T cell immune deficiency results in inefficient anti-tumor immune response in most patients with hematological malignancies, thereby tumor cells escaping the attack of host immune system, that is the immune escape of tumor cells.Recently, new insights regard to the CD8+ T cell exhaustion which is one of the most important mechanisms of tumor immune suppression. The CD8 + T cell exhaustion is mediated by abnormal expression of T-cell immunosuppressive receptors such as PD-1 and CTLA-4.Blocking these molecules may restore the partial or all functions of T cells. This article reviewed the advances on the role of the PD-1 and CTLA-4 in hematological malignancies, including their functions on mediating T cell-immune tolerance and the progression of targeted immunotherapy,to provide the new strategy for hematological malignancies.

Objective To investigate the role of osteopontin (OPN) in the pathogenesis ot cholesterol gallstone formation in bile. Methods The nucleation time of OPN in model bile and human gallbladder bile was studied by the nucleation time assay, the effect of OPN on cholesterol crystal growth in model bile was examined by the cholesterol crystal growth assay. The effect of OPN on vesicle was detected by the transmission electron microscopy in model bile and gallbladder bile; then the content of OPN and calcium were detected via the commercial kits in human bile. Results Osteopontin prolonged nucleation time in a dose dependent manner in model bile and human bile, and this effect was correlated with calcium.Compared with control group, the nucleation times were prolonged by 1.50and 1.93 times in lithogenic bile at the concentration of osteopontin 50 μg/ml and 100 μg/ml (P<0.01),respectively. Nucleation time were prolonged by 1.17 and 1.33 times in normal bile (P<0.01) and by 1.29 and 1.48 times in model bile (P<0.01),respectively.The rate of cholesterol crystals growth was not influenced by calcium ions, but inhibited by osteopontin in a dose dependent manner in the model bile.Furthermore, the formation, aggregation and fusion of vesicles were delayed by osteopontin in bile samples as indicated by the transmission electron microscopy. The concentration of osteopontin [(0.53± 0.08) mg/ml vs. (0.65 ± 0.14) mg/ml,P<0.05] and the calcium ions [ (0.71 ± 0.17) mmol/L vs. ( 0.84 ± 0.08 ) mmol/L,P < 0.05 ] were lower in lithogenic bile than in control. Conclusions Osteopontin can inhibit the cholesterol gallstone formation in model and human gallbladder bile as the anti nucleating factor.

Objective To explore the therapeutic effect of hyperbaric oxygen therapy (HBO) in the treatment of neuropsychological disorders in senile patients with type 2 diabetes mellitus combined with cerebral infarction. Methods A total of 70 senile patients with type 2 diabetes mellitus combined with cerebral infarction were randomly divided into two groups: treatment group receiving HBO plus routine medicine treatment and control group receiving routing medicine treatment only. A suit of senile neuropsychological tests was administered pre and post treatment by an experienced psychometrician who was blinded to treatment group assignments. Results Neuropsychological disorders were improved partly in control group while the efficacy in treatment group was markedly improved(P<0.01). Spearman rank correlation analysis revealed that the improvement degree of neuropsychological disorders was positively correlated with hyperbaric oxygen therapy (HBO) (r= 0.502,P<0.05), the time of test before therapy (r=0.260, P<0.05) and the time of test after therapy (r=0.386, P<0.05), and was negatively correlated with hyperlipoidemia (r=-0.261, P< 0.05), glycosylated hemoglobin(GHbAlc) (r=-0.321,P<0.05) and body mass index(BMI) (r= 0.360,P<0.05). Multiple stepwise regression analysis showed that HBO and hyperlipoidemia were the independent predictors for the improvement degree of neuropsychological disorders. Conclusions HBO is an efficacious way to treat neuropsychological disorders in senile patients with type 2 diabetes mellitus combined with cerebral infarction.
