1.Establishment and application of virtual craniofacial surgical platform based on personal computer

Anatomic structure in craniofacial region is very complicated and orthognathic surgery is difficultly performed.Therefore,it is necessary to generate virtual surgical model from CT scan data before operation on personal computer in order to make surgery more accurate.This technique has been increasingly interested in the field of orthognathic surgery both at home and abroad.The sample was scanned with thin-layer CT.CT image data (336 layer,1.0 mm slice thickness) were analyzed with the Mimics software.Simultaneously,three-dimensional model and virtual surgery of the facial cranium were established.The digitized virtual surgical platform of facial cranium was preliminarily created,and three common surgical methods in maxillofacial surgery,i.e.,LeFort Ⅰ osteotomy,mandibular angle osteotomy,and genioplasty,for maxillofacial surgery were simulated.The results showed that the digitized virtual surgical platform of facial cranium could be generated with the Mimics software on personal computer,which provides convenient and quick methods for research,teaching,and clinical surgery.More importantly,it creates theoretical basis for virtual surgical platform which can be widely used on personal computer.
2.Metadherin promotes gastric carcinoma metastasis through induction of epithelial-mesenchymal transi-tion

Objective To explore the relationship between gene MTDH expression and its role in promo-ting gastric carcinoma metastasis .Methods We collected clinical specimens and cultured gastric carcinoma cell lines.By Western blotting and Real -time PCR methods,protein and mRNA levels in tissues and MTDH relation-ship with EMT were detected .Results There was 86%of patients who expressed MTDH positively and 13%of normal gastric mucosa was positive expression .The results showed that the expressive level of MTDH gene in gas-tric carcinoma was higher than in the normal tissues .The expression of MTDH was correlated with TNM stage、mi-crovascular invasion、recurrence and metastasis .The expressive level of MTDH was correlated with two epithelial mesenchymal transition markers ( E-cadherin and N-cadherin ) .Conclusion MTDH may promote gastric car-cinoma metastasis through the induction of EMT process and may be a candidate biomarker for therapeutic target .
3.Multidisciplinary team confirmed the simultaneous diagnosis of CLL and AML-M5

A 54-year-old man diagnosed with chronic lymphocytic leukemia (CLL) was admitted in our department in June 2014. After one cycle of FC chemotherapy, a bone marrow examination revealed normalized lymphocyte count and another acute myeloid leuke-mia (AML)-M5 clone. The patient refused sequential treatment and only received follow-up examination. He had continuous hemato-logically stable disease and died of pulmonary infection on July 2015. After a multidisciplinary team discussion, we confirmed the si-multaneous diagnosis of CLL and AML-M5. Through this discussion,tumor second hit model,tumor evolution model,andtumor heterogeneitywere further defined.
4.Relationship between survivin expression and paclitaxel drug-resistance and research of reversed drug-resistance by matrine in non-small cell lung cancer

Objective To study the correlation between drug-resistance of paclitaxel and the expres- sion levels of anti-apoptotic protein survivin and the role of matrine reversal resistance of PTX in non-small cell lung cancer.Methods The expressions of survivin in 120 samples of human lung cancer with paclitaxel chemotherapy were detected by immunohistochemical staining.Results The positive expression rate of sur- vivin was 57.5%.In the positive expression of survivin group,chemotherapy combined matrine could improve response rate and survival time(P0.05).Conclusion The survivin expression was related to the response rate of paclitaxel in lung cancer.It might be a valuable new marker to predict the drug-resistance of paclitaxel.In this prospective research,survivin protein,which promoted the apoptosis caused by paclitaxel,may be the target of martine.It could partly reverse the drug re- sistance to paclitaxel.
5.Repair of wound defect with exposed tibia with facial flap pedicled with vascular perforating branch of leg

Objective To explore the method of repairing wound defects with exposed tibia. Methods 32 patients with soft tissue defects with exposed tibia were treated with three kinds of flaps pedicled with interior,lateral and posterior vascular perforating branches in the leg,respectively.Re- suits One flap with distal part necrotic was treated with change of dressings and got one-stage healing. One case had delayed union for muscular infection under the flap.Other flaps all were successful and sur- vived.No ostemyelitis was found.Conclusion The flap pedicled with vascular perforating branches of leg has abundant blood supply and is a good method for repairing small or middle wound defect with ex- posed tibia.
6.Genotyping of Epstein-Barr virus in Epstein-Barr virus associated gastric carcinoma.

Tumor samples from 236 gastric carcinoma patients and throat washings (TWs) from 135 healthy adults were collected and screened for EBV genome. 17 Epstein-Barr virus associated gastric carcinoma (EBVaGC) and 33 EBV positive TWs were further examined for type 1/2 EBV and polymorphism at Bam HI F, Bam HI W1/I1 boundary regions and Xho I restriction site in LMP1 gene. No type f of Bam HI F polymorphism was found in all the cases, nor type 2 in EBVaGC. 25 of 33 TWs (75.8%) analyzed detected type 1 virus, while 8 TWs (24.2%) detected type 2 virus. Type I and i of Bam HI W1/I1 polymorphism accounted for 1(5.9%) and 16 (94.1%) in EBVaGC and 11 (33.3%) and 19 (57.6%) in TWs, respectively. LMP1 Xho I(+) and (-) polymorphism accounted for 0(0) and 15 (88.2%) in EBVaGCs and 12 (36.4%) and 18 (54.5%) in TWs, respectively. Among those specimens with determinable genotypes through all the polymorphism, the distribution rate of type 1/i/Xho I(-) in EBVaGC (15/15, 100%) was significantly higher than that in TWs (4/28, 14.3%) (chi2 = 29.098, P < 0.0001), suggesting that the distinctive EBV strain (1/i/Xho I(-)) could be associated with Chinese EBVaGC.

7.Effect of millimeter therapy in burning mouth syndrome.

OBJECTIVETo observe the millimeter wave therapy responses in patients with burning mouth syndrome.
METHODSEighty patients were randomized divided into 4 groups. The first group was treated with both millimeter wave irradiation and routine medication, the second group with millimeter wave irradiation, the third with pretending millimeter wave irradiation and routine medication and the fourth with routine medication. Pain, extravasated blood level and autonomic nerve system condition were double-blindly evaluated either before or after the treatment.
RESULTSStatistically significant difference (P < 0.05) was found as the degree of pain was compared before and after treatment of all the 4 groups. The first and second group, which were affected by the millimeter wave irradiation, had obvious improvements in the extravasated blood level and autonomic nerve system condition (P < 0.05). When the 4 groups were compared with each other, there were significant differences (P < 0.05) between the first and the fourth groups, and the second and the fourth groups regarding the reduction of pain. According to extravasated blood level, significant differences (P < 0.05) were found in the first and third or fourth groups, the second and third or fourth groups.
CONCLUSIONSThe irradiation of holographic point by millimeter wave can improve the patients' pain, extravasated blood level and autonomic nerve system condition. It might provide a new treatment method for burning mouth syndrome.

8.A novel SELEX method for screening of HIV-1 P24 antigen.

OBJECTIVETo screen aptamers that can bind P24 antigen tightly and specificly, and verify its specificity and affinity.
METHODSPolycarbonate PCR plate was coated with P24 antigen and SELEX technology was used to screen aptamer on the PCR plate. The primary and secondary structure of these aptamers was analyzed by software. Through HRP labeled streptavidin and biotin labeled aptamers, the affinity and specificity of obtained aptamers were verified by ELISA.
RESULTSThe polycarbonate PCR plate could be coated with P24 antigen. Electrophoretic analysis showed the aptamers had been enriched. Sequence aligment analysis showed that these aptamers have consensus sequence and their apatial structure was multiple; ELISA verified that aptamers had high affinity with P24 antigen.
CONCLUSIONA simple method was established for screening aptamers that can bind HIV-1 P24 antigen specificly and tightly.

9.Amplification of EMS1 gene in oral carcinogenesis.

OBJECTIVETo investigate the amplification of EMS1 gene in the carcinogenesis of oral mucosa.
METHODSA total of 78 subjects, including 30 patients with oral leukoplakia (OLK), 33 with oral squamous cell carcinoma (OSCC), and 15 healthy controls, were studied. By using microdissection method, we obtained normal mucosa, hyperplastic epithelia, mild-dysplastic epithelia, moderate-dysplastic epithelia, severe-dysplastic epithelia and primary OSCC tissue. Then we analyzed EMS1 amplification by using differential PCR.
RESULTSEMS1 amplification began from moderate-dysplastic epithelia and occurred in 20.0% OLK cases and 57.6% OSCC cases. In the progress of OSCC, no gene amplification was observed in normal tissues, non-dysplastic OLK and mild-dysplastic OLK, while in the cases with metastasis, amplification frequency increased significantly (P = 0.015).
CONCLUSIONSEMS1 amplification parallels with the progress of oral carcinogenesis, indicating their potential roles in oral carcinogenesis.

10.Effect of Changtong oral liquid on fibroblast proliferation in normal and adhesive rat peritoneal tissues.

OBJECTIVETo investigate the effect of Changtong oral liquid (CTOL) on the proliferation of cultured fibroblasts derived from normal peritoneum (NFs) and adhesive peritoneum (AFs) of rats.
METHODSTwenty male SD rats were randomized into 4 groups, including a normal serum group and 3 CTOL groups with CTOL treatment at low, medium or high doses. Serum samples were obtained from the abdominal arteries of the rats after oral treatment with CTOL for 7 days. The fibroblasts were isolated from the peritoneum by means of tissue culture, and the passage 3-8 cells were cultured with the sera of the normal control and CTOL groups for 24, 48, 72 and 96 h. MTT assay was used to observe the proliferation of the fibroblasts.
RESULTSThe dose of CTOL was inversely correlated to the absorbance but positively to the growth inhibition rates. Compared with the NFs cultured in normal control rat serum, the NFs in serum from CTOL groups showed no obviously changes in the absorbance at 24 and 48 h, but displayed significant reduction at 72 and 96 h (P<0.01). Compared with the AFs in normal rat serum, the AFs in the 3 CTOL groups all showed significantly decreased absorbance at 24, 48, 72 and 96 h (P<0.05). At the same time point, the inhibition rate of AFs in low-dose CTOL group showed no significant difference from that in the normal control group, but CTOL at a medium dose resulted in a significantly higher inhibition rate of AFs at 72 h (P<0.05). High-dose CTOL produced significant differences in the inhibition rates of AFs and NFs (P<0.05).
CONCLUSIONCTOL can inhibit the proliferation of AFs and NFs in vitro. AFs appear to be more sensitive to CTOL, which has a dose-dependent inhibitory effect of AF proliferation.
