1.Preparation and characterization of E.coli “bacterial ghost”

Objective To prepare the “bacterial ghost” of E.coli with heat-induction and analyse its lysis rate and configuration. Methods Through immediately shifting the culture temperature from 37℃ to 42℃, E.coli DH5? including plasmid pMuH36 was induced to lyse, and the OD value of culture media was measured every 30 minutes during the induction. After 4 hours of induction, the bacteria samples were collected to examine the lysis rate by CFU (colony formation unit) and the configuration of lysed bacteria was observed by transmission electron microscopy (TEM). Results The OD value of DH5? (pMuH36)began to decline after 1 hour of induction, and increased slowly after 4 hours of induction. The CFU assay showed that the lysis rate was about 95%. TEM observation proved that most of the lysed bacteria were emptied, whereas the whole outmembrane structure, i.e. “bacterial ghost”, remained. Conclusions The E.coli “bacterial ghost” was efficiently prepared and identified, which might provide the basis for further development of a more effective “bacterial ghost” vaccine and adjuvant.

3.Effects of different catheterization on patients with oral poisoning gastric lavage:a meta-analysis with trial sequential analysis

Objective To assess on the effect of the different catheterization on patients with oral poisoning gastric lavage systematically and objectively. Methods A computerized search of PubMed, the Cochrane Library, EMBASE, Springer Link, Science Direct,China Biology Medicine (CBM),China National Knowledge Internet(CNKI), Wanfang Data and VIP database was performed for relevant randomized controlled trials (RCTs) which investigated the effects of the oral intubation gastric lavage on patients with oral poisoning relative to nasogastric lavage, retrieval to January 2016, and back into the study of references. According to include and exclude standard to screening literature, literature which met inclusion criteria was selected for quality evaluation and data extraction. Meta-analysis and trial sequential analysis were performed by using Rev-Man 5.3 and TSA soft-ware to estimate the required information size (RIS). Results 13 studies incorporated into Meta-analysis including a total o 1 296 patients. Meta-analysis results showed that the oral intubation gastric lavage group was better than nasogastric lavage in shortening the time of intubation [MD=-9.05, 95% CI(-12.86--5.23), P=0.00], improving the rate of intubation for the first time [MD=1.36, 95% CI (1.14-1.62), P =0.00], mucosal bleeding complication [MD=0.11, 95% CI (0.04-0.64), P=0.010], nausea and vomiting [MD=0.60, 95%CI (0.37-0.98), P=0.04], kinking [MD=0.14, 95% CI (0.02-0.80), P=0.03], reflex cardiac arrest [MD=0.24, 95% CI (0.08-0.71), P=0.01] and asphyxia [MD=0.45, 95% CI (0.26-0.80), P=0.007], the difference were statistically significant. But no significant difference existed in gastric tube fall off. Conclusions The oral intubation gastric lavage could shorten intubation time of patients, improve the first time intubation rate and reduce certain complications. It suggests that the oral intubation be extensively applied to care patients with oral poisoning.
4.Impact of image quality with scan parameters and reconstruction algorithms in head dual-energy computed tomography angiography

Objective To study the difference of image quality which based on different reconstruction in head dual‐energy computed tomography angiography (DECTA) scanning ,and evaluate the radiation dose which using different tube voltage in DEC‐TA .Methods An anthropomorphic head phantom with simulated arteries was used for DECTA scanning by different scan parame‐ters (tube voltage 80/140 kV and tube voltage 100/140 kV) ,the data was reconstructed with filtered back projection (FBP) and Si‐nogram Affirmed Iterative Reconstruction (SAFIRE) .The CT values with SD were measured and calculate to SNR and CNR ,the dose length product (DLP) was recorded and invert to effective dose .The image quality and radiation dose were evaluated in differ‐ent imaging condition .Results The image noise of SAFIRE reconstruction was lower than FBP reconstruction in same san parame‐ters .The increase of tube current‐time product from 100 mAs to 300 mAs ,it shows the characteristics of linear tendency .When it was on 260 mAs ,the image quality become stable .The difference between the two groups(80/140 kV ,100/140 kV)of SD of CT value by SAFIRE was significant (P< 0 .05) ,the difference of SNR and CNR between the two groups was not significant (P>0 .05) .The effective doses of group A (80/140 kV) was significantly lower than group B(100/140 kV) .Conclusion Using tube voltage of 80/140 kV combine with SAFIRE algorithm in head DECTA can ensure the image quality ,which can significantly reduce radiation dose .
5.Detecting of nitric oxide content in pulmonary artery-and superior vena cava-derived plasma in the patients with congenital heart disease

Objective:To explore the relationship between the nitric oxide(NO)and the pathogenesis of the pulmonary hypertension(PH)induced by congenital heart disease(CHD). Method:NO content in plasma of CHD patient's pulmonory artery and superior vena cava was detected with NO kit. Result:NO content in the pulmonary artery of the patients with PH was much higher than that of patients without PH(37.58?9.99?mol/L vs 19.03?15.25?mol/L,P0.5). Conclusion:NO content in the pulmonary artery of CHD patients with PH increase,which may be involved in the pathogenesis of PH induced by CHD.
6.Endogenous hydrogen sulfide participating in the ischemic postconditioning against cardiac ischemia/reperfusion injury

Aim To investigate whether endogenous hydrogen sulfide participated in the ischemic postconditioning against ischemia/reperfusion injury in the isolated rat hearts.Methods Isolated SD rat hearts were subjected to ischemia by stopping perfusion for 30 min followed by reperfusion for 60 min in Langendorff perfusion system,at the onset of reperfusion,4 cycles of 15 seconds reperfusion followed by 15 seconds ischemia were given for postconditioning.Intraperitoneal injection of L-propargylglycine(PAG,the inhibitor of cystanthionine-?-lysase) half an hour before operation or administration of exogenous hydrogen sulfide donor-NaHS for postconditioning was performed to observe the impact on ischemic postconditioning.The heart rate(HR),left ventricular developed pressure(LVDP),coronary arterial flow(CF)were recorded.The activity of lactate dehydrogenase(LDH)in the coronary effluent fluid,and the activity of CSE and the content of H2S in the isolated rat hearts were measured with colorimetry method.The infarction sizes were measured with TTC dying method.Results In ischemic/reperfusion group,LVDP decreased,the activity of LDH in the coronary effluent fluid and the size of myocardial infarction increased(vs control,P

Aim To investigate the expression of phosphorylated JNK1/JNK2 and the protection of 11,12-EET in ischemic and reperfusion rat heart.Method The expression of JNK1/JNK2 was detected with western blot method and the changing of heart function during ischemia/reperfusion process was observed in different groups. Results The cardiac function (+dp/dt_(max)%,-dp/dt_(max)% and LVDP)of reperfusion periods(30 min) apparently decreased in ischemia/reperfusion (I/R) group contrasted with Sham group, short ischemia(SI)+I/R group and EET+I/R group,and the expression of phosphorylated JNK1/JNK2 increased in I/R group contrasted with nromal group,Sham group and EET+I/R group.Conclusion The myocardial protection of 11,12-EET ( 6.24?10~(-8) mol?L~(-1)) is able to inhibit the expression of phosphorylated JNK1/JNK2.
8.Effects of platelet-rich plasma on proliferative and osteogenetic activity of skeletal muscle stem cells in vitro

BACKGROUND:Studies has confirmed that platelet-rich plasma(PRP) can affect the proliferation and differentiation of bone marrow mesenchymal stem cells and adipose derived stem cells. OBJECTIVE:To observe the effects of PRP on the proliferative and osteogenetic activity of skeletal muscle stem cells(SMSCs) in vitro,in addition,to elucidate the potential mechanism by which PRP affects SMSCs. DESIGN,TIME AND SETTING:A randomized controlled animal experiments was completed in the Center Laboratory of the Sixth People's Hospital Affiliated to Shanghai Jiao Tong University from June to October in 2008. MATERIALS:Nine New Zealand,ordinary grade rabbits,weight of 2.5-3.0 kg,1 year old,irrespective of gender. METHODS:The right soleus was dissected and cultured for SMSCs. PRP was prepared with central artery of rabbits’ ears. The cells were randomly divided into the experimental group and the control group. In the experimental group,the SMSCs were interfused with PRP(special culture media including 12.5% PRP) . There was no intervention in the control group. MAIN OUTCOME MEASURES:①Morphological observation. ②The proliferative ability of the cells was detected with MTT. ③ The osteogenetic ability was measured with alkaline phosphatase(AKP) staining,alizarin red staining and osteocalcin immunofluorescence assay. 9.Meta analysis of clinical and pathological features of breast cancer in young women in China

Objective To compare the clinical and pathological features of Chinese young breast cancer(age ≤ 35)with elder patients(> 35)using Meta analysis .Methods Published studies concerning clinical and pathological features of young breast cancer in China were searched systemically and assessed .Stata12 .0 software was used for data analyzing and calculating OR and its 95%CI .Results Totally 31 studies were selected for Meta analysis ,and most of them were classified as 6 - 7 scores ,which showed the quality of articles was high .The risk factors of breast cancer and its pooled odds ratio values with statistical significance were as fol‐lows 6 .42(95% CI :4 .22 - 9 .79) ,0 .61(95% CI :0 .50 - 0 .74)when clinical staging of 0 - Ⅱ phase or Ⅰ - Ⅱ phase ,2 .25(95% CI :1 .69 - 2 .99)when histological type of Invasive carcinoma ,1 .73(95% CI :1 .23 - 2 .43)when histological grade of III grade ,1 .80 (95% CI :1 .23 - 2 .43)when positive of lymph node metastasis .Conclusion Compared with elder breast cancer ,the clinical and pathological characteristics of young breast cancer were mainly for the high misdiagnosis rate ,the late clinical stage ,the high pro‐portion invasive carcinoma ,the poor histological differentiation and the lymph node metastasising easily ,the hint of young breast cancer screening and treatment may be different principles and measures should be adopted .
10.Expression of BIM gene in papillary thyroid carcinoma and its association with the BRAFV600E mutation

To study the link between BRAFV600E status and the expression of BIM gene in papillary thyroid carcinoma( PTC) tissues and to analyze the association of these factors with clinicopathological characteristics. BRAFV600E status was determined by MASA-PCR, and qPCR was applied to detect the expression of BIM gene. Finally, the associations of these factors with clinicopathological characteristics were analysed. The rate of mutant BRAFV600E in PTC was 54. 1% , and the expression of BIM gene was lowered in BRAFV600E positive PTC tissues. Additionally, there was significant association( P < 0. 05) between BRAFV600E positiveness and raised TNM Staging (Ⅲ/ Ⅳ), and lowered BIM expression was significantly associated (P<0. 05) with the tumor size and raised TNM Staging(Ⅲ/ Ⅳ). These findings may help us to know more about the mechanism of PTC and to develop new diagnostic biomarkers or prognostic indicators of PTC.
