1.Effect of hand intrinsic muscle exercise on the postoperative recovery of fine movement in children with hand injury

Objective To study the effect of hand intrinsic muscle exercise on postoperative recovery of fine movement in children with hand injury.Methods From January 2009 to December 2011,64 children with hand injury were randomly divided to exercise group and control group equally.Hand intrinsic muscle exercise and rehabilitative care were conducted in the exercise group while traditional rehabilitative therapies in control group.After 3-month therapy,MMDT(Minnesota Manual Dexterity Test)and PPT(Purdue Pegboard Test)were conducted to evaluate the functional recovery of hands.Result The results of MMDT and PPT were obviously better in the exercise group than in the control group(P<0.01).Conclusion Hand intrinsic muscle exercise may be effectively in promoting the recovery of fine movement in children with hand injury.
2.Effects of local delivery c-myc antisense oligodexynucleotides on neointimal hyperplasia and vascular remodeling

Objective To investigate the effects of local delivery c-myc antisense oligodexynucleotides on neointimal hyperplasia after angioplasty in rabbits. Methods The celiac arterial endothelium of 75 male Japanese white rabbit (2.5?0.5 kg) were denuded after being fed with cholesterol-supplemented diet to establish the model of atherosclerotic stenosis of celiac arteries. The animals were randomized into the antisense DNA group (Group Ⅰ), sense DNA group (Group Ⅱ), saline control group (Group Ⅲ) and simple angioplasty group (Group Ⅳ). The drugs were injected intramurally using a porous balloon. The local vessels were collected at the 24 th hour and the 4 th week after the procedure for morphological observation. C-myc mRNA level was measured by means of RT-PCR and c-myc protein with immunohistochemical staining. Results RT-PCR showed that c-myc mRNA level in Group Ⅰ was lower than that of Group Ⅱ, Group Ⅲ and Group Ⅳ after the first 24 hours. The ratio of neointimal area (NEA) to medial area (MA) and NEA/MA in Group Ⅰ were significantly less than those of Group Ⅱ, Group Ⅲ and Group Ⅳ. Immunohistochemical staining showed that the percentage of c-myc protein positive cells was less in Group Ⅰ than those of the other groups. Conclusion Transcatheter local delivery of c-myc antisense oligonucleotides reduces neointimal hyperplasia after angioplasty in rabbits.
3.MicroRNA-10a induces cisplatin resistance by targeting PTEN in human cervical cancer cells

Objective To explore the roles of miR-10a in the cisplatin resistance in cervical cancer cell lines,and further study the mechanism.Methods The cells were transfected with the mimics of miR10a and its negative control RNA (NC).Methyl thiazolyl tetrazolium (MTF) assay and fluorescence activated cell sorter were used to analyze drug sensitivity and apoptosis after treating with cisplatin,luciferase reporter assay to identify that miR-10a directly targets phosphatase and tensin homologue deleted on chromosome ten (PTEN).The expression of PTEN gene and its protein levels after tansfection was measured,respectively.Results after transfected with miR-10a mimics:(1) The 50% inhibition concentration of Hela cells (7.2 μg/ml) was more than in the NC group (5.6 μg/ml).The 50% inhibition concentration of Siha cells (6.4 μg/ml) was significantly more than in the NC group (3.8 μg/ml) (P<0.05).(2) The apoptosis rates of Hela cells and Siha cells were significantly lower compared to the NC group (P < 0.05).(3)miR-10a might directly target the 3'-untranslated region (3'-UTR) of PTEN,which significantly changed the PTEN protein level (P < 0.05).Conclusions miR-10a may modulate cisplatin-induced apoptosis in cervical cancer by inhibiting PTEN protein level.
4.Preventive Medication of Antiboitics in Patients Undergoing Thyroidectomy:Retrospective Analysis of 120 Cases

OBJECTIVE:To investigate the rationality of preventive use of antibiotics for patient undergoing thyroidectomy.METHODS:120 patients undergoing thyroidectomy from Jan 2005 to Mar 2007 in our hospital were enrolled.The exclusion criteria were the following:patients with infection,coronary heart diseases,diabetes,and other severe diseases or with body temperature above 38 ℃ and aged above 60.The 120 patients were assigned to antibiotic group and non-antibiotic group.Then the healing condition of incisions in two groups was evaluated.RESULTS:There was no significant difference between the two groups in incision healing.CONCLUSION:Antibiotics don't have to be used in patients undergoing thyroidectomy.
5.Uterine artery embolization with Pingyangmycin lipiodol emulsion for treatment of symptomatic uterine fibroids

Objective To evaluate the effectiveness and side effects of uterine arterial embolization with Pingyangmycin(a homogenous bleomycin) lipiodol emulsion(PLE) for symptomatic uterine fibroids. Methods Uterine arterial embolization with PLE was performed in 25 patients.The improvement of symptoms and uterine size changes were followed up in 3-18 months(mean 6 months) after the procedure. Results All but 2 cases were successfully treated bilaterally.Superselective angiography showed enlargement of uterine artery,accompanied by tortuous branches.The uterine size was increased.The uterus itself was significantly stained and emptied slowly. Coagulation necrosis was found in resected fibroids after embolization in 3 patients. One month after the procedure, a mean 40% reduction of uterine volume was obtained in 18 followed up cases.The clinical symptoms were relieved significantly. The main side effects were hypogastic pain(13/25),which was intense in 6 cases. Conclusion Uterine arterial embolization with PLE is a good non surgical therapy in symptomatic uterine fibroids with mild side effects.
6.Meta-Analysis of Oral Preparation of Ginkgo Biloba Extract in The Treatment of Mid Cognitive Impairment

Objective To evaluate the efficacy and safety of oral ginkgo biloba extract EGB761 in patients with mild cognitive impairment.Methods They searched PubMed,Embase,the Cochrane Library ,CNKI,Wanfang and VIP databases for randomly controlled trials of oral ginkgo biloba extract for mild cognitive impairment.After assessed the quality of studies included,RevMan5.2 software was used to analyze data.Results Seven studies which including 815 patients were involved by our inclusion criteria.The results of meta-analysis showed,compared with the control group,ginkgo biloba was superior in improving mild cognitive impairment patients' MMSE level[MD=1.81,95%CI(0.02,3.60),P=0.05;MD=1.96,95%CI(1.48,2.43),P<0.000 01;MD=1.79,95%CI(0.99,2.58),P<0.000 1] after treated three months、six months and twelve months.Ginkgo biloba was also superior in improving mild cognitive impaimant patients.CDT level[MD=0.43,95%CI(0.30,0.57),P<0.000 01;MD=0.57,95%CI(0.39,0.75),P<0.000 01] after treated six months and twelve months.The effect of preventing MCI patients into dementia was better than that of the control group[RR=0.27,95%CI(0.06,1.27),P=0.10;RR=0.32,95%CI(0.16,0.63),P=0.001]after treated six months and twelve months.Conclusion Oral preparation of ginkgo biloba extract in the treatment of MCI clinical efficacy and prevention of dementia occurrence rate was better than that of blank control group.

8.Effects of losartan and ginseng on cardiomyocyte Bcl-2 gene expression induced by ischemia and reperfusion

BACKGROUND: The studies found that cardiomyocyte apoptosis is related to ischemia-reperfusion injury directly. The clinical and experimental studies proved that ginseng and losartan could improve myocardial iscbemia and prevent the ischemia-reperfusion injury.But the comparative study of their effects on cardiomyocyte injury induced by ischemia and reperfusion has not been reported.OBJECTIVE: To compare the effects of ginseng and losartan on cardiomyocyte Bcl-2 gene expression after ischemia and reperfusion in vivo.DESIGN: Randomized controlled experiment.SETTING: Department of Cardiovascular Internal Medicine, Tongji Hospital, Tongji Medical College, Huazhong University of Science and Technology.MATERIALS: The experiment was carried out in the Laboratory of Cardiovascular Internal Medicine of Tongji Hospital of Tongji Medical College Affiliated to Huazhong University of Science and Technology from November 2002 to April 2003. Totally 40 healthy adult Wistar rats, weighting 200-250 g, of either gender, were selected and divided into 4 groups:sham operated group, ischemia-reperfusion group, ginseng treated group and losartan treated group with 10 rats in each group.METHODS: Rats were modeled in ischemia-reperfusion group, ginseng treated group and los arran treated group but not in sham operation group.20 mg/kg (1 mL in volume) losartan was given by stomach. The first administration was 2 hours prior to operation. Subsequently, the second and third administrations were given in immediate and 24 hours after operation,respectively; 1 mL of radix ginseng rubra (1 g/mL) was given by stomach.The first administration was 2 hours before operation. Subsequently, the second and third administrations were given in just and 24 hours after operation, respectively. The rats in ischemia-reperfusion and sham-operated group were given the normal saline with the same volume and at the same time. Immunohistochemistry and in situ hybridization histochemistry were used to measure mRNA and protein of Bcl-2 gene expression that were compared with those in the control group.MAIN OUTCOME MEASURES: The expression level of Bcl-2 mRNA and Bcl-2 protein in ginseng treated group and losartan treated group.RESULTS: Data of totally 40 rats was entered the final analysis. ① Content of Bcl-2 mRNA and protein were not significantly different both in ginseng group and control group (P > 0.05). ② Contents of Bcl-2 mRNA and Bcl-2 protein were higher in ginseng treated group than those in the ischemia-reperfusion group (P < 0.05 or 0.01).CONCLUSION: Expressions of Bcl-2 mRNA and Bcl-2 protein in ginseng treated group are higher than those in losartan treated group, which suggests that ginseng has not same effect with losartan in inhibiting cardiomyocyte apoptosis and in preventing and curing cardiomyocyte injury induced by ischemia-reperfusion.
9.Acquiring and analyzing of ornithine aminotransferase gene of Schistosoma japonicum

Objective To screen and identify the novel genes from the adult worm cDNA library of Schistosoma japonicum by the expressed sequence tags (EST) strategy.Methods The cDNA clones were selected randomly and the ESTs were obtained.The novel gene was searched for homologue identity with NCBI blast programmer.The homologue of the two sequences with a high identity was compared at amino acid and nucleotide level with the BLAST programmer,and the analysis of protein was carried out with the pcgene software. Results There was a novel gene of Schistosoma japonicum which included 1 677 bp coding for 424 amino acid residues, and it was given the accession number of sequence in Genbank(AY336497).The cDNA sequence was homologous to the known ornithine aminotransferase gene of Xenopus laevis. The identity of amino acid sequence was 66%. The academic pI was 8.52, and the antigen determinant was probably from 795 to 846 on the cDNA sequence. Conclusion EST strategy is an effective measure to discover new genes of Schistosoma japonicum. The novel gene is homologous to the ornithine aminotransferase gene of Xenopus laevis.This study is helpful to further sequential and functional search of the gene.[
10.Application of Shikani optical stylet to orotracheal intubation in critically conscious patients

Objective To investigate the practical value of Shikani optical stylet used for orotracheal intubation in critically conscious patients and to evaluate the successful rate. Method A total of 48 conscious patients with severe respiratory failure were selected from January 2008 to June 2009. Of them, there were 31 males and 17 females,aging 21-86 years old with an average of 57 years old. All enrolled patients needed endotracheal intubation for mechanical ventilation support, and they were assigned to Shikani group (group S, n = 25) and Macintosh group (group M, n = 23) according to the odd and even number of date of admission to this study. The time consumed for intubation, the number of failure in intubation, the adverse effects or complications such as hemodynam-ic changes, injury to the pharyngo-oral cavity, choking and breath-hold were observed and recorded. The rank test and chi-square test were used for statistical analysis. Results The ratio of the successful intubation at first attempt was much higher in group S (96.0%) than that in group M (60.9%) (P <0.01). Compared with group M, the time consumed for intubation was significantly shortened, the cardiovascular reactions were much mild, and the incidence of injury to pharyngo-oral cavity, choking and breath-hold were less in the group S ( P < 0.01 all). Conclusions For the acute and critical patients, especially the conscious ones, orotracheal intubation with the Shikani optical stylet is rapid, successful, safe and less injurious, resulting in mild cardiovascular reactions.
