1.Homozygous deletions of MTS1/p16 gene in laryngeal squamous cell carcinoma

To study the status of homozygous deletions of MTS1/p16 gene in squamous cell carcinoma of the larynx (LSCC) and to discuss the relations of homozygous deletions to the pathological grade and clinical stage of the tumor,Fifty-nine primary LSCC were examined for homozygous deletions of the MTS1/p16 gene by comparative PCR.Three specimens were eliminated for being incapable of comparison with others.Homozygous deletions of the MTS1/p16 gene were found in 9 of 56 tumors (16.07%).The rates of homozygous deletions of MTS1/p16 gene in well,medially,poorly differentiated tumors were 17.24%(5/29),11.76%(2/17),and 20.00%(2/10) respectively.There had no significant difference in various pathological grades of tumors.Homozygous deletions rates of tumors in stagesⅠ~Ⅱand in stagesⅢ~Ⅳwere 5.41% (2/37) and 36.84%(7/19) respectively.The rate of homozygous deletions in stages Ⅲ~Ⅳtumors was significantly higher than that in stagesⅠ~Ⅱ(P<0.01).Homozygous deletions of the MTS1/p16 gene were correlated well with clinical stages.Our data suggested that homozygous deletions of MTS1/p16 gene might be one of the genetic events in the development of the tumor and might play a role in malignant progression of LSCC.
2.A domestic cat infected with a large number of Clonorchis sinensis

Adult Clonorchis sinensis not only occurs in human hepatic duct,but also in the liver,gallbladder and bile ducts of animals,including dogs and cats,thus causing clonorchiasis—one of important parasitic zoonoses. In present study,we dis?sected a domestic cat in which a total of 736 pieces of trematodes,identified as Clonorchis sinensis,were detected in the liver and cholecyst. The findings indicate that Clonorchis sinensis may be endemic in domestic animals in Wuhu area,and observe our awareness in prevention of the parasites in house pets.
3.Comparisons of clinical characteristics of primary Sj gren's syndrome onset at different ages

Objective To study the difference in clinical characteristics and experimental profiles of primary Sj gren's syndrome(pSS)with a different onset age.Methods The clinical and laboratory data of 136 pSS patients were retrospectively analyzed.Results The prevalence of anti-SSA and anti-SSB antibodies de- clined with later onset.As compared with patients with disease onset older than 45,patients with disease onset younger than 45 had higher rheumatoid factor,circulating immune complex and lower C3.The prevalence of parotitis was higher in patients with disease onset younger than 45 than those with disease onset older than 45. The two groups had similar extra-glandular involvement.Conclusion The younger onset pSS patients have more abnomlalities in inmmnological parameters,and more parotiditis,implying that young onset pSS have more strong immune responses.
4.The impact of angiogenic and adipogenic microenvironment on adipose tissue regeneration in tissue engineering chamber.

OBJECTIVEBy observing the adipogenic and angiogenic microenvironment impact on the morphology of newly generated tissue for exploring the key factors which inducing mature adipose tissue regeneration in tissue engineering model.
METHODS24 healthy 6 months' New Zealand rabbits were picked and put into four groups according to different microenvironment. Every group has 6 rabbits and divided as follows: no axial-blood supply fat flap(0 ml), granular fat only(0.6 ml), axial blood vessel only (0.05 ml), axial vascularized fat flap ((0.6 ml). We separated or combined adipogenic and angiogenic environment within these groups. After 8 weeks, samples were harvested for histologic observation including macroscopic observation, volume analysis and HE testing.
RESULTSIn granular fat group, its volume decreased by (0.25 ± 0.10) ml after 8 weeks as the shortage of blood supply and finally it could be enveloped. In axial blood vessel group, its volume increased by (0. 37 ± 0. 04) ml after 8 weeks with fibrous tissue formation as shortage of adipogenic microenvironment. The no axial-blood supply fat flap group grew into(0.12 ± 0.03) ml, which can' t support large volume adipose tissue formation because of lacking independent blood supply. Only axial vascularized fat flap model could generate mature adipose tissue in large volume(3.45 ± 0.48) ml. The number of new capillary in every group was different after 8 weeks. By counting the numbers in every single view, no axial-blood supply fat flap group 15 ± 3.5)and granular fat only group(5 ± 2.5)had a significant difference with axial vascularized fat flap group 22 ± 5) respectively.
CONCLUSIONOnly both adipogenic or angiogenic microenvironment exist could induce mature adipose tissue in large volume in tissue engineering chamber model.

5.Study on Histiostomas apromyzarum breeding in traditional Chinese medi-cine Rhizoma bletillae

Objective To investigate the Histiostomas apromyzarum breeding in traditional Chinese medicine Rhizoma Bletillae. Methods The scurf and dust of R. bletillae were obtained by brushing,and the mites samples were separated and identified. Results Totally 520 g samples were collected,and 56 adult mites(female?male ratio 0.12)and 18 dormant bodies were obtained from the samples,and they were identified as H. apromyzarum with an average breeding density of 0.14/g. Con?clusion H. apromyzarum could breed on the traditional Chinese medicine R. bletillae,and the corresponding control mea?sures should be strengthened.
6.EGFR inhibitor enhances cisplatin sensitivity of human glioma cells.

Epidermal growth factor receptor (EGFR) is found to express at high levels in a variety of solid tumors including gliomas. This study was to examine the effect of an EGFR-tyrosine kinase inhibitor (AG1478) alone or in combination with cisplatin (CDDP) on the growth of glioma cells (U87). U87 glioma cells were treated with AG1478 (10 μmol/L) or CDDP (25 μmol/L) as a single agent or in combination for 24 or 48 h. The expression of EGFR and the components in its downstream signaling pathway [extracellular signal-regulated kinase (ERK), protein kinase B (AKT)] in U87 glioma cells was detected by Western blotting. Cell growth, cell cycle distribution and cell apoptosis were determined by MTT method and flow cytometry, respectively. The results showed that CDDP could induce the activation of EGFR and the components in its downstream signaling pathways in a concentration-dependent manner. The combined treatment of AG1478 with CDDP could inhibit the proliferation of U87 glioma cells, arrest the cell cycle and promote cell apoptosis. In the EGFR signaling pathway, AG1478 decreased the phosphorylation of ERK, AKT and EGFR in U87 glioma cells. It was concluded that the combined treatment of AG1478 and CDDP may exert synergistic inhibitory effects on the growth of glioma cells by suppressing the activities of EGFR, AKT and ERK.
7.Detection of Low-abundance Point Mutations by Competitive Strand Assisted Endonuclease Ⅳ Signal Amplification System

Genetic mutations are important molecular biomarkers for cancer diagnosis and surveillance.Therefore,the development of methods for mutation detection characterized with straightforward,highly specific and sensitive to low-level mutations within various sequence contexts is extremely needed.Although some of the currently available methods have shown very encouraging results,their discrimination efficiency is still very low.Herein,we demonstrate a fluorescent probe coupled with blocker and property of melting temperature discrimination,which is able to identify the presence of known or unknown single-base variations at abundances down to 0.1% within 20 min.The discrimination factors between the perfect-match target and single-base mismatched target are determined to be 10.15-38.48.The method is sequence independent,which assures a wide range of application.The new method would be an ideal choice for high-throughput in vitro diagnosis and precise clinical treatment.
8.Cloning and functional characterization of phytoene desaturase in Andrographis paniculata.

A full-length cDNA of phytoene desaturase (PDS) gene from Andrographis paniculata was obtained through RACE-PCR. The cDNA sequence consists of 2 224 bp with an intact ORF of 1 752 bp (GeneBank: KP982892), encoding a ploypeptide of 584 amino acids. Homology analysis showed that the deduced protein has extensive sequence similarities to PDS from other plants, and contains a conserved NAD ( H) -binding domain of plant dehydrase cofactor binding-domain in N-terminal. Phylogenetic analysis demonstrated that ApPDS was more related to PDS of Sesamum indicum and Pogostemon cablin. The semi-quantitative RT-PCR analysis revealed that ApPDS expressed in whole aboveground tissues with the highest expression in leaves. Virus induced gene silencing (VIGS) was performed to characterize the functional of ApPDS in planta. Significant photobleaching was not observed in infiltrated leaves, while the PDS gene has been down-regulated significantly at the yellowish area. To the best of our knowledge, this represents the first report of PDS gene cloning and functional characterization from A. paniculata, which lays the foundation for further investigation of new genes, especially that correlative to andrographolide biosynthetic pathway.

9.The expression level of serum IL-7 and its influence on prognosis in patients with acute coronary syndrome

Objective: To explore the expression level of serum interleukin (IL)-7 in patients with acute coronary syndrome (ACS) and analyze the relationship between IL-7 level and prognosis. Methods: A total of 130 ACS patients [ACS group, including 70 cases with acute myocardial infarction (AMI) and 60 cases with unstable angina pectoris (UAP)], 33 cases with stable angina pectoris (SAP,SAP group) and 89 healthy subjects (healthy control group) were selected. IL-7 level was measured using enzyme linked immunosorbent assay (ELISA) and compared among all groups. The 130 ACS patients were followed up, and Logistic regression analysis was used to analyze the relationship between IL-7 level and prognosis. Results: Compared with healthy control group and SAP group, there was significant rise in IL-7 level in UAP group and AMI group [(1.84±0.47) pg/ml, (2.11±0.63) pg/ml vs. (4.87±0.52) pg/ml, (5.15±0.71) pg/ml, P<0.05 or P<0.01]. There were no significant difference in IL-7 level between healthy control group and SAP group, UAP group and AMI group (P>0.05 both); Logistic regression analysis indicated that expression level of serum IL-7 was an independent risk factor for adverse cardiovascular events in ACS patients (OR=1.212, 95%CI:1.061-1.418). Conclusion: Interleukin-7, as an important inflammatory cytokines, its serum level abnormally elevated in patients with acute coronary syndrome, it may have important prognostic value.
10.Analysis of effects of arthroscopic treatment for recurrent patellar dislocation by lateral patellar retinacular release outside the synovial bursa of knee joint combined with reconstruction of medial patellofemoral ligament.

OBJECTIVETo introduce an arthroscopic technique in managing recurrent dislocation of the patella and its clinical results.
METHODSSixteen patients with recurrent patellar dislocation were reviewed, including 3 males and 13 females. The average age was 17.6 years old (ranged from 14 to 32 years). The patients suffering from patellar sub-luxation averaged 18.5 months (ranged from 6 to 23 months)before operation. These patients were treated with lateral patellar retinacular release outside the synovial bursa of knee joint and medial patellofemoral ligament reconstruction using the semitendinosus tendon free autograft. The Lyshohm scores before and after operation were used to evaluate outcomes at the final follow-up.
RESULTSAll the patients were followed up, and the duration ranged from 6 to 48 months, with an average of 12 months. There was no recurrence. The Q angle decreased from (16.4 ± 3.7)° to (10.1 ± 1.4)°; insall index decreased from 1.37 ± 0.25 to 1.28 ± 0.23; congruence angle decreased from (21.3 ± 2.6)° to (5.86 ± 2.23)°; Lysholm score improved from 76.1 ± 5.2 to 89.8 ± 4.1 at 6 months after operation.
CONCLUSIONCompared with conventional procedure, arthroscopic surgery for recurrent dislocation of the patella achieves excellent outcomes with minimum invasion.
