The viscoelastic properties of hepatocellular carcinoma (HCC) cells were measured by means of micropipette aspiration technique. Further,the authors studied the changes in viscoelastic coefficients by treating with colchicine and cytochalasin D. The results showed that the elastic coefficients of HCC cells were obviously higher than the corresponding value of hepatocytes. By treating with colchicine, the effects on viscoelastic properties of HCC cells were obviously different in ways and extents from those on viscoelastic properties of hepatocytes.,but the viscoelastic properties of hepatocytes by treated with cytochalasin D had the same trend of decreasing as those of HCC cells. These results represent the change in cytoskeleton structure and function among hepatocytes and HCC cells, this change might affect tumor cells invasion and metastasis.

3.Association study of dopamine D1 receptor gene and cognitive function of first-episode schizophrenic patients

Objective To investigate the relationship between cognitive function of first-episode schizophrenic patients and dopamine D1 receptor gene. Methods A total of 112 first-episode schizophrenic patients and 60 healthy controls were evaluated with Wechsler adult intelligence scale ( WAIS-R), Wechsler memory scale (WMS) and Wisconsin card sort test (WCST) ,and genotyped one polymorphism (rs4532) within DRD1 gene using TaqMan SNP genotyping assay. Results There were no significant differences on the frequencies of the genotypes and alleles of rs4532 polymorphism between patients with schizophrenia and normal controls ( x2 =2.90, P=0.35; x2 = 0.01, P= 0. 93 ). There were significant differences in all index of WCST between two groups (P <0.01 ). Patients with rs4532G allele had worse WCST performance than those without G allele ((60.9 ± 13.2)%vs (44.9 ±21.3)%, t=4.79, P=0.00002). Conclusion Rs4532 polymorphism of DRD1 gene may be associated with executive function impairment in schizophrenic patients.
4.Experiment studies on viscoelastic properties of erythrocyte membrane in patients with pulmonale during acute exacerbation.

The membrane viscoelasticity of erythrocyte taken from both normal subjects and patients with cor pulmonale during acute exacerbation was investigated using a micropipette aspiration technique. Experimental results were analysed with vogit viscoelaticity model based on pioneering theory of Chein et al. The results showed that the erythrocyte membrane elastic moduli ((6.970 +/- 1.050) x 10(-3) dyn/cm) and viscous coefficients ((0.936 +/- 0.242) x 10(-4) dyn x s/cm) of the cor pulmonale patients was significantly higher than those of the normal subjects ((5.203 +/- 1.051) X 10(-3) dyn/cm, (0.620 +/- 0.053) x 10(-4) dyn x s/cm). The membrane elastic moduli, viscous coefficients, rigidity of erythrocyte, and viscosity were all increased. It may be the important subcellular mechanism to cause the decrease of erythrocyte deformability and hyperviscosity of blood in these patients.

5.Olanzapine May Inhibit Colonic Motility Associated with the 5-HT Receptor and Myosin Light Chain Kinase.

OBJECTIVE: To study whether the effects of olanzapine on gastrointestinal motility is related to the serotonin antagonism and myosin light chain kinase. METHODS: Male Sprague-Dawley rats were randomly divided into four groups. Olanzapine gavage was performed for each treatment group during the course of 30 continuous days, while the same volume of saline was given to the rats in the control group. Defecation of the rats was observed on days 7 and 30 after olanzapine gavage. The effects of olanzapine on contraction of colonic smooth muscles were observed in ex vivo experiments. A Western blot was used to evaluate expression levels of the serotonin transporter (SERT) and MLCK in colon segments of the rats. RESULTS: ResultsaaCompared to the control group, 5-160 µM of olanzapine could inhibit dose-dependently the contraction of colonic smooth muscle ex vivo experiments. The maximum smooth muscle contraction effects of 5-HT and acetylcholine significantly decreased after treatment with 40-160 µM of olanzapine. Constipation was found in the olanzapine-treated rats on day 7 and have sustained day 30 after gavage. Expression of MLCK in olanzapine-treated rats was significantly decreased, whereas the expression of SERT significantly increased on the day 7, then significantly decreased on the day 30 after olanzapine gavage. CONCLUSION: SERT and MLCK may involve in the inhibition of colonic contraction induced by olanzapine.

6.The influence shuxuetong on the membrane viscoelasticity of erythrocyte taken from patients with chronic pulmonary heart disease.

The present paper was aimed to explore the effect of Shuxuetong on the membrane viscoelasticity of erythrocyte taken from the acute phase patients suffering from chronic pulmonary heart disease. The membrane viscoelasticity of erythrocyte was taken from the acute phase patients suffering from chronic pulmonary heart disease. The changes of membrane viscoelasticity of erythrocyte after treated with shuxuetong were detected by micropipette aspiration technique. The results showed that the Shuxuetong of certain concentration could cause the decrease of membrane elastic modulus and viscous coefficients in acute phase patients suffering from chronic pulmonary heart disease. The study offers experimental evidences that the comprehensive treatment of pulmonary heart disease should involve the drug or measure to improve the erythrocyte deformability.

7.Investigation on the Mechanics of Adhesion to the Selective Extracellular Matrix Coated Surfaces of Lung Cancer Cells

The adhesion properties of tumor cells with extracellular matrix(ECM) are closely associated with their invasion and metastasis.Our work reported here was intended reveal the relevant biomechanical and biorheological manifestations of human lung cancer. Using micropipette aspiration technique, we investigated quantitatively the adhesive mechanics properties of high metastatic human giant cell carcinoma(PG) cells as well as low metastatic adenocarcinoma(PAa) cells of lung based on cell culture in vitro. The results showed that the adhesion forces of PAa and PG cells to collagen Ⅳ were significantly higher than those to glass surfaces, but at the lower concentrations(1.00μg/ml and 2.00μg/ml) of collagen Ⅳ, the amplitude for the increase of adhesion forces of PG cells were less than the amplitude for that of PAa cells, and most of the adhesion force values of PAa cells to the coated surfaces of incorporation of laminin along with 2 μg/ml collagen Ⅳ were significantly greater than those of PG cells. At the lower concentrations(0.625μg/ml for PAa cells,and 0.625 μg/ml, 1. 25 μg/ml for PG cells) of laminin tested,the adhesion force values of PAa and PG cells all decreased, but the amplitude and level for the decreased values of adhesion forces of PG cells were greater than those for the PAa cells. In conclusion, the adhesive and proteolytic behaviour of cancer cells to extracellular matrix might be mediated mainly by tumor cell membrane receptors such as integrin receptors and laminin receptors, it might affect the biological characteristics and the metastasis of the tumor cells. The results may benefit to explain some questions in biomechanical views about how the highly metastatic PG cells are prone to migration and invasion.
8.Research status and hot spots visualization analysis of operating room efficiency at home and abroad based on citespace

Objective To analyze the current research status and hotspots in the field of operating room efficiency both domestically and internationally,and provide theoretical and practical guidance for the improvement and evaluation of operating room efficiency.Methods Using bibliometric methods to search for domestic and foreign literature on operating room efficiency in CNKI and Web of Science core collection databases,Citespace software was used for visual analysis of publication volume,core authors,publication institutions,keyword co-occurrence,clustering,and emergence.Results A total of 387 foreign litera-ture on operating room efficiency were retrieved,with the United States ranking first in terms of publication volume.Research in-stitutions mostly collaborate closely,with research hotspots mainly focusing on time management,anesthesia management,cost management,and quality management.Research frontiers include artificial intelligence,quality improvement,etc;A total of 235 domestic literature have been searched,but a close cooperation network has not yet formed.The research hotspots mainly include lean management,process management,nursing management,etc.The research frontiers include information systems,quality and safety,etc.Conclusion China should integrate research capabilities,strengthen institutional cooperation,pay attention to research hotspots and frontiers,and increase the importance of research on operating room efficiency.
9.Research progress of bio-metal organic frameworks in drug delivery system

In recent years, bio-metal organic frameworks (Bio-MOFs) synthesized with biocompatible ligands have been widely investigated as a potential drug delivery carrier due to their large specific surface area and porosity, rich host-guest intermolecular interactions, and good biocompatibility.In this review, we summarized the design methods of Bio-MOFs including structural and toxic factors, as well as a variety of drug loading methods including click chemistry, with particular focus on recent research advances in Bio-MOFs for pulmonary drug delivery systems, improving pharmaceutical properties of drugs, sustained and controlled drug release, stimulation response and targeted drug delivery systems.Finally, we summarized the bottlenecks that constrain the development of Bio-MOFs in clinical studies of actual pharmaceutical formulations and their future directions for approved formulations, aiming to provide some theoretical reference for promoting the application of Bio-MOFs in drug delivery systems.
10.Predictive value of anti-RA33,IL-6 and hs-CRP levels for treatment response in rheumatoid arthritis based on decision curve analysis

Objective To analyze the predictive value of anti-rheumatoid arthritis-33(anti-RA33),interleu-kin-6(IL-6)and high-sensitivity C-reactive protein(hs-CRP)levels for treatment response in patients with rheumatoid arthritis(RA)based on the decision curve.Methods A total of 102 RA patients admitted to the hospital from January 2021 to August 2023 were selected.The clinical data of the patients were collected and their serum anti-RA33,IL-6 and hs-CRP levels were detected.After half a year of treatment with methotrex-ate and etanercept,the patients were divided into good response group and non-response group according to the treatment response.Pearson correlation analysis was used to analyze the correlation between the levels of anti-RA33,IL-6,hs-CRP and disease activity score 28(DAS28)in RA patients.Multivariate Logistic regres-sion analysis was used to analyze the influencing factors of non-response in RA patients.Receiver operating characteristic(ROC)curve was drawn to analyze the efficacy of anti-RA33,IL-6 and hs-CRP in non-response to RA.The decision curve was used to analyze the net benefit of anti-RA33,IL-6 and hs-CRP alone or in com-bination to predict the non-response of RA patients to treatment.Results After half a year of treatment,80 patients had good or moderate response(good response group)and 22 patients had no response(non-response group).The course of disease in the good response group was shorter than that in the non-response group,and DAS28 score was lower than that in the non-response group(P<0.05).The levels of serum anti-RA33,IL-6 and hs-CRP in the good response group were lower than those in the non-response group(P<0.05).Pearson correlation analysis showed that the levels of serum anti-RA33,IL-6 and hs-CRP were negatively correlated with DAS28 score(P<0.05).Multivariate Logistic regression analysis showed that DAS28 score,anti-RA33,IL-6 and hs-CRP levels were the influencing factors of non-response to treatment in RA patients(P<0.05).ROC curves showed that the area under the curve of serum anti RA33,IL-6,hs-CRP single and combined for predicting patients without reaction were 0.729,0.814,0.831,0.948,and the combined prediction value was higher.The decision curve analysis showed that within the scope of the most reasonable threshold.The overall net benefit of serum anti-RA33,IL-6 and hs-CRP combined predicted treatment responsiveness in RA patients was higher than the net benefit predicted alone.Conclusion The levels of anti-RA33,IL-6 and hs-CRP are closely related to the treatment response of RA patients.The combination of the three has high clinical value and net benefit in predicting no response to treatment.
