1.Study on the correlation between COX-2, ER, PR and the formation of endometrial polyps and the increased malignant change rate of endometrial polyps after menopause

Objective:To investigate the correlation between cyclooxygenase-2(COX-2), estrogen receptor (ER), progesterone receptor(PR) and the formation of endometrial polyps and the increased malignant rate of postmenopausal endometrial polyps.Methods:From January 2018 to January 2019, a total of 160 postmenopausal women treated in the People's Hospital of Changxing County were selected and divided into endometrial polyps group(83 cases) and oothectomy group(42 cases), normal menopause group(35 cases) according to different diseases.The levels of COX-2, ER, PR in the three groups were measured.The expression of COX-2 in mesenchymal cells and glandular epithelial cells, the expression of ER in mesenchymal cells and glandular epithelial cells, the expression of PR in mesenchymal cells and glandular epithelial cells in the three groups were compared, and the correlation between COX-2, ER, PR and increased malignant rate of endometrial polyps was analyzed.Results:The positive expression rates of COX-2 in mesenchymal cells and glandular epithelial cells in the endometrial polyps group(16.9%, 30.1%) were higher than those in the normal menopause group(0.0%, 11.4%) and the oothectomy group(4.8%, 7.1%)(χ 2=4.568, 5.806, all P<0.05 ). There were no statistically significant differences in the expression of ER in mesenchymal cells and glandular epithelial cells among the three groups(χ 2=1.333, 1.412, all P>0.05). The expression of PR in mesenchymal cells and glandular epithelial cells in the endometrial polyps group was lower than that in the normal menopause group and the oothectomy group(χ 2=4.890, 5.022, all P<0.05). COX-2 was positively correlated with the increase in malignant rate of endometrial polyps( r=4.335, P<0.05), PR was negatively correlated with an increase in the malignant rate of endometrial polyps( r=-4.256, P<0.05), and ER had no significant correlation with an increase in the malignant rate of endometrial polyps( r=1.203, P>0.05). Conclusion:COX-2 and PR are significantly related to the formation of endometrial polyps and the increase in malignant rate of postmenopausal endometrial polyps, but there is no significant expression of ER.Therefore, the detection of COX-2 and PR levels is beneficial to provide a certain scientific basis for clinical treatment.
2.Clinical Observation of NB-UVB Combined with Total Glucosides of White Paeony Capsules and Urea Cream in the Treatment of Psoriasis Vulgaris

OBJECTIVE:To observe therapeutic efficacy and safety of NB-UVB combined with Total glucosides of white paeony(TGP)capsules and Urea cream in the treatment of psoriasis vulgaris. METHODS:A total of 75 patients with psoriasis vulgaris in dermatology department of our hospital during Jan. 2015-Dec. 2016 were divided into control group(37 cases)and observation group(38 cases)according to random number table. Control group was given TGP capsules 0.6 g orally,3 times a day,reducing to 0.3 g,3 times a day if diarrhea or stool increased significantly after taking the medicine+Urea cream,smearing on the skin,day and evening,for consecutive 12 weeks. Observation group was additionally given NB-UVB irradiation with initial dose of 0.36 J/cm2,2 min/time,every other day,adjusted according to skin reaction for consecutive 8 weeks,on the basis of control group. Clinical efficacies of 2 groups were observed,and PASI scores before and after treatment and the occurrence of ADR were observed. RESULTS:One patient of observation group withdrew from therapy after suffering from obvious edematous erythema with pain due to irradiation. All patients of control group completed treatment. Total response rate of observation group (86.49%)was significantly higher than that of control group(56.76%),with statistical significance(P<0.05). Before treatment, there was no statistical significance in PASI scores between 2 groups(P>0.05). After treatment,PASI scores of 2 groups were significantly lower than before treatment,and observation group was significantly lower than control group,with statistical significance(P<0.05). There was no statistical significance in total incidence of ADR between 2 groups(P>0.05). CONCLUSIONS:NB-UVB combined with TGP capsules and Urea cream show good therapeutic efficacy and safety for psoriasis vulgaris,and can significantly decrease PASI score of patients.
3.A comparative study on the expression of EFhd1/2 in AD patients and model mice

Objective :
To investigate the expression differences of helix⁃loop⁃helix hand domain family member D 1/2(EFhd1/2) in Alzheimer′s disease (AD) patients and model mice.
Methods :
The expression changes of EFhd1/2 in AD patients were compared by using data from Alzheimer′s disease database (AlzData), and the changes in mRNA levels and protein levels of EFhd1/2 in brain tissues of AD patients and healthy control group were detected by qPCR and Western blot. The changes of EFhd1/2 mRNA and protein expression in brain tissues of AD models and wild⁃type mice of 3⁃month⁃old and 6⁃month⁃old were detected. Microglia were isolated from AD models and detected the changes of EFhd1/2 by RNA⁃sequencing.
Results :
The Analysis of Alzheimer′s Disease Database data showed that the mRNA levels of EFhd1 in AD patients increased, while EFhd2 decreased. AD patients brain tissue samples showed an upward trend in the expression of EFhd1 in different brain regions of AD patients compared with healthy controls, while EFhd2 was not different. In the AD mice model, the mRNA levels of EFhd1 were similar in both 3⁃month⁃old and 6⁃month⁃old AD mice compared with wild⁃type mice, but the protein levels of EFhd1 increased; the mRNA levels of EFhd2 increased in 3⁃month⁃old AD mice, and protein levels remained similar in the brain tissue from AD mice aged 3 months and 6 months. However, EFhd2 increased in microglia from 6⁃month old AD mice.
Conclusion
EFhd1 increased in both AD model mice and brain tissue of AD patients, suggesting that EFhd1 might play an important role in the development of AD, while EFhd2 increased in microglia in AD model mice, indicating that EFhd2 might be involved in AD related microglia activation and neuroinflammation.
