1.Mechanism of inhibition of platelet activation by tanshinone type ⅡA though G protein signal pathway

Objective To discuss the mechanism of inhibition of platelet activation by tanshinone type ⅡA(TanⅡA) through G protein signal pathway.Methods Methylthiazolyldiphenyl-tetrazolium bromide(MTT) test was used to determine the optimum effective concentration and optimal time of thrombin and TanⅡA on platelet.Northern blot and Western blot were used to detect the transcription and expression levels of G protein and related signal molecules,including protease activated receptors(PARs),P2Y1 and P2Y12 receptors,α2A-adrenergic receptor and thromboxane A2(TXA2) receptor,in control group,thrombin treated group and TanⅡA treated group,and the platelet aggregation rate was also detected.Results Platelet aggregation rate,and the transcription and expression levels of G protein and related molecules in thrombin treated group were higher than control group(P<0.05).The transcription and expression levels of G protein and related molecules in different concentrations of TanⅡA treated groups were lower than thrombin treated group(P<0.05).Conclusion TanⅡA could inhibit the activation of platelet by inhibiting the transcription and expression of G protein and the related molecules.
2.Effect of retention enema with Chinese Traditional Medicine at different hour on the patients with chronic renal failure

Objective To study the impact of medicine retention time and the therapeutic effect to the chronic renal failure by retention enema with Chinese traditional Medicine at different hour.Methods Totals of 60 patients with chronic renal failure disease were randomly divided into observation group (n=30)and control group (n=30).The observation group adopted enema at 7:00-9:00, and the control group adopted enema at routine hour ( 19:00 -21:00 ) .Following items were observed and compared between two groups: subjective feeling, adverse reflection, defecating ratio before enema, the appetency of defecating after enema, medicine retention time after enema, the Scr before and after enema, the change of BUN and Ua.The observation period was 30 days ( one period of the treatment) .Results Compared with the control group, the adverse reflection, such as bellyache, abdominal distension, anus ache, downfallen pressure feeling at anus were significantly relieved in observation group (P<0.05).The times & severity of the appetency of defecating after enema in observation group was reduced apparently than that in control group (P<0.05).The average defecating ratio before enema in observation group was significantly improved (83.3%vs 20.0%;χ2 =79.452,P<0.01).The retention time in observation group was ( 324.12 ±129.47 ) min, and that in control group was ( 225.32 ± 104.21)min,and the difference was statistically significant (t=4.605,P<0.01).The blood BUN, Scr, Ua were down after enema in both groups, but the improvement in observation group was significantly better than that in the control group (P <0.05).Conclusions By practicing retention enema with Chinese traditional Medicine at 7:00-9:00, the medicine retention time in intestines can be extended, and the therapeutic effect to the chronic renal failure can be improved.

Depressive disorder ranks as a major bur-den of disease worldwide,yet the current antidepressant medications are limited by frequent non-responsiveness and significant side effects.The lateral septum(LS)is thought to control of depression,however,the cellular and circuit substrates are largely unknown.Here,we identified a subpopulation of LS GABAergic adenosine A2A receptors(A2AR)-positive neurons mediating depres-sive symptoms via direct projects to the lateral habenula(LHb)and the dorsomedial hypothalamus(DMH).Activa-tion of A2AR in the LS augmented the spiking frequency of A2AR-positive neurons leading to a decreased activation of surrounding neurons and the bi-directional manipula-tion of LS-A2AR activity demonstrated that LS-A2ARs are necessary and sufficient to trigger depressive pheno-types.Thus,the optogenetic modulation(stimulation or inhibition)of LS-A2AR-positive neuronal activity or LS-A2AR-positive neurons projection terminals to the LHb or DMH,phenocopied depressive behaviors.Moreover,A2AR are upregulated in the LS in two male mouse mod-els of repeated stress-induced depression.This identifica-tion that aberrantly increased A2AR signaling in the LS is a critical upstream regulator of repeated stress-induced depressive-like behaviors provides a neurophysiological and circuit-based justification of the antidepressant poten-tial of A2AR antagonists,prompting their clinical transla-tion.
4.Integrated analysis of DNA methylation and gene expression profiles of hepatocellular carcinoma to construct miR-1180-3p relevant ceRNA regulatory network

Objective:This study analyzes the expression level of miR-1180-3p and constructs the regulatory network of relevant ceRNA by integrating the DNA methylation and gene expression profile of hepatocellular carcinoma from the Cancer Genome Atlas (TCGA).Methods:Firstly, the expression level of miR-1180-3p in hepatocellular carcinoma and adjacent tissues was analyzed by TCGA database, and the differential expression of lncrna and mRNA was screened. Secondly, the LncBase database and the TargetScan database were used to predict the relationship between miR-1180-3p and lncRNA and mRNA, and the DNA methylation-mediated lncRNA was screened by the DNA methylation profile of lncRNA. Finally, Cytoscape software was used to construct miR-1180-3p relevant ceRNA network, and WebGestalt website was used to perform GO and KEGG analysis of related mRNA in ceRNA.Results:Compared with patients with low expression of miR-1180-3p (mean overall survival duration, 5.69 ± 0.35 years), patients with high expression of miR-1180-3p had shorter overall survival time (mean overall survival duration, 3.99 ± 0.47 years), indicating that the high expression of miR-1180-3p was hepatocellular carcinoma risk factor affecting the prognosis ( HR = 1.28, 95% CI = 1.1 ~ 1.5, P < 0.01). A miR-1180-3p related ceRNA regulatory network was constructed in this study, which contained 2 lncRNAs (F11-AS1 and LINC01511) and 37 mRNAs. Conclusion:This study has successfully constructed miR-1180-3p relevant ceRNA regulatory network, and DNA methylation-mediated F11-AS1 and F11-AS1/miR-1180-3p/C11of54 ceRNA regulatory axis has played an important role in the occurrence and development of hepatocellular carcinoma.
