, n=10). Motoneuron conduction and morphology (including anatomical measures and ultrastructure) of the regenerated axons were investigated at 12 weeks postoperatively. Regeneration rate of the motoneuron in spinal cord was also evaluated by using HRP label technique. Results Higher rate of nerve regeneration or number of labeled motoneurons in spinal cord was observed using PLGA conduits seeded with GDNF enhanced expressing Schwann cells (group C) than that with PLGA conduits (group A) alone or PLGA conduits seeded with control Schwann cells (group B), but there was no significant difference as compared with that in the autograft group (group D). Conclusion These results suggest that using gene transfer techniques to increase neurotrophic factor expression in Schwann cells added to nerve grafts may be a promising method for improving nerve regeneration.

Objective To establish the electric controlled cortical impact (eCCI)-induced traumatic brain injury (TBI) model in rats with different severity in degree, which may serve as a suitable platform to provide experimental evidence for the pathophysiological following TBI.Methods A total of 40 male Wistar rats were randomly divided into 3 experimental groups and sham group.TBI rats (n=10/group) were positioned beneath the controlled cortical impactor device (eCCI) and subjected to impact injury at 2 mm depth of penetration, for a sustained depression of 200 ms,at 4 m/s,5 m/s,6 m/s velocity for mild, moderate, and severe TBI,respectively.Sham-operated rats (n=10) underwent identical surgical procedures, including craniotomy, without receiving the cortical impact.Neurological function and regional cerebral flow (24 h after CCI),contusion volume, histopathological, and ultrastructural changes (48 h after CCI) were measured, respectively. Results The severity of the pathological changes in rats was increased as the injury aggravated. The eCCI device impacted the brain at 4 m/s,5 m/s,6 m/s velocity for mild, moderate, and severe TBI,respectively.TBI groups showed impaired neurological function, and decreased rCBF lower than that of sham-operated group (all P<0.01).Furthermore,neuronal pathological abnormalities in TBI groups, including neuron shrinking, perineuronal vacuole, and structural abnormalities of mitochondria.Increased severity of injury was apparent following the increased level of the impacted velocity, and significant differences were observed between TBI groups (P<0.05).Conclusion The TBI animal model with mild, moderate, and severe brain injury can be established successfully by 4 m/s,5 m/s,and 6 m/s of impact velocity respectively with the eCCI-6.3 device. The novel eCCI-induced TBI model in rats possibly serves as a novel useful approach in the development of TBI models.
