2.A Randomized Controlled Clinical Study on Prevention and Treatment for Acute Radiation Esophagitis by Compound Kushen Injection

Objective To investigate the prevention and treatment effects of Compound Kushen Injection on acute radiation esophagitis. Methods Eighty-two eligible patients with esophageal cancer were randomly divided into the treatment group (41 cases) and the control group (41 cases). All the patients received radiotherapy. Throughout the course of radiotherapy, patients in the treatment group received Compound Kushen Injection, and patients in the control group received Kangfuxin Liquid. Occurrence time and level of radiation esophagitis, and dosage of painkillers were observed. Results Different degrees of acute esophageal toxicity were observed in the two groups. The occurrence rate of high level (degree III and degree IV) acute radiation esophagitis was 7.3%(3/41) in the treatment group, and 31.7%(13/41) in the control group. There was significant difference between the two groups (P<0.05). The dosage of the analgesic drug (Fentanyl Transdermal System) in the treatment group was far less than the controlled group (P<0.001). Conclusion Compound Kushen Injection could decrease the incidence rate of acute radiation esophagitis, and reduce the high-level esophagitis and the dosage of the analgesic drug, which can help the completion of radiation.
3.Immune response of human immunodeficiency virus-1 specific T lymphocytes in human immunodeficiency virus-1 monoinfected or human immunodeficiency virus-1/hepatitis C virus coinfected individuals

Objective To investigate the features of immune response of human immunodeficiency virus type-1 (HIV-1) antigen specific T lymphocytes in HIV-1 monoinfected or HIV 1/hepatitis C virus (HCV) coinfected individuals.Methods Twenty-six HIV-1 monoinfected and 23 HIV-1/HCV coinfected individuals were enrolled.Immunomagnetic microbeads were used to isolate T lymphocyte subpopulation CD4+ T cells and CD8+ T cells from human peripheral blood mononuclear cells (PBMC).Frequencies of interferon-γ (IFN-γ) secreting cells of CD4+,CD8+ T lymphocytes and PBMC stimulated by a peptide pool containing 12 overlapping peptides in HIV-1 P24 from 49 patients were assessed by enzyme-linked immunospot (ELISPOT) assay.HIV-1 RNA levels of these patients were also detected by real-time fluorescence quantitative polymerase chain reaction.The data were compared by one-way ANOVA and Mann-Whitney U test,and Spearman test was used for correlation analysis.Results Frequencies of HIV-1 antigen specific CD4+ T lymphocytes [median =25 spot-forming cells (SFC)/106 cells] were significantly lower than those of CD8+T lymphocytes (median=38SFC/106 cells,F=4.592,P=0.037) and PBMC (median=53 SFC/106 cells,F=5.436,P=0.025) in HIV-1 monoinfected group.Frequencies of HIV-1 antigen specific CD4+ T lymphocytes (median=5 SFC/106 cells,Z=-2.432,P=0.015),CD8+T lymphocytes (median=5 SFC/106 cells,Z=-1.996,P=0.046) and PBMC (median=10 SFC/106 cells,Z=-2.306,P=0.021) in HIV-1/HCV coinfected group were significantly lower than those in HIV-1 monoinfected group.Conclusions In HIV-1 infection,antigen specific immune response of CD4+ T cells can be activated,but weaker than that of CD8+ T cells.Co-infection with HCV might down-regulate the responses of HIV-1 antigen specific T lymphocytes in HIV-1 infected individuals.
4.MRI study on predicting the collapse of avascular necrosis of the femoral head

Objective To study the risk factors of MRI for the prediction of collapse in patients with avascular necrosis of the femoral head.Methods Twenty-two patients (39 hips) diagnosed avascular necrosis of femoral head by MR were enrolled in our study.The following MR appearances were evaluated:bone marrow edema,joint fluids,signal intensity and location of the lesion.The volume and surface area of the necrosis zone were calculated.The time of follow-up was 18-84 months (median,25 months).Logistic regression analysis was used to predict the risk factors by SPSS 13.0.The maximum value of Youden index was selected as the critical point to predict the collapse of femoral head and to define the sensitivity,specificity and accuracy.Results In the 39 hips with femoral head necrosis,21 hips had collapse.Bilateral collapse occurred in 5 cases.In 25 hips with the necrosis surface larger than 25%,collapse occurred in 21 (84%); In 8 hips with the volume of femoral head necrosis larger than 30%,collapse occurred in all cases; 1n 33 hips with the necrosis locating at the superolateral quadrant,collapse occurred in 21 (63.6%); In 22 hips with necrotic areas showing heterogeneous signal intensity,collapse occurred in 18(81.8%) ;In 25 hips with large amount of joint effusion,collapse occurred in 16 (64%) ;in 18 hips with bone marrow edema,collapse occurred in 13 (65%).Joint fluid,heterogeneous signal intensity and lesions in the superolateral quadrant,volume ratio,and area ratio were the high risk factors,while bone marrow edema was a relatively low risk factor.The area under ROC curves for area ratio of NASA was greater than that for volume ratio (0.987 vs 0.902).When the critical value for area ratio was 26.7%,the true positive rate was 95.2%,true negative rate was 94.4%,and Youden's index was 0.896.Conclusions The collapse of necrosis of femoral head may result from many factors.The femoral head was easy to collapse when it had large enough area of necrosis and mixed signal intensity,a large amount of joint effusion,bone marrow edema,and superolateral quadrant location.The critical value for area ratio to predict the collapse of femoral head was about 26.7%.The area ratio is more accurate than volume ratio in predicting the collapse of necrosis of femoral head.
5.Research on the tumor suppression of Chinese caterpillar fungus water decoction combined cisplatin on tumor burdened mice

Objective To study the tumor suppression of Chinese caterpillar fungus water decoction combined cisplatin on a tumor burdened mice .Methods The beforehand vaccinal HCT8 tumor burdened mice were randomly devided into 5 groups(n=10) ,then 3 groups of Chinese caterpillar fungus water decoction were given a tumor burdened mice stomach (2 .5 ,5 .0 ,10 g/kg) ,1 times a day ,12 d .1 group of mice were given saline .1 group were given cisplatin .Determination of different concentrations of cordyceps water decoction lavage ,then observe the changes of tumor‐burdened bone tumors in mice weight .Results For vaccination HCT8 tumor burdened bone tumors had an average weight of each dose group mice compared with saline group have significant difference(P<0 .01) ,while Chinese caterpillar fungus water decoction (2 .5 ,5 .0 g/kg) and cisplatin group mice tumor block is more significant difference(P< 0 .01) ,the tumors had the lighter weight .Conclusion Chinese caterpillar fungus water decoction has tumor suppression effect on tumor burdened mice .
6.RANTES, MCP-1, and SDF-1β levels in healthy people and AIDS patients: a comparative analysis

Objective To evaluate the differences of serum RANTES(regulated on activation, normal T cell expressed and secreted), MCP-1 (monocyte chemoattractant protein), and SDF-1β (stromal cell-derived factor-1β) in patients with acquired immunodeficiency syndrome (AIDS) and healthy people.Methods 38 AIDS patients who were admitted to a hospital between January 2010 and January 2015 were as AIDS groups, 38 healthy persons were as a healthy group, serum levels of RANTES, MCP-1, and SDF-1β in two groups were detected, and the subgroup analysis was carried out according to the viral load.Results Serum levels of RANTES, MCP-1, and SDF-1β in AIDS group were (1 392.55±227.69)pg/mL,(450.91±103.04)pg/mL, and(104.82±22.52)pg/mL respectively,all were significantly higher than those in healthy group([120.58±55.87] pg/mL, [74.25±33.62] pg/mL, and [39.04±11.43]pg/mL respectively)(all P<0.05).Among AIDS patients with HIV viral load 4≤Log(VL)<5 and Log(VL)≥5, serum RANTES were (1 470.34±155.01)pg/mL and (1 408.29±181.54)pg/mL respectively,which were both significantly higher than patients with HIV viral load Log(VL)<4([1 183.12±174.54]pg/mL);serum MCP-1 and SDF-1β levels in AIDS patients with HIV viral load 4≤Log (VL)<5 were (537.93±89.32)and(149.31±18.05)pg/mL respectively,which were significantly higher than patients with HIV viral load Log(VL)≥5([410.26±80.57] pg/mL, [81.53±20.31]pg/mL respectively) and HIV viral load Log(VL)<4([381.71±77.26] pg/mL, [72.90±21.62]pg/mL respectively), differences were both statistically significant(both P<0.05).Conclusion Serum levels of RANTES, MCP-1, and SDF-1β are significantly increased in AIDS patients, which are related to the level of viral load.

Objective To study the relationship between tyrosine phosphorylation(TP)and protein expression of insulin receptor substrate-1(IRS-1)and insulin resistance in patients with gestational diabetes mellitus(GDM).Methods IRS-1 expression and TP in skeleton muscle tissue were determined by Western blot and immunoprecipitation in women with GDM(GDM group,n=22),normal pregnant women(normal pregnancy group,n=22)and normal nonpregnant women(normal nonpregnant group,n=13).Fasting plasma glucose(FPG)and fasting insulin(FINS)were measured by oxidase assay and immunoradioassay. The osmotic pressure of ammonium sulfate solutions has been measured by the well-established freezing point osmometry in dilute solutions and we recently reported air humidity osmometry in a much wider range of concentration. Air humidity osmometry cross-validated the theoretical calculations of osmotic pressure based on the Pitzer model at high concentrations by two one-sided test (TOST) of equivalence with multiple testing corrections, where no other experimental method could serve as a reference for comparison. Although more strict equivalence criteria were established between the measurements of freezing point osmometry and the calculations based on the Pitzer model at low concentration, air humidity osmometry is the only currently available osmometry applicable to high concentration, serves as an economic addition to standard osmometry.
10.Effect of DC-CIK cells combined with oridonin on cytotoxicity against RPMI 8226 cells

Objective To investigate the changes in cytotoxicity of DC-CIK cells to human multiple myeloma RPMI 8226 cells before and after treatment with oridonin. Methods Normal human peripheral blood mononuclear cells were isolated and induced to obtain DC-CIK cells. Cytotoxicity of DC-CIK cells against RPMI 8226 cells which were treated by oridonin was analyzed by LDH releasing assay. The variation for expression of NKG2D ligands on RPMI 8226 cells were measured by flow cytometry. Results DC-CIK cells were successfully induced from the peripheral blood mononuclear cells. At the same effector to target ratio, oridonin obviously enhanced the cytotocixity of DC-CIK cells against RPMI 8226 cells (P<0.01). Flow cytometry showed the expression of NKG2D ligands ULBP1 of RPMI8226 cells was most significantly increased as the cells were treated by oridonin [(9.19 ± 1.85) vs. (15.47 ± 0.67), P<0.01]. Correlation analysis indicated that cytotocixity was positively correlated with changes in ULBP1. Conclusions Oridonin can improve the cytotoxicity of DC-CIK cells against RPMI 8226 cells, which may be related with the increased expressions of NKG2D ligands on the tumor cell surface.
