Amplified in breast cancer 3 (AIB3) is a newly discovered pleiotropic nuclear receptor co-activator.AIB3 regulates cell proliferation,growth,migration and apoptosis by interacting with many nuclear receptors.At the mean time,some reports show that AIB3 interacts with variety of transcription factors,which indicates AIB3 may play an important role in cancer occurrence and progression.
2.Advancement in surgical treatment of intrahepatic cholangiocarcinoma

Intrahepatic cholangiocarcinoma (ICC) has the significant characteristics of regional spread and lymphatic metastasis. Radical resection provides the only chance for long-term survival of patients with ICC. Anatomic or extended hepatectomy is the most optimal treatment. The value of hepatic resection with lymph node dissection and the area of lymph node dissection are controversial. Palliative resection improves prognosis of some patients. Secondary hepatectomy also benefits some patients with late (>1 year) intrahepatic recurrence. Liver transplantation is only performed for highly selected prophase-patients or prudent clinical trials, and a combination with adjuvant treatment such as chemo-radiotherapy after operation is suggested.
3.Simultaneous ion pair RP-HPLC determination of oxymatrine and matrine in Compound Kushen Injection

AIM: A RP HPLC was developed to determine the contents of oxymatrine and matrine simultaneously in Compound Kushen Injection(Radix Sophorae Flavescentis, Rhizoma Heterosmilacis Japonicae). METHODS: The analysis was carried out on a column of Hypersil ODS 2(4.6mm?200mm,5?m) with a mobile phase of methanol acetonitrile water phosphate acid(10∶30∶65∶0.05,33mmol?L -1 ), flow rate being 1.0mL?min -1 , detection wavelength being at 210nm and the column temperature at room temperature. RESULTS: The linear range was in the range of 17.44~174.4?g?mL -1 ( r =0.9996, n =6) for oxymatrine and in the range of 1.96~ 39.20 ?g?mL -1 ( r=0.9997,n =6) for matrine, respectively. The average recoveries of oxymatrine and matrine were 100.4%( RSD =2.1%, n =9) and 99.7%( RSD =1.4%, n =9), respectively. CONCLUSION: The method is found to be simple and accurate for simultaneous analysis of oxymatrine and matrine in Compound Kushen Injection, and a reliable way to quality evaluation.
4.Application of 13 High-risk HPV infection Test Combined with Thinprep Cytologic Test on Screening Cervical Carcinoma in Dali Region, Yunnan Province

Objective To investigate relativity between the epidemiology of HPV and cervical carcinoma in Dali region,Yunnan province,through detecting the 13 high-risk human papillomavirus infection and Thinprep cytologic test in 2153 cases.Methods Real-time PCR was used to detect the 13 high-risk HPV (16,18,31,33,35,39,45,51,52,56,58,59,68) in2153 cases and 1604 cases were checked with Thinprep cytologic test.Results In 2153 samples,260 cases were infected with HPV,with the positive rate of 12.08%.The highest positive rates were >60 years old (18.18%),then >20 and ≤30 years old (14.41%);there was no significant difference in the positive rate among the various age groups (P =0.384).There were 1465 negative for intraepithelial lesion ormalignancy (NILM) cases (91.33%),86 atypical squamous cells of undetermined significance (ASC-US) and atypical squamous cells cannot exclude HSIL (ASC-H) cases (5.36%),32 low-grade squamous intraepithelial lesion cases (LSIL) cases (2.00%),21 high-grade squamous intraepithelial lesion cases (1.31%) through Thinprep cytologic test.The correlation coefficient is 0.893.Conclusions The infection rate of HPV in Dali region,Yunnan Province,has no significant difference among the various age groups.Application of 13 high-risk HPV infection test combined with Thinprep cytologic test could be more effective in screening cervical carcinoma.
5.Clinical application of the combined the anterior malleolus flap and anterior tibia flap

Objective To investigate the clinical application of the combined the anterior malleolus flap and anterior tibia flap. Methods Based on the dissection of the perforating branches of anterior tibial artery on the middle and inferior section, the combined the anterior malleolus flap and anterior tibia flap was designed to repair the necrotic skin of anterior foot for 5 patients. The sizes of the flaps ranged from 17 cm×10 cm-10 cm× 5 cm. And the area of the flap was from tibial tuberosity(upper bound) to the line between internal malleolus and external malleolus (lower bound), and from the median line of one side of leg to the other side. Results Postoperatively, all flaps survived, and the primary healing of transplanted skin in donor site was achieved. The texture of flaps were excellent, the phenomenon of abrasion did not happen, and the clinical therapeutic efficacy was satisfactory after a follow up of 2-24 months. Conclusion It's a good method that combined the anterior malleohls flap and anterior tibia flap, which not only could enlarge the area of the flap but also has reliable blood supply, in repair of large size skin defect of anterior foot.
7.Laparoendoscopic single-site radical cystectomy and urinary diversion: initial experience using homemade single-port device

Objective We report our initial experience with laparoendoscopic single-site surgery (LESS) for radical cystectomy and urinary diversion performed by a single surgeon using homemade singleport device.Methods From December 2010 and April 2011,7 LESS radical cystectomy were performed using a home-made single-port device composed of an inverted cone device of polycarbonate and a powderfree surgical glove.7 patients were all male,mean age was 66 (54-81) years and mean BMI (Body mass index) was 23.1 (20.9-25.3) kg/m2.The port was placed into a 5 cm periumbilical skin incision.The conventional laparoscope and laparoscopic instruments were inserted through the single-port.No additional ports were needed for radical cystectomy and bilateral standard pelvic lymphadenectomy.Cutaneous ureterostomy (3 cases) and ileal conduit urinary diversion (4 cases) were used for patients.Operative time,estimated blood loss,postoperative recovering time,oncologic data and complications were collected and analyzed.Results All the procedures were completed successfully.The mean operative time was 210.1 (155-280) minutes.The estimated blood loss was 300 (100-500) ml.The bowel recovering time 8 (4-12) days and postoperative hospital stay was 20 (13-34) days.One patient required a transfusion of 400 ml red blood cells.The pathologic evaluation revealed that there were one case of pT1N0M0,two of pT2aN0M0,two of pT2bN0M0 and two of pT3aN0M0.The high grade tumor in 4 cases and low grade in 3 cases.The surgical margins were negative in all the patients.All patients were node negative.After the operations,one case had a small bowel obstruction after three days and was treated by abrosia,fluid infusion and gastrointestinal decompression.Another patient died of cardiac disease at first day postoperative.Conclusions In our experience,LESS for radical cystectomy could be clinically feasible for selected patients,but it requires the learning curve.
8.Correlation of cholangiocarcinoma and K-ras gene mutation of bile duct tissues

Objective To explore the relationship between K-ras gene and cholangiocarcinoma by detecting the K-ras gene mutation of bile duct tissues.Method We studied all the patients who presented to our hospital from June 2010 to June 2012 with stenosis of the bile duct.There were 17 cases of cholangiocarcinoma and 17 cases of benign stenosis.From the DNA extracted from the paraffin tissues,we used the HRM assay for K-ras gene mutation.Result We found that the HRM method and the DNA typing had exactly the same result for the DNA content which confirmed the effectiveness of the HRM assay.Of note,the K-ras mutation rate was found to be significantly higher in the cholangiocarcinoma cases (11/17) when compared with the benign cases (3/17).Conclusion The mutation of the K-ras gene was closely related to cholangiocarcinoma.Our results suggest a new way to diagnosis cholangiocarcinoma.
9.The impact of using a Swiss ball to exercise the lumbo-abdominal muscles

Objective To explore any changes in surface electromyogram (sEMG) signals from the lumboabdominal muscles during exercise on stable and unstable surfaces. Methods sEMG signals from the lumbo-abdominal muscles of 33 healthy young persons [18 male and 12 female; average age (26.5 ± 4.3 ) years] were measured with the FlexComp Infiniti apparatus.Each subject performed 5 exercises on and off a Swiss ball:sit,bridge,bridge with both knees flexed,reverse bridge as well as press-up. Results ①In bridging there was a significant increase in the activation of the erector spinae during exercise on the ball compared with on the stable surface.②Bridging with both knees flexed gave a significant increase in activations of the erector spinae,the external obliques and the transverses abdominus/internal obliques during exercise on the unstable surface compared with the stable surface.③During reverse bridging there was a significant increase in activation of the erector spinae and rectus abdominus during exercise on the unstable surface compared with the stable surface.④During press-ups there was a significant increase in activation of the rectus abdominus,the external obliques and the transverses abdominus/internal obliques during exercise on the ball compared with the stable surface. Conclusion The unstable surface provides better training stimulus for the activation of the lumbo-abdominal muscles.
10.Expression and purification of spike protein of severe acute respiratory syndrome coronavirus in Saccharomyces cerevisiae

Objective To construct the recombinant plasmid pYES6-S and express and purify spike protein of severe acute respiratory syndrome(SANS)coronavirus in Saccharomyces cerevisiae. Methods DNA fragments of SANS coronavirus were obtained by reverse transeription.Four over- lapped fragments of spike protein genes were amplified by polymerase chain reaction(PCR)and ligated into an integral spike protein gene by restriction enzyme digestion.The spike protein gene recombined with pYES6 and cloned into E.coll.The recombinant plasmid pYES6-S was induced and expressed in Saccharomyces cerevisiae(INVScl)by galactose.Results The recombinant plasmid pYES6-S was confirmed that inserted fragment was right in length,direction and base matching by restriction enzyme digestion and sequencing.The purified protein encoded by the whole spike protein gene was about Mr 110?10~3 identified by electrophoresis.Conclusion The whole spike protein gene of SARS coronavirus is cloned into E.coli and the protein is expressed in Saccharomyces cerevisiae successful ly.which can be helpful in SARS vaccine research.
