4.Pharmacodynamic Study on Xiaolin Powder

Objective:To study the pharmadynamics in association with the clinical effects of Xiaolin powder. Methods: The bacteriostatic, anti inflammatory, antipyretic, analyesic, diuretic and immunologic actions of xiaolin powder were studied on the corresponding pharmacological models.Results: Xiaolin power had these actions above.Conclusion: Xiaolin powder can be used in the treatment of urinary infection such as part of damp heat strangury and blood strangury.
5.Effect of Rosiglitazone on Insulin Resistance and ROS . IKK Signaling Pathway in Vascular Endothelial Cells

Objective To explore the protective effect of rosiglitazone on insulin resistance( IR)induced by high glucose in vascular endothelial cells and its possible mechanism. Methods Human umbilical vein endothelial cells( HUVECs) was divided into 3 groups:the normal control group cultivated in DEME medium with 5. 5 mmol·L-1 glucose;the high glucose group( HG)cultivated in DEME medium with 33 mmol · L-1 glucose for 24 h after the IR model was set up;the rosiglitazone group cultivated in DEME medium with 33 mmol·L-1 glucose and 10 μmol·L-1 of rosiglitazone for 24 h after the IR model was set up. The cell viability,nitric oxide(NO),endothelin-1(ET-1),mitochondrial membrane potential,reactive oxygen species ( ROS),p-IKK and IkBa protein levels were detected. Results Compared with the normal control,the cell viability,the level of NO and the mitochondrial membrane potential were decreased,levels of ET-1 and ROS increased,p-IKK expression was up-regulated,and IκBα expression was down-regulated in HG group(all P〈0. 01). Rosiglitazone reversed these changes in a time-dependent manner(P〈0. 05). Conclusion Rosiglitazone has the protective effect on insulin resistance induced by high glucose in vascular endothelial cells via inhibiting ROS/IKK signaling pathway.
6.Effects of recombinant gene lentivirus containing LIM mineralization protein-1 on proliferation effect and expression of bone marrow mesenchymal stem cells in rats.

OBJECTIVETo explore method of recombinant gene lentivirus containing LIM mineralization protein-1 (LMP-1) in transfecting bone marrow mesenchymal stem cells (BMSC), and to observe the effect of gene LMP-1 on proliferation effect and expression of BMSC.
METHODSSix clean SD rats aged 4 weeks were selected, bone marrow mesenchymal stem cells were extracted under sterile conditions and cultured to the third generation, then divided into three groups:control group (the third generation of BMSC), lentiviral vector transfection group (PGC-FU-GFP and Polybrene were injected into the third generation of BMSC) and recombinant gene transfection group (PGC-FU-LMP-1-GFP and Polybrene transfection were injected into the third generation of BMSC). After 48 hours' transfection, fluorescent expression were detected under immuno-fluorescence microscopy; lentiviral transfection efficiency were detected by flow cytometry; effect of lentiviral transfection on BMSC were evaluated by MTT; gene expression of transfected cells were determined by Western Blot.
RESULTS1) The third generation of BMSC was cultured successfully,and transfected with MOI:100. After 48 hours, green fluorescent expression were detected and transfection efficiency was 67% under immuno-fluorescence microscopy; 2) Compared to control group, there were no statistical differences between control group and other two groups; 3) Western blot teast showed that 72KDa specific band was observed in recombinant gene transfection group and its size was similar to LMP-1 fusion protein (50 kDa+28 kDa=78 kDa).
CONCLUSIONThere is no effect of recombinant gene lentivirus containing LIM on BMSC, and can effectively influence the expression of LMP-1.

Objective:To investigate the association between MIC allele polymorphism and susceptibility to Ureaplasma urealyticum infection. Methods:PCR-SSP and PCR-SBT were used to analyze the gene polymorphism of every one. Results:Twelve allele genes of MICA and five MICA-STR and fourteen MICB were found in the participants,the frequency of MICA?010 and MICB?009N were higher in ureaplasma urealyticum infection patients than in healthy controls(MICA?010:OR=3. 85,95%CI:2. 12-6. 99, Pc<0. 05;MICB?009N:OR=3. 22,95%CI:1. 33-7. 80,Pc<0. 05);the frequencies of MICA-A5. 1 and MICB?00502 were lower in ureaplasma urealyticum infection patients than in healthy controls(MICA-A5. 1:OR=0. 61,95%CI:0. 40-0. 94,Pc<0. 05;MICB?00502:OR=0. 58,95%CI:0. 40-0. 83,Pc<0. 05),the differences were statistically significant;MICA?010/010,MICA?01201/01201 homozygote were higher in ureaplasma urealyticum infection patients than in healthy controls(MICA?010/010:OR=14. 84, 95%CI:1. 90-115. 9,Pc<0. 05:MICA?01201/01201:OR=10. 83,95%CI:1. 35-86. 79,Pc<0. 05),the differences were statistically significant. The distribution frequency of MICB?00502/00502 in patients with ureaplasma urealyticum was higher,but the difference was not statistically significant(Pc>0. 05). Conclusion:MIC allele gene polymorphism may be associated with ureaplasma urealyticum infection.
8.Transplantation using donation after cardiac death meeting Chinese standard Ⅲ : 4 cases report

Objective To discuss the curative effect of transplantation using donation after cardiac death (DCD) according with Chinese standard Ⅲ (C-Ⅲ).Methods The organs were obtained from 4 DCDs from 2011 to 2012,and the clinical data of DCD transplantation were retrospectively analyzed.Withdrawal of life support occurred in the operating room.Donor warm ischemia time was 7-15 min.Results The biopsy of liver was performed on the 3rd DCD.Eight cases were subjected to renal transplantations,and 3 to liver transplantations.One patient exhibited delayed graft function of the kidney from the 4th DCD.All patients made an uneventful recovery and were discharged from the hospital without rejection or surgical complication.They were followed up in outpatient department.Conclusion The use of DCD according with C-Ⅲ is an effective way to increase the number of organs available for transplantation,and can obtain satisfactory effects.

The combination effect of high hydrostatic pressure and moderate heat on the inactivation kinetics of Bacillus coagulans spore in phosphate buffer and UHT(Ultra High Temperature)whole milk was investigated.The pressure come-up time and corresponding log-reduction of spore inactivation were considered during pressure-thermal treatment.Bacillus coagulans spore had a much higher resistance to pressure in UHT whole milk than in phosphate buffer.Survival data were modeled using the linear,Weibull and log-logistic models to obtain relevant kinetic parameters.The tailing phenomenon occurred in all survival curves,indicating the linear model was not adequate for describing these curves.The log-logistic model produced best fits to survival curves,following by Weibull model.

P300/CBP-associated factor(PCAF),an important member of histone acetyltransferase family(HATs) within eukaryotic cells,is capable of inducing the acetylation of histone,promoting the transcription of specific genes and involving in many biological effects.In the present study,full-length cDNA of PCAF was inserted into plasmid pGEX-5x-1,then the soluble protein GST-PCAF was expressed in E.coli BL21(DE3) after the optimization of inducing conditions.The recombinant protein was further purified with affinity chromatography and tested the activity by in vitro acetylation assays.High efficient PCAF protein produced by this method could serve for the study on the role of PCAF in gene regulation and the interaction between PCAF and other proteins.
