1.Preparation and characterization of E.coli “bacterial ghost”

Objective To prepare the “bacterial ghost” of E.coli with heat-induction and analyse its lysis rate and configuration. Methods Through immediately shifting the culture temperature from 37℃ to 42℃, E.coli DH5? including plasmid pMuH36 was induced to lyse, and the OD value of culture media was measured every 30 minutes during the induction. After 4 hours of induction, the bacteria samples were collected to examine the lysis rate by CFU (colony formation unit) and the configuration of lysed bacteria was observed by transmission electron microscopy (TEM). Results The OD value of DH5? (pMuH36)began to decline after 1 hour of induction, and increased slowly after 4 hours of induction. The CFU assay showed that the lysis rate was about 95%. TEM observation proved that most of the lysed bacteria were emptied, whereas the whole outmembrane structure, i.e. “bacterial ghost”, remained. Conclusions The E.coli “bacterial ghost” was efficiently prepared and identified, which might provide the basis for further development of a more effective “bacterial ghost” vaccine and adjuvant.
2.Antisense oligonucleotides targeting TNF-? suppress on Co-Cr-Mo particle-induced osteolysis

[Objective]To investigate the effect of a single subcutaneous dose of an antisense oligonucleotide(ASO) on particle-induced osteolysis.[Method]The murine calvaria osteolysis model was utilized in ICR mice.Bone resorption was measured with the toluidine blue staining.Osteoclasts were detected by tartrate resistant acid phosphatase(TRAP) staining assay and were quantified by a TRAP quantification kit.[Result]Bone resorption was 0.347 ? 0.09 mm2 in animals with particle implantation,and decreased to 0.123 ? 0.05 mm2 and 0.052 ? 0.02 mm2 after ASO treatment in low and high doses,respectively.The bone resorption was reestablished in animals given an additional TNF-?.The number of osteoclasts in animal calvaria treated with ASO was reduced obviously compared with those untreated animals and the quantification results indicated that about 90% osteoclastgenesis was suprressed by the ASO.Additionally,the osteoclastgenesis was reestablished by the addition of TNF-?.[Conclusion]An antisense oligonucleotide targeting an inflammatory factor,TNF-?,has been to suppress the osteolysis induced by particle for the first time.This new finding holds a great promise.It is a therapeutic strategy for the component loosening.
3.The curative effects and related analysis of bezafibrate combined medroxyprogesterone acetate in the treatment of AML

Objective To observe the curative effects of bezafibrate combined medroxyprogesterone acetate in the treatment of AML .Methods 40 cases of AML patients were collected and devided into experimental group (bezafibrate combined medroxypro-gesterone acetate ,6 cases) ,control group (chemotherapy ,28 cases) and non-treatment group (6 cases) ,then through a prospective analysis we studied the curative effects and related results of bezafibrate combined medroxyprogesterone acetate in the treatment of AML .Results The median survival time of the experiment group (accepted bezafibrate combined medroxyprogesterone acetate ) was 15 weeks ;the median survival time of the control group (accepted chemotherapy ) was 24 weeks ;the median survival time of the giving up group(accepted any therapy ) was 8 weeks .Conclusion Bezafibrate combined medroxyprogesterone acetate could prolong the survival time of AML patients and there were no obvious adverse reactions .

5.Measurement of healthy young female's auricular lobules and its related factors

Objective:To investigate auricular lobule index difference in healthy young female of different height or weight, and to provide an additional reference for otoplasty of auricular lobule.Methods:Subjects were made up of 266 healthy young females of Henan residents. Their height, weight, physiognomic ear breadth, morphological ear breadth, physiognomic ear length, morphological ear length, auricular lobule length, auricular lobule breadth were measured and calculated. All the data were analyzed by SPSS 15.0 software.Results:We obtained the means, standard deviation of each items, and data were treated statistically. In the 266 subjects, average height was (162.9±4.1) cm, weight (55.3±6.5) kg, morphological ear breadth (4.9±0.6) cm, morphological ear length (2.9±0.3) cm, physiognomic ear breadth (6.2±0.4) cm, physiognomic ear length (3.3±0.3) cm, auricular lobule breadth (1.6±0.3) cm, auricular lobule length (1.8±0.2) cm, lobule physiognomic ear breadth index (25.6±4.9)%, lobule-physiognomic ear length index (53.69±7.69)%, and auricular lobule index (91.4±19.3)%. The differences among the indices mentioned above were of statistical significance ( P<0.05); the indices of subjects of different height were significantly different ( P<0.05); the indices of subjects of different weight were of no statistical difference. ( P>0.05). Conclusions:The subjects of different height or weight have no different morphological ear breadth, morphological ear length, physiognomic ear breadth, physiognomic ear length, auricular lobule length, lobule-physiognomic ear length index and auricular lobule index; hence, the indices are no related to body height and weight. However, the subjects of different height have different auricular lobule breadth and lobule-physiognomic ear breadth index.

7.Perioperative management of abdominal aortic balloon occlusion in patients compli-cated with placenta percteta:a case report

SUMMARY When placenta previa complicated with placenta percreta, the exposure of operative field is difficult and the routine methods are difficult to effectively control the bleeding, even causing life-threatening results. A 31-year-old woman, who had been diagnosed with a complete type of placenta pre-via and placenta percreta with bladder invasion at 34 weeks gestation. Her ultrasound results showed a complete type of placenta previa and there was a loss of the decidual interface between the placenta and the myometrium on the lower part of the uterus, suggestive of placenta increta. For further evaluation of the placenta, pelvis magnetic resonance imaging was performed, which revealed findings suspicious of a placenta percreta. She underwent elective cecarean section at 36 weeks of gestation. Firstly, two ureteral stents were placed into the bilateral ureter through the cystoscope. After the infrarenal abdominal aorta catheter was inserted via the femoral artery ( 9 F sheath ) , subarachnoid anesthesia had been estab-lished. A healthy 2 510 g infant was delivered, with Apgar scores of 10 at 1 min and 10 at 5 min. Imme-diately after the baby was delivered, following which there was massive haemorrhage and general anaes-thesia was induced. The balloon catheter was immediately inflated until the wave of dorsal artery disap-peared. With the placenta retained within the uterus, a total hysterectomy was performed. The occluding time was 30 min. The intraoperative blood loss was 2 500 mL. The occluding balloon was deflated at the end of the operation. The patient had stable vital signs and normal laboratory findings during the recovery period and the hemoglobin was 116 g/L. She was discharged six days after delivery without intervention-related complications. This case illustrates that temporary occlusion of the infrarenal abdominal aorta using balloon might be a safe and effective treatment option for patients with placenta previa complicated with placenta percreta, who were at high risk for peripartum hemorrhage. Early removal of the endovascu-lar catheter and close postoperative surveillance of the vascular system are required with this procedure to minimize the risk of vascular complications. However, further studies are needed to determine whether the potential benefits of temporary occlusion of the infrarenal abdominal aorta using balloon outweigh the potential risks.

Background and purpose:Smac/DIABLO was the only apoptosis-related protein that could inhibit IAPs directly and simultaneously.The four amino-residual AVPI(Ala-Val-Pro-lie)in its N-terminal was the very important domain that could stimulate apoptosis.This study investigated the effect of synthetic Smac peptide (SmacN7) on chemotherapy sensitivity of bladder cancer cells.Methods:SmacN7 penetratin peptide was synthesized and delivered into T24 cells.MTT assay was adopted to evaluate the viability of T24 cells induced by low-dosage of MMC. Flow cytometry was applied to analyze the proportion of apoptosis and Western blot was used to detect the expression of XIAP and caspase-3;The activity of caspase-3 was measured and the effect of SmacN7 combined with MMC on T24 cell lines was also determined.Results:SmacN7 penetratin peptide could successfully interact with endogenous XIAP and increase the proportions of apoptosis of T24 cell lines induced by low-dosage of MMC in a dose-and time- dependent manner.An obvious down-regulation of XIAP expression and up-regulation of caspase-3 was identified by Western blot.The activity of caspase-3 in experimental group was significantly increased as compared with that in the control group;Combining the treatment with SmacN7 penetratin peptide,the viability of T24 cells decreased to 55% and 72.7% in 24 hrs and 48 hrs respectively.Conclusion:SmacN7 penetratin peptide could act as a cell-permeable IAP inhibitor,inhibit the proliferation,induce apoptosis and enhance the chemo-sensitivity of bladder cancer cells to MMC. When combined with chemotherapy,it may be a very promising strategy for bladder cancer therapy.
9.The effects of mutation of PKA phosphorylation site of Mfn2 on the intimal proliferation after balloon injury of carotid artery in rats

Objective To study the effects of protein kinase A (PKA) phosphorylation site' s mutation of mitofusin-2 (Mfn2) on intimal proliferation after balloon injury of carotid artery of rats. Method Rat model of carotid artery balloon injury was established and infected with Adv-LacZ, Adv-Mfn2, AdvMfn2-S442A or Adv-Mfn2-S442D from the peri-arterial sheathes of vessels, while phosphate buffered solution (PBS) used instead of above infectious adenovirus as uninfected group and sham operation as control group. The rats were sacrificed 14 days after balloon injury of carotid artery in order to measure the level of Mfn2 protein and the prol9iferation cell nuclear antigen (PCNA) with immunohistochemistry staining. The morphology of vessels was observed with HE staining. All data were statistically analyzed with ANOVA and Dunnett-t test. Results Fourteen days after surgery, the levels of Mfn2 protein significantly increased in arteries infected with Adv-Mfn2, Adv-Mfn2-S4442A and Adv-Mfn2-S442D compared with those in control group, sham operation group and Adv-LacZ infected group. The ratio of intimal area/medial area (I/M) and percentage of PCNA positive cells in both Adv-Mfn2 and Adv-Mfn2-S442A groups markedly decreased compared with control group (P <0. 01 ) . Compared with the Adv-Mfn2 group, the I/M and the percentage of PCNA positive cells reduced more significantly in Adv-S442A group (P < 0. 01 ) . However,the I/M and the percentage of PCNA positive cells in Adv-LacZ and Adv-S442D groups were not significantly different from those found in the control group. Conclusions The over-expression of Mfn2 gene may effectively inhibit intimal proliferation after balloon injury of carotid artery of rats. The inhibitory effects of Mfn2-S442A are more obvious than those of Mfn2. However, the Mfn2-S442D is out of the inhibitory effect on neo-intimal proliferation.
