1.Analysis of tongue figure features in 990 cases of sexually transmitted and intravenous drug use spread HIV infected population in Xinjiang.

OBJECTIVETo investigate the tongue manifestation features of sexually transmitted and intravenous drug use spread HIV infected population in Xinjiang.
METHODSRecruited were 990 HIV infected subjects in Xinjiang from May 2011 to March 2012, who were assigned to the intravenous drug use spread HIV infected (498 cases) and the sexually transmitted (492 cases). By using tongue figure shoot combined with analyses of experts, tongue manifestations were analyzed and compared between the sexually transmitted and the intravenous drug use spread from four aspects, i.e., the tongue color, the tongue shape, the fur color, and the fur property.
RESULTSCompared with the sexually transmitted population, red tongue, fissured tongue, yellow fur, thick fur, eroded fur, deficiency of fur fluid were more often seen, showing statistical difference (P < 0.05). Compared with the intravenous drug use spread population, pale tongue, white fur, and thin fur were more often seen, showing statistical difference (P < 0.05).
CONCLUSIONSThe tongue manifestations of the intravenous drug use spread HIV population reflected inner exuberance of evil toxin and heat impairing qi and yin. Compared with the intravenous drug use spread population, the attack of HIV infection was more hiding in the sexually transmitted population, with milder internal injury. Their Wei-qi was not damaged and no obvious change occurred in the tongue figure.

3.Kaempferol alleviates ox-LDL-mediated endothelial cell injury via regulating AMPK/Nrf2/HO-1 signaling pathway

Objective:To investigate the effects of kaempferol on the oxidized low density lipoprotein(ox-LDL)-induced injury in human umbilical vein endothelial cell (HUVECs) and to explore its underlying molecular mechanism.Methods: HUVECs were randomly divided into 6 groups:control group,ox-LDL group,Kaempferol+ox-LDL group,Kaempferol+ox-LDL+Compound C group, Kaempferol+ox-LDL+si-Nrf2 group and Kaempferol+ox-LDL+si-HO-1 group.The cell activity was measured by MTT assay.The cell apoptosis was analyzed by flow cytometry.The protein expression were measured by Western blot.The ROS levels were evaluated by flow cytometry.Results: Compared with control group,ox-LDL treatment decreased the cell viability,increased the cell apoptosis,up-regulated the protein levels of cleaved-caspase 3 and down-regulated the Bcl-2 level,elevated the expression of TNF-α,IL-1β,IL-6, vascular cell adhesion molecule (VCAM1),intercellular adhesion molecule (ICAM1) and selectin E (E-selectin),promoted the reactive oxygen species (ROS) generation and reduced the superoxide dismutase(SOD) levels,and decreased the protein levels of p-AMPK,Nrf2 and HO-1.Compared with the ox-LDL group,kaempferol treatment reversed the ox-LDL-induced cell activity inhibition, apoptosis and oxidative stress damage,and increased the protein levels of p-AMPK,Nrf2 and HO-1.Compound C,si-Nrf2 and si-HO-1 inhibited AMPK/Nrf2/HO-1 signaling pathway,elevated the production of ROS and thus inhibiting the protective effects of kaempferol on ox-LDL-treated HUVECs.Conclusion:Kaempferol alleviates ox-LDL-induced endothelial cell injury,which may be related with the activation of AMPK/Nrf2/ HO-1 signaling pathway.
5.Effects of 75 gram glucose oral tolerance test and standard meal test on insulin secretion function of islets of Langerhans and free fatty acid in patients with type 2 diabetes mellitus

Objective To observe the effects of 75 gram glucose oral tolerance test (75 g OGTT) and standard mixed meal test (SMMT) on insulin secretion function of the islets of Langerhans and plasma free fatty acid (FFA) in patients with type 2 diabetes mellitus.Methods Seventy-six patients with type 2 diabetes without using insulin and with no obvious complications were recruited for 75 g OGTT following overnight fasting on the first day and SMMT (bread 50 g,egg 50 g and milk 250 ml) on the 7th day.Blood specimens were collected from each patients before the tests and 30 min,60 min,120 min and 180 min after glucose or meal load to measure their levels of plasma glucose,serum insulin,C peptide,FFA and lipids (total cholesterol,triglyceride,high-density and low-density lipoprotein cholesterol).Results No difference in fasting plasma glucose,serum insulin,C peptide,FFA and lipids between 75 g OGTT and SMMT was found.Postprandial plasma glucose 30 min,60 min,120 min and 180 min after 75 g OGTT was significantly higher than that after SMMT,with (15.3?3.5) vs (9.9?3.4) mmol/L,(18.2?4.8) vs (12.8?4.0) mmol/L,(16.3?5.8) vs (12.2?4.9) mmol/L and (10.6?5.4) vs (9.5?4.5) mmol/L (F=28.1,P
6.Effects of 1-bromopropane exposure on cognitive function in rats.

OBJECTIVETo study the effects of 1-bromopropane (1-BP) on the functions of learning-memory and the central cholinergic system in rats.
METHODSForty male Wistar rats were randomly divided into four groups: low 1-BP group (200 mg/kg), middle 1-BP group (400 mg/kg), high 1-BP group (800 mg/kg) and control group, and the exposure time was 7 days. The Morris water maze (MWM) test was applied to evaluate the learning-memory function in rats. After the MWM test, the rats were sacrificed, the cerebral cortex and hippocampus were quickly dissected and homogenized in ice bath. The activity of acetylcholine esterase (AChE) and choline acetyltransferase (ChAT) in supernatant of homogenate were detected.
RESULTSThe latency and swim path-length of rats in middle and high 1-BP groups prolonged significantly in place navigation test and the efficiency of searching strategy obviously decreased, as compared with control group (P < 0.05 or P < 0.01). In spatial probe test, the number of crossing platform in three 1-BP groups decreased significantly, as compared with control group (P < 0.05 or P < 0.01). The cortical AChE activity of rats in middle and high 1-BP groups was significantly higher than that of control and low 1-BP group (P < 0.05 or P < 0.01). The AChE activity in rat hippocampus of high 1-BP group obviously increased, as compared with control group as compared with control group (P < 0.05). There was no significant difference of cortical ChAT activity between three 1-BP groups and control group (P > 0.05). In the hippocampus, there was no difference of ChAT activity among the groups (P > 0.05).
CONCLUSION1-BP exposure could significantly influence the learning-memory function in rats due to the increase of AChE activity.

7.Analysis of mitochondrial DNA D-loop region mutation and its significance in human oncocytoma.

OBJECTIVETo investigate the mutation in mitochondrial DNA displacement-loop (mtDNA D-loop) region in oncocytoma and its relationship with tumorigenesis and tumor development.
METHODSThe mtDNA D-Loop region of 20 thyroid or renal oncocytomas and the adjacent normal tissues were amplified by PCR, and then sequenced. Five human fetal renal tissues were collected as matched controls.
RESULTSAmong the 20 oncocytomas, 21 mutations which focused on hypervariable region I (HVI) were found in 7 tumor tissues and 1 normal tissue with the mutation rates of 35% and 5%, respectively. At the same time, 191 polymorphisms were found in the 20 cases.
CONCLUSIONmtDNA D-loop region, especially HV I, is the mutational hotspot of oncocytomas, which may be closely related with mtDNA duplicating rate and the function of mitochondria.

8.Study on the AIDS-related high risk behaviors of men who have sex with men under the experience of exchanging money for the same sex

Objective To investigate the AIDS-related high risk behaviors of men who havesex with men (MSM) who ever exchanged money for the same sex. Methods Target sampling forcross-sectional study and valid anonymous questionnaires were adopted to compare the differences ofAIDS-related high risk behaviors between MSM with the experience of exchanging money for thesame sex and those without that experience. 1959 rstadied samples were recruited in nine cities. SPSS13.0 was used and t, x 2 and Mann-Whitney tests were taken for statistical analysis. ResultsCompared to corresponding ones without "buying" the same sex experience, MSM who ever engagedin "buying" sex had the characteristics of being prone to in marriage and living in larger cities witholder age and higher income (P<0.01). They also had a significant larger number in the followingevents: total sexual panners, anal sex and oral sex episodes with same sex, number of sexualpartners, anal sex and oral sex in the previous six months, with the figure of median 50.0, 20.0,20.0, 5.0,4.0,5.0 respectively. 31.5% had ever participated in 'group sex', 48.0% had sex with malepartners away from his own region in the previous year, 70.5% had sexual intercourse withstrangers at MSM avenues in the last six months. The OR (95% CI) values were 2.288( 1.702-3.077), 3.231 (2.462-4.241 ), 2.840 (2.140-3.770). All the above mentioned figures werehigher than those without the experience, with significant differences. They had a significant lower rate of 45.7% of condom use while having sex with female partners (P<0.05). Conclusion MSM with the experience of exchanging money for the same sex would have more AIDS-related high risk behaviors, AIDS preventative measures should be taken to target different subgroups.
9.Combining CpG and Al(OH)3 enhances the immunogenicity of hepatitis C virus recombinant ptotein combined vaccine

Objective To research CpG and Al(OH)3 adjuvants enhancing immunogenicity of hepatitis C virus(HCV) recombinant ptotein combined vaccine(TIE).Methods BALB/c mice were immunized with candidate vaccine TFE using CpG,Al(OH)3,Al(OH) 3 + CpG,or freund's adjuvant(FA) as the adjuvant.Five mice were sacrificed after 10 d of the last immunization.Specific antibodies in sera were tested by enzyme-linked immunosorbent assay(ELISA).Splenic cells were isolated and levels of IFN-γ,IL-4 and cytotoxic T lymphocyte(CTL) cytotoxicity assay were messuredin vitro.The remaining mice were subcutaneouly injected with 1 × 106 SP2/0-NS3 cells on the back to investigate the protective effects.The differences of means between groups were compared by LSD-t test.Results The specific CTL activity of TFE + A1(OH) 3 + CpG group was higher than TFE + FA group and TFE + CpG group(P < 0.05).The level of IFN-γsecreting cells in TFE + Al(OH)3 + CpG group was higher than that in TFE + M(OH)3 group or TFE + CpG group(P < 0.05).Conclusion Combining Al(OH) 3 and CpG could enhance specific cellular immunogenicity of candidate HCV vaccine TFE.TFE + M(OH) 3 + CpG could effetively prevent the attack of tumor cell SP2/0-NS3 expressing nonstructural protein NS3 of HCV.
10.Beneficial effects of a diabetes specific formula on insulin sensitivity and free fatty acid in patients with type 2 diabetes mellitus.

BACKGROUNDThis prospective, randomized, controlled study was designed to investigate the effects of a diabetes specific formula (Diason low energy: 313.8 kJ/100 ml), compared with a standard formula, on insulin sensitivity, serum C peptide, serum lipids and free fatty acid (FFA) in type 2 diabetics.
METHODSIn total of 71 type 2 diabetics completed the study. Enteral formulas were given orally as the sole source of nutrition to the subjects for 6 days. Venous blood samples (0.5, 1, 2, 3 hours) were collected at day-7 after a 75 g oral glucose tolerance test (OGTT), day 1 after a standard test meal (1673.6 kJ) and after 6 days of either the test diabetes specific formula or a standard formula. Plasma glucose, serum insulin, C peptide and lipids were measured.
RESULTSAfter the intervention period, the diabetes specific formula resulted in a significantly lower postprandial rise in blood glucose concentrations at 0.5 hour (P < 0.05) and 1 hour (P < 0.01); significantly lower peak height of plasma glucose (P = 0.05); significantly lower plasma insulin concentrations at 0.5 hour (P < 0.01), 1 hour (P < 0.01) and 2 hours (P < 0.01); and a significantly lower plasma insulin peak compared to controls; both OGTT and a standard test meal (P < 0.05). The glucose and insulin area under the curve after the diabetes specific formula compared to the standard formula were significantly lower. The C peptide level was lower after 6 days of both nutrition formulas compare to 75 g OGTT, but not different from the standard mixed meal. Both formulas were well tolerated.
CONCLUSIONSIn summary the diabetes specific formula with a relatively high monounsaturated fatty acid and high multi fiber proportion significantly improved glycemic control. On top of this, the insulin sensitivity (HOMA-IS) was significantly improved and may therefore directly improve the impact on long term complications. The disease specific formula should therefore be the preferred option to be used by diabetic and hyperglycemic patients in need of nutritional support.
